    EndoNet: model for automatic calculation of H-score on histological slides

    H-score is a semi-quantitative method used to assess the presence and distribution of proteins in tissue samples by combining the intensity of staining and percentage of stained nuclei. It is widely used but time-consuming and can be limited in accuracy and precision. Computer-aided methods may help overcome these limitations and improve the efficiency of pathologists' workflows. In this work, we developed a model EndoNet for automatic calculation of H-score on histological slides. Our proposed method uses neural networks and consists of two main parts. The first is a detection model which predicts keypoints of centers of nuclei. The second is a H-score module which calculates the value of the H-score using mean pixel values of predicted keypoints. Our model was trained and validated on 1780 annotated tiles with a shape of 100x100 ÎĽm\mu m and performed 0.77 mAP on a test dataset. Moreover, the model can be adjusted to a specific specialist or whole laboratory to reproduce the manner of calculating the H-score. Thus, EndoNet is effective and robust in the analysis of histology slides, which can improve and significantly accelerate the work of pathologists

    Human wellbeing and automotive industry: correlations in the era of economical digitalization

    This research studies the relation between human wellbeing and automotive industry, as to whether the performance of automobile industry translates to an overall well-being of the populace. The study is based on secondary-data, and mainly takes into account the Prosperity-Index / ranking and its possible linkage with automotive sale volume of the nations. Findings of this study confirms that that the higher the prosperity index or ranking, the higher the automotive sales volume for most of the nations but several factor should be taken in to consideration, for example India automotive sale volume is bigger than the automotive sales volume of France but France prosperity rank is 19 and India in on 88 rank, it is because of the size and population of the country, India might have 10 times more population than France when divided among the population France may investing much more than India on their citizen. Findings reveal that Canada leads the table on first position with Australia on second position in the prosperity ranking, due to they provide enough opportunities for their people to live a good and healthy lives and these can be observed in terms of good automotive sales volume by these two nations. Further, finding also reveals that the automotive sales number of United States of America or several other countries have large value but that does not mean that they are also good in ranking in Prosperity Index which implies that the prosperity ranks has no direct relation with the automotive sales volume that a country generates

    Industry 4.0 readiness: the impact of digital transformation on supply chain performance

    In the context of Digital Economy the usage of information and communication technology to promote commercial operations has grown significantly with the advent of electronic business and networking systems. This research focuses on the new generation technologies and on communications between all supply chain components. The study enable Russian and Thai retail supply chain administrators and market leaders (including policy makers) to share their expert opinion about impact of digital transformation on supply chain performance. Once each organization affirmed its commitment to be engage in the interview, analytical conclusions were made on the basic of semi-structured open discussions and surveys of 18-36 employees from each organization which were combined in such a way as to present a complete profile of the organizations readiness to Industry 4.0 strategy. 236 semi-structured open interviews focused on a few topics and keywords have been performed. In fact, five generalized qualitative interviews (profiles) present the analytical evidence from five leading brands. This approach helps to consider more fully how they perceive and understand digital transformation. The purpose was to gain a rich basis of the interviewed person’s content and this approach is quite well adapted to analyses. The research implemented the quantitative methodology throughout using the named data and had been treated with SPSS statistical tool for hypotheses validation. As the result statistically evidenced presence of a relationship between digitalization, the Industry 4.0 technologies and supply chain performance

    The digitalization of monetary and foreign exchange circulation is one of the leading institutional and infrastructural transformations at the beginning of the 21st century, affecting not only the long-term interests and priorities of business development and state progress, but also the very principles of public-private relations within the socio-economic system. The article assess opinions of 350 experts from 8 sectors of the economy of Russia and Thailand for the prospects of cryptocurrency (on the example of Bitcoin) introducing into official calculations, turnover, accumulation, as well as the readiness of business and state for this digital transformation in context of the Digital Economy trend globalization. The research implemented the quantitative methodology throughout using a survey and the data had been treated using SPSS statistical tool for hypotheses validation. The empirical data obtained confirmed the priorities of the factors for accelerating the currencies cryptotization, identified the main problems of Russian and Thai business, pessimizing the potential for using Bitcoins, and the concept of state financial regulation modernizing in the dynamics of digitalization

    Heterologous Expression of Xylanase xAor from Aspergillus oryzae in Komagataella phaffii T07

    Xylanases (EC 3.2.1.8) hydrolyze the hemicellulose of plant cell walls. Xylanases are used in the food and paper industries and for bioconversion of lignocellulose to biofuel. In this work, the producer-strain with four copies of the xAor xylanase gene was organized in two tandem copies for optimal expression in Komagataella phaffii T07 yeast. The secreted 35 kDa xylanase was purified from culture medium by gel filtration on Sephadex G-25 and anion exchange chromatography on DEAE-Sepharose 6HF. Tryptic peptides of the recombinant enzyme were analyzed by liquid chromatography-tandem mass spectrometry where the amino acid sequence corresponded to Protein Accession # O94163 for Endo-1,4-beta-xylanase from Aspergillus oryzae RIB40. The recombinant xylanase was produced in a bioreactor where the secreted enzyme hydrolyzed oat xylane with an activity of 258240 IU/mL. High activity in the culture medium suggested xylanase could be used for industrial applications without being purified or concentrated. The pH optimum for xylanase xAor was 7.5, though the enzyme was active from pH 2.5 to pH 10. Xylanase was active at temperatures from 35 °C to 85 °C with a maximum at 60 °C. In conclusion, this protocol yields soluble, secreted xylanase suitable for industrial scale production
