    炭素窒素安定同位体比を用いた広島湾の海産生物24種の栄養段階の推定

    本研究は広島湾に生息する魚類や頭足類などの栄養段階を炭素・窒素安定同位体分析を用いて明らかにした。分析した魚類の中で最もδ15N値が低かったのはカタクチイワシとサヨリの14.4‰で,逆に高かったのはカサゴの16.8‰であった。δ13C 値が低かったのはサヨリとスズメダイの-17.6‰で,高かったのはマダイとシロギスの-15.3‰であった。頭足類を加えると,アオリイカのδ15N 値とδ13C 値は最も高く,それぞれ17.3‰と-14.8‰であった。このような種間の栄養段階の違いは,食性や栄養源の違いを反映していると考えられた。本研究結果は,瀬戸内海でも屈指の漁場として知られている広島湾において,魚類資源の持続的利用を行うために有益な知見となるであろう。Stable isotopes can provide useful knowledge about sources and processes within an ecosystem. The stable isotopes of carbon (δ13C) and nitrogen (δ15N) were used to investigate trophic relationships of relatively commercially important 21 finfish species, cephalopods in Hiroshima Bay. Among 21 finfish species, the lowest mean δ15N of 14.4‰ was recorded for Engraulis japonicus and Hyporhamphus sajori while the highest mean δ15N of 16.8‰ was recorded for Sebasticus marmoratus. The lowest and highest mean δ13C were noted -17.6‰ for Chromis notata and H. sajori, and -15.3‰ for Pagrus major and Sillago japonica, respectively. Including with cephalopods, the highest mean δ13C and δ15N noted at -14.8‰ and 17.3‰ for Sepioteuthis lessoniana. Information of stable isotope variation and trophic level in aquatic species of Hiroshima Bay can be used for monitoring and managing sustainable fisheries

    Generation of Vascular Endothelial Cells and Hematopoietic Cells by Blastocyst Complementation.

    In the case of organ transplantation accompanied by vascular anastomosis, major histocompatibility complex mismatched vascular endothelial cells become a target for graft rejection. Production of a rejection-free, transplantable organ, therefore, requires simultaneous generation of vascular endothelial cells within the organ. To generate pluripotent stem cell (PSC)-derived vascular endothelial cells, we performed blastocyst complementation with a vascular endothelial growth factor receptor-2 homozygous mutant blastocyst. This mutation is embryonic lethal at embryonic (E) day 8.5-9.5 due to an early defect in endothelial and hematopoietic cells. The Flk-1 homozygous knockout chimeric mice survived to adulthood for over 1 year without any abnormality, and all vascular endothelial cells and hematopoietic cells were derived from the injected PSCs. This approach could be used in conjunction with other gene knockouts which induce organ deficiency to produce a rejection-free, transplantable organ in which all the organ's cells and vasculature are PSC derived

    Generation of Germline-Competent Rat Induced Pluripotent Stem Cells

    Recent progress in rat pluripotent stem cell technology has been remarkable. Particularly salient is the demonstration that embryonic stem cells (ESCs) in the rat (rESCs) can contribute to germline transmission, permitting generation of gene-modified rats as is now done using mouse ESCs (mESCs) or mouse induced pluripotent stem cells (iPSCs; miPSCs). However, determinations of whether rat iPSCs (riPSCs) can contribute to germ cells are not published. Here we report the germline competency of riPSCs.We generated riPSCs by transducing three mouse reprogramming factors (Oct3/4, Klf4, and Sox2) into rat somatic cells, followed by culture in the presence of exogenous rat leukemia inhibitory factor (rLIF) and small molecules that specifically inhibit GSK3, MEK, and FGF receptor tyrosine kinases. We found that, like rESCs, our riPSCs can contribute to germline transmission. Furthermore we found, by immunostaining of testis from mouse-rat interspecific chimeras with antibody against mouse vasa homolog, that riPSCs can contribute to embryonic development with chimera formation in mice (rat-mouse interspecific chimeras) and to interspecific germlines.Our data clearly demonstrate that using only three reprogramming factors (Oct3/4, Klf4, and Sox2) rat somatic cells can be reprogrammed into a ground state. Our generated riPSCs exhibited germline transmission in either rat-rat intraspecific or mouse-rat interspecific chimeras

    Eleven Novel Polymorphic Microsatellite Loci for Oval Squid Sepioteuthis Lessoniana (Shiro-Ika Type)

    The oval squid Sepioteuthis lessoniana is one of the most economically important squid species in Japan; however, its population structure is poorly understood due to the lack of hypervariable markers. Such information is critical for managing sustainable fisheries, as well as for ensuring the existence of wild S. lessoniana stocks. Eleven candidate microsatellite loci were isolated from a small insert genomic DNA library. Polymorphisms in these 11 loci were screened in 24 wild individuals. The number of alleles per locus was found to range from 5 to 19 alleles, and the observed heterozygosity ranged from 0.292 to 0.958. No evidence for linkage disequilibrium was detected among all the loci. The genotypic proportions conformed to Hardy-Weinberg equilibrium, except at one locus. In conclusion, these polymorphic microsatellite loci may be used to develop a genetic framework to manage S. lessoniana in the future

    Development of Wearable Sheet-Type Shear Force Sensor and Measurement System that is Insusceptible to Temperature and Pressure

    A sheet-type shear force sensor and a measurement system for the sensor were developed. The sensor has an original structure where a liquid electrolyte is filled in a space composed of two electrode-patterned polymer films and an elastic rubber ring. When a shear force is applied on the surface of the sensor, the two electrode-patterned films mutually move so that the distance between the internal electrodes of the sensor changes, resulting in current increase or decrease between the electrodes. Therefore, the shear force can be calculated by monitoring the current between the electrodes. Moreover, it is possible to measure two-dimensional shear force given that the sensor has multiple electrodes. The diameter and thickness of the sensor head were 10 mm and 0.7 mm, respectively. Additionally, we also developed a measurement system that drives the sensor, corrects the baseline of the raw sensor output, displays data, and stores data as a computer file. Though the raw sensor output was considerably affected by the surrounding temperature, the influence of temperature was drastically decreased by introducing a simple arithmetical calculation. Moreover, the influence of pressure simultaneously decreased after the same calculation process. A demonstrative measurement using the sensor revealed the practical usefulness for on-site monitoring

    Pathological complete response of initially unresectable multiple liver metastases achieved using combined peptide receptor radionuclide therapy and somatostatin analogs following pancreatic neuroendocrine tumor resection: a case report

    Abstract Background Peptide receptor radionuclide therapy (PRRT) serves as a novel and effective treatment option for somatostatin receptor-positive unresectable liver metastases of pancreatic neuroendocrine tumors (PNETs). However, there are few reported cases of surgical resection for initially unresectable liver metastases of PNET that were converted to resectable after PRRT. Here we report a case where PRRT and somatostatin analogs (SSAs) led to a pathological complete response of initially unresectable multiple liver metastases following PNET resection. Case presentation A 52-year-old man underwent pylorus-preserving pancreaticoduodenectomy for PNET at age 40 and subsequent hepatectomies for resectable liver metastases at 44 and 47 years of age. At age 48, a follow-up examination revealed unresectable multiple liver metastases, and PRRT with 177Lu-DOTATATE therapy was initiated. After four cycles of PRRT, most liver metastases diminished according to imaging studies, and the remaining two hepatic lesions continued to shrink with additional lanreotide. Conversion surgery for liver metastases was successfully performed, revealing no viable tumor cells in tissue specimens. Seventeen months after surgery, imaging showed no detectable residual tumor or recurrence. We present a review of the relevant literature that highlights the significance of our findings. Conclusions This rare case highlights the pathological complete response of initially unresectable multiple liver metastases achieved by PRRT and SSAs following PNET resection, suggesting their potential as a multimodality treatment option for unresectable PNET
