2024 Recommendations for Validation of Noninvasive Arterial Pulse Wave Velocity Measurement Devices
BACKGROUND: Arterial stiffness, as measured by arterial pulse wave velocity (PWV), is an established biomarker for cardiovascular risk and target-organ damage in individuals with hypertension. With the emergence of new devices for assessing PWV, it has become evident that some of these devices yield results that display significant discrepancies compared with previous devices. This discrepancy underscores the importance of comprehensive validation procedures and the need for international recommendations. METHODS: A stepwise approach utilizing the modified Delphi technique, with the involvement of key scientific societies dedicated to arterial stiffness research worldwide, was adopted to formulate, through a multidisciplinary vision, a shared approach to the validation of noninvasive arterial PWV measurement devices. RESULTS: A set of recommendations has been developed, which aim to provide guidance to clinicians, researchers, and device manufacturers regarding the validation of new PWV measurement devices. The intention behind these recommendations is to ensure that the validation process can be conducted in a rigorous and consistent manner and to promote standardization and harmonization among PWV devices, thereby facilitating their widespread adoption in clinical practice. CONCLUSIONS: It is hoped that these recommendations will encourage both users and developers of PWV measurement devices to critically evaluate and validate their technologies, ultimately leading to improved consistency and comparability of results. This, in turn, will enhance the clinical utility of PWV as a valuable tool for assessing arterial stiffness and informing cardiovascular risk stratification and management in individuals with hypertension
Validation of <i>N</i>-myristoyltransferase as Potential Chemotherapeutic Target in Mammal-Dwelling Stages of <i>Trypanosoma cruzi</i>
BACKGROUND:Trypanosoma cruzi causes Chagas disease, an endemic and debilitating illness in Latin America. Lately, owing to extensive population movements, this neglected tropical disease has become a global health concern. The two clinically available drugs for the chemotherapy of Chagas disease have rather high toxicity and limited efficacy in the chronic phase of the disease, and may induce parasite resistance. The development of new anti-T. cruzi agents is therefore imperative. The enzyme N-myristoyltransferase (NMT) has recently been biochemically characterized, shown to be essential in Leishmania major, Trypanosoma brucei, and T. cruziž and proposed as promising chemotherapeutic target in these trypanosomatids. METHODOLOGY/PRINCIPAL FINDINGS:Here, using high-content imaging we assayed eight known trypanosomatid NMT inhibitors, against mammal-dwelling intracellular amastigote and trypomastigote stages and demonstrated that three of them (compounds 1, 5, and 8) have potent anti-proliferative effect at submicromolar concentrations against T. cruzi, with very low toxicity against human epithelial cells. Moreover, metabolic labeling using myristic acid, azide showed a considerable decrease in the myristoylation of proteins in parasites treated with NMT inhibitors, providing evidence of the on-target activity of the inhibitors. CONCLUSIONS/SIGNIFICANCE:Taken together, our data point out to the potential use of NMT inhibitors as anti-T. cruzi chemotherapy
Mass incarceration and neoliberal penality : a response to Lloyd and Whiteheadâs Kicked to the Curb
This paper is written in response to Lloyd and Whiteheadâs (2018) Kicked to the Curb: The triangular trade of neoliberal polity, social insecurity, and penal expul-sion. Using the âtriangular tradeâ that underpinned chattel slavery as an analytical metaphor, Lloyd and Whitehead (2018) argue that the growth of mass incarcera-tion is an endogenous feature of neoliberalism. They conclude a distinctive form of penality - neoliberal penality has developed over the past forty years. Lloyd and Whitehead (2018) propose that a tripartite model - neoliberalism, precarity and mass incarceration - as the basis for a model of neoliberal penality. This paper uses an exploration of the arguments raised by Lloyd and Whitehead (2018) to examine the links between neoliberalism and the expansion of the penal state. Whilst rec-ognising the centrality of race to these issues, the paper argues that the model that Lloyd and Whitehead (2018) present offers a partial explanation for mass incarcer-ation. This paper acknowledges that the triangular trade metaphor is a powerful one but will conclude that it has limitations. In particular, the comparison be-tween mass incarceration and chattel slavery is overstated. The economic impact of slavery and its centrality to the modern capitalism (Williams, 2014) cannot be compared to the exploitation that occurs in the current prison system. Rewriting Conjunctive Queries under Description Logic Constraints
Abstract. We consider the problems of conjunctive query answering and rewriting under Description Logic constraints. We present a query rewriting algorithm for ELHI knowledge bases, and use it to show that query answering in this setting is PTime-complete w.r.t. data complexity. We show that our algorithm is worst-case optimal for languages with data complexity of query answering ranging from LogSpace to PTimecomplete.
