    A History of Vocational Education in Oregon Since 1917

    86 pagesThere has long been a need of an adequate history of vocational education in Oregon. The compilation of this history I am attempting to do in a manner which will not be too detailed, but which will nevertheless trace the progress and evolution of the different phases of vocational education of the secondary level from approximately 1917 until the present time, and treat all the significant aspects with such dexterity and conciseness as to make the survey readily readable. The information for this study was assembled from various sources. The State Board for Vocational Education through the avenues of the several department heads were willing and valuable contributors. Superintendents of city schools, heads of vocational schools, and instructors in the various divisions of the vocational filed were contacted, as well as those who were pioneers in the development of this field of education. Courses of study, surveys, vocational texts, and pamphlets have all been used in this study

    The Use of a Stringent Selection System Allows the Identification of DNA Elements that Augment Gene Expression

    The use of high stringency selection systems often results in the induction of very few recombinant mammalian cell lines, which limits the ability to isolate a cell line with favorable characteristics. The employment of for instance STAR elements in DNA constructs elevates the induced number of colonies and also the protein expression levels in these colonies. Here, we describe a method to systematically identify genomic DNA elements that are able to induce many stably transfected mammalian cell lines. We isolated genomic DNA fragments upstream from the human Rb1 and p73 gene loci and cloned them around an expression cassette that contains a very stringent selection marker. Due to the stringency of the selection marker, hardly any colony survives without flanking DNA elements. We tested fourteen ~3500 bp DNA stretches from the Rb1 and p73 loci. Only two ~3500 bp long DNA fragments, called Rb1E and Rb1F, induced many colonies in the context of the stringent selection system and these colonies displayed high protein expression levels. Functional analysis showed that the Rb1 DNA fragments contained no enhancer, promoter, or STAR activity. Our data show the potential of a methodology to identify novel gene expression augmenting DNA elements in an unbiased manner

    Membrane traffic in eukaryotic cells involves transport of vesicles that bud from a donor compartment and fuse with an acceptor compartment. Common principles of budding and fusion have emerged, and many of the proteins involved in these events are now known. However, a detailed picture of an entire traffickingorganelle is not yet available. Using synaptic vesicles as a model, we have now determined the protein and lipid composition; measured vesicle size, density, and mass; calculated the average protein and lipid mass per vesicle; and determined the copy number of more than a dozen major constituents. A model has been constructed that integrates all quantitative data and includes structural models of abundant proteins. Synaptic vesicles are dominated by proteins, possess a surprising diversity of trafficking proteins, and, with the exception of the V-ATPase that is present in only one to two copies, contain numerous copies of proteins essential for membrane traffic and neurotransmitter uptake.status: publishe
