    Association of Single Nucleotide Polymorphisms in the IL-18 Gene with Production of IL-18 Protein by Mononuclear Cells from Healthy Donors

    IL-18 has proinflammatory effects and participates in both innate and adaptive cellular and humoral immunity. A number of SNPs that influence IL-18 production are found in the gene promoter region. We investigated the association of SNPs in the IL-18 promoter at −607 and −137 with the level of IL-18 protein production by PBMC from healthy donors from Southwestern Siberia. The genetic distribution of these SNPs in the promoter site was established by PCR. IL-18 protein production was determined by ELISA. Our results showed that PBMC from donors carrying allele 137C have lower levels of both spontaneous and LPS-stimulated IL-18 production. In contrast, PBMC from donors carrying allele 607A showed significant increases in spontaneous and stimulated IL-18 production compared to wild type. Our study suggests that the SNPs −607 and −137 in the promoter region of the IL-18 gene influence the level of IL-18 protein production by PBMC from healthy donors in Southwestern Siberia

    THE FREQUENCY OF ALLELIC VARIANTS OF GENES TNFRI AT POSITIONS -609 AND -1207 AND TNFRII AT POSITIONS -1709 AND -3609 IN HEALTHY DONORS AND PATIENTS WITH RHEUMATOID ARTHRITIS

    In this paper we studied the frequency of allelic variants of genes TNFRI at positions -609 and -1207 and TNFRII at positions -1709 and -3609 in patients with rheumatoid arthritis and in population control group. It was shown that the resulting frequency distribution of alleles and genotypes of TNFRI gene promoter in a group of population control is characteristic for Asian populations, for TNFRII gene polymorphisms and the frequency distribution differed from the European and Asian populations. Comparative analysis of allele and genotype frequencies of TNFRI gene promoter at positions -609 and -1207 and TNFRII and -3609 and positions -1709 in patients with rheumatoid arthritis and in population control group showed no statistically significant differences. However in the analysis of combinations has been shown that in patients with rheumatoid arthritis compared with the control population was significantly less common combination of genotypes TNFRI-609GT + TNFRII-3609CC

    Полиморфизм микросателлитных локусов у белого (Hypophthalmichthys molitrix Val.) и пестрого (Hypophthalmichthys nobilis Rich.) толстолобиков, выращиваемых в аквакультуре в Республике Беларусь

    In this study, we evaluated the genetic diversity of silver (Hypophthalmichthys molitrix Val.) and bighead (Hypophthalmichthys nobilis Rich.) carps grown in aquaculture in the territory of the Republic of Belarus. Genotyping was obtained for 11 STR-loci - Hmo11, Hmo13, Hmo15, Hmo25, Hmo26, Hmo31, Hmo33, Hmo 34, Hmo36, Hmo37, and Hmo40. The following parameters were calculated: the average number of alleles per locus, the effective number of alleles, the levels of expected and observed heterozygosity, the value of the Shannon information index and the fixation indexes FIS and FST. The obtained results indicate a moderate (for silver carp) and sufficiently high (for bighead carp) genetic diversity of the studied samples of carps up to the possibility to allocate several groups for subsequent work on obtaining linear material and further reproduction, including commercial fish. However, to achieve these goals, it is necessary to pay special attention to the selection of pairs of producers taking into account the results of molecular genetic analysis. The scheme for the differentiation of hybrid individuals between silver and bighead carps and the use of molecular genetic analysis of STR-loci would be proposed. This approach can be used as a minimally invasive rapid test in breeding and reproductive programs for the studied fish species.Communicated by Academician A. V. KilchevskyДана оценка генетического разнообразия белого (Hypophthalmichthys molitrix Val.) и пестрого (Hypophthalmichthys nobilis Rich.) толстолобиков, выращиваемых в аквакультуре на территории Республики Беларусь, по данным генотипирования 11 микросателлитных локусов - Hmo11, Hmo13, Hmo15, Hmo25, Hmo26, Hmo31, Hmo33, Hmo34, Hmo36, Hmo37, Hmo40. Рассчитаны следующие показатели: среднее число аллелей на локус, эффективное число аллелей, уровни ожидаемой и наблюдаемой гетерозиготности, значение информационного индекса Шеннона, индексы фиксации FIS и FST Полученные результаты свидетельствуют об умеренном (для пестрого толстолобика) и достаточно высоком (для белого толстолобика) генетическом разнообразии изученных выборок, что дает возможность выделить несколько групп для получения линейного материала и дальнейшего воспроизводства, в том числе товарной рыбы. Вместе с тем следует уделить особое внимание подбору пар производителей с учетом результатов молекулярно-генетического анализа. На основании молекулярно-генетического анализа пяти микроса-теллитных локусов предложена схема по дифференциации гибридных особей между пестрым и белым толстолобиками, что может быть использовано в качестве малоинвазивного экспресс-теста в селекционных и воспроизводительных программах для данных видов растительноядных рыб

    Since 1960s UNIIM performs research in the field of needs in reference materials of composition and properties of substances and materials (RM) as well as develops it. During the research UNIIM has developed 757 types of RMs for metrological measurement assurance of factors of composition and properties of substance and materials for test laboratories of chemical, pharmaceutical, fuel, food industry, agriculture, metallurgy and ecological monitoring laboratories. List ofRMs enlarges thanks to development of UNIIM standards and transmission measurement facility from State standards of units. Taking into account the actual requirements in the field of measurements, the UNIIM's key destination is to assure the accuracy and the metrological traceability of measurements. The present-day system of RMs to be developed in UNIIM includes RMs of composition of inorganic and organic compounds and their solutions, fuels, stable isotopic materials, water, grounds, food products, biomaterials, nanomaterials, metals, alloys and other materials offerrous and non-ferrous industry, RMs of properties (thermodynamic, magnetic, physical-chemical, technical) of substances and materials. The present article considers history of RMs list development which were created by UNIIM and the strategy of this direction.Исследования в области мониторинга потребности в стандартных образцах состава и свойств веществ и материалов (СО), создание стандартных образцов УНИИМ проводит c 60-х годов ХХ века. За годы исследований УНИИМ разработал 757 типов СО для метрологического обеспечения измерений показателей состава и свойств веществ и материалов, предназначенных для испытательных лабораторий химической, фармацевтической, топливной, пищевой промышленности, сельского хозяйства, металлургии, лабораторий экологического мониторинга. Развитие номенклатуры СО, выпускаемых УНИИМ, неразрывно связано с развитием эталонной базы УНИИМ и формированием средств передачи измерительной возможности от государственных эталонов единиц величин. Учитывая современные требования в области измерений, ключевым предназначением СО УНИИМ является обеспечение точности и метрологической прослежи-ваемости измерений. Современная система СО, разрабатываемых и выпускаемых УНИИМ, включает СО состава неорганических и органических соединений и их растворов, топлив, стабильных изотопных материалов, воды, почвы, пищевых продуктов, биологических материалов, наноматериалов, металлов, сплавов и других материалов цветной, черной металлургии, СО свойств (термодинамических, магнитных, физико-химических, технических) веществ и материалов. Настоящая статья посвящена историческим этапам развития номенклатуры СО, разрабатываемых и выпускаемых УНИИМ, а также перспективам развития в этом направлении
