    Change of the composition of splenic lymphocytes at the effect of argentogalactomanann and argento-poly-1-vinyl-1,2,4-triazole on the experimental animals

    The article presents the results of the research of effect of metal-containing nanocomposites on the content of splenic Band T-lymphocytes of white mice. Phenotypic analysis of the spleen cells suspension was conducted by flow cytometry on BD FACSCanto ™ П. Stimulating effect of these medications on the immune response formation is showed and is proved by the increase of content of B-lymphocytes and T-helper cells and it indicates cooperative interactions between the cells of immune system. It was experimentally showed that the medications 2-N-HTP-Ag, GM-Ag can be recommended for the further research in order to increase protective properties of an organism

    Results of the study of the immunogenic activity of cell envelopes of Francisella tularensis different subspicies (Report 2)

    The effect of cell envelopes preparations of four subspecies of Francisella tularensis, derived from urea lysates of tularemia microbe on cytokine production by immunocompetent cells of experimental animals has been studied. Stimulating influence of F. tularensis cell envelopes preparations on cytokine synthesis on the 7th day after inoculation was shown. The results showed that the cell envelopes of different subspecies F. tularensis have different stimulatory effect on the production of proinflammatory cytokines and growth factors GM-CSF and G-CSF, further defining the immune system of host.The results allow to consider cell envelopes preparations of F. tularensis as constituent component in the development of subunit vaccines

    ACTION OF 1-VINYL-1,2,4-TRIAZOLE COPOLYMER WITH N-VINYLPIROLIDONE AND METAL-CONTAINING NANOCOMPOSITES ON FUNCTIONAL STATE OF PHAGOCYTIC CELLS

    The creation of immuno-biological drugs to increase the effectiveness of specific immunotherapy and immunoprophylaxis is a current trend in medicine to date. Therefore, the search and development of new safe and effective artificial bioorganic complexes (immunomodulators) capable of stimulating individual responses of immune system to reduce infectious morbidity is an important area of research. At the moment, such promising preparations can be considered water-soluble polymeric materials and organo-inorganic polymer nanocomposites with nanoparticles of various metals which possess immunomodulating and bactericidal properties. The study results of the effects of 1-vinyl-1,2,4-triazole copolymer with N-vinylpyrrolidone and nanocomposites based on it with gold and silver nanoparticles on acute toxicity and functional state of immunophagocytic cells of experimental animals, as well as the production of IFN-γ, TNF-α and IL-4 by human blood lymphocytes are represented in this article. It was established that the tested preparations do not cause increase in body temperature, decrease in mass and death of animals. It was shown that they also have the multidirectional action on cytokine production by human blood cells in vitro, the ability to increase activity of оxidative stress-related enzymes and antioxidant protection of phagocytes. The data obtained by us allow to explain the need for further investigation of the actions of nanocomposites and copolymer in vitro and in vivo

    Study of Bacillus anthracis 34F2 Sterne antigenic preparation S-2 and its combined use with nanostructured cobalt-arabinogalaktan (Со-АG) is presented. The ability of these preparations to stimulate proliferation and differentiation of B-lymphocytes is demonstrated. However, content of the B-lymphocyte circulating subpopulations depends on the time of observation. Co-AG exhibits adjuvant properties enhanced the immunogenic features of the S-2 B. anthracis 34F2 Sterne that may indicate its availability as an adjuvant in the construction of chemical vaccines

    Relevance. In Russia, the live plague vaccine (LPV) is used for specific prophylaxis of plague. Immunological monitoring of humans vaccinated by LPV in order to search for informative diagnostic markers, as well as to improve the tactics of epidemiological surveillance of plague enzootic territories is an urgent area of research.The aim is to assess the parameters of cellular and humoral immunity in humans revaccinated by LPV who permanently reside on territory of the Tuvinian natural plague focus.Materials and methods. The study involved 76 volunteers from the Republic of Tuva, revaccinated by LPV. Blood sampling was performed before vaccination and 1, 3, and 6 months after revaccination. The study included the determination of cytokine production (IFN-γ, IL-4, TNF-α), specific antibodies, immunoglobulins (IgM, IgG, IgA and IgE) and lymphocyte subpopulation composition (CD3, CD4, CD8, CD16, CD19, immunoregulatory index).Results. A decrease in IgG and IgM and an increase in IgA were found after vaccination with LPV and their increase after revaccination. Correlation relationships were revealed between immunoglobulins, B cells and IL-4. Revaccination leads to an increase seroconversion. The activation of humoral immunity in humans vaccinated against plague is also evidenced by dynamics of changes in the subpopulation composition: an increase in B-lymphocytes and natural killer cells, a decrease in T-helpers and immunoregulatory index, and cellular immunity stimulation is an increase in spontaneous and induced production of pro- and anti-inflammatory cytokines.Conclusion. It has been shown that LPV is capable of causing the body’s immune restructuring and activating the cellular and humoral mechanisms of immunological protection. For a complete understanding of the development and preservation of  antiplague immunity, it is necessary to continue the annual immunological monitoring of the population living on the territory of the Tuvinian natural plague focus, using additional modern research methods
