    A fluorescence anisotropy study of tetramer-dimer equilibrium of λ repressor and its implication for function

    Tetramer-dimer equilibrium of λ repressor has been studied by fluorescence anisotropy techniques. We have chosen 1-dimethylamino naphthalene-5-sulfonyl chloride (dansyl chloride)-labeled repressor to study the dissociation-association equilibrium, because of relatively long life-time of the probe (> 10 ns). Polarization of the dansyl-labeled repressor decreases with decreasing protein concentrations in the range of 20 to 0.2 μM. The decrease of anisotropy was shown to be due to reversible dissociation of the protein. Size exclusion high-performance liquid chromatography studies and polyacrylamide gel electrophoresis under native conditions (Ferguson plot) confirmed that at around 20 μM concentrations the repressor exists in predominantly tetrameric form, whereas in lower concentrations it exists in predominantly dimer form. A dissociation constant of 2.3 ± 0.9 μM was estimated in 0.1 M potassium phosphate, pH 8.0, at 25° C. A stoichiometric amount of isolated single operator shifted the tetramer-dimer equilibrium toward the dimer. Increased ionic strength had only a modest effect on the dissociation constant. The thermodynamic constants for the dissociation reaction calculated from the Van't Hoff plot was +26.6 kcal/mol for ΔH and +64.7 e.u. for ΔS. The rotational correlation times derived from isothermal Perrin plot indicated elongated dimers and tetramers

    Search for optical bursts from the gamma ray burst source GBS 0526-66

    Attempts were made to detect optical bursts from the gamma-ray burst source GBS 0526-66 during Dec. 31, 1984 to Jan. 2, 1985 and Feb. 23 to Feb. 24, 1985, using the one meter reflector of the Kavalur Observatory. Jan. 1, 1985 coincided with the zero phase of the predicted 164 day period of burst activity from the source (Rothschild and Lingenfelter, 1984). A new optical burst photon counting system with adjustable trigger threshold was used in parallel with a high speed photometer for the observations. The best time resolution was 1 ms and maximum count rate capability was 255,000 counts s(-1). Details of the instrumentation and observational results are presented

    An operator-induced conformational change in the C-terminal domain of the λ repressor

    4,4'-bis(1-anilino-8-naphthalenesulfonic acid (Bis-ANS), an environment-sensitive fluorescent probe for hydrophobic region of proteins, binds specifically to the C-terminal domain of λ repressor. The binding is characterized by positive cooperativity, the magnitude of which is dependent on protein concentration in the concentration range where dimeric repressor aggregates to a tetramer. In this range, positive cooperativity becomes more pronounced at higher protein concentrations. This suggests a preferential binding of Bis-ANS to the dimeric form of the repressor. Binding of single operator OR1 to the N-terminal domain of the repressor causes enhancement of fluorescence of the C-terminal domain bound Bis-ANS. The binding of single operator OR1 also leads to quenching of fluorescence of tryptophan residues, all of which are located in the hinge or the C-terminal domain. Thus two different fluorescent probes indicate an operator-induced conformational change which affects the C-terminal domain. The significance of this conformational change with respect to the function of λ repressor has been discussed

    One-Dimensional Approximation of Viscous Flows

    Attention has been paid to the similarity and duality between the Gregory-Laflamme instability of black strings and the Rayleigh-Plateau instability of extended fluids. In this paper, we derive a set of simple (1+1)-dimensional equations from the Navier-Stokes equations describing thin flows of (non-relativistic and incompressible) viscous fluids. This formulation, a generalization of the theory of drop formation by Eggers and his collaborators, would make it possible to examine the final fate of Rayleigh-Plateau instability, its dimensional dependence, and possible self-similar behaviors before and after the drop formation, in the context of fluid/gravity correspondence.Comment: 17 pages, 3 figures; v2: refs & comments adde

    Atomic Configuration of Nitrogen Doped Single-Walled Carbon Nanotubes

    Having access to the chemical environment at the atomic level of a dopant in a nanostructure is crucial for the understanding of its properties. We have performed atomically-resolved electron energy-loss spectroscopy to detect individual nitrogen dopants in single-walled carbon nanotubes and compared with first principles calculations. We demonstrate that nitrogen doping occurs as single atoms in different bonding configurations: graphitic-like and pyrrolic-like substitutional nitrogen neighbouring local lattice distortion such as Stone-Thrower-Wales defects. The stability under the electron beam of these nanotubes has been studied in two extreme cases of nitrogen incorporation content and configuration.     Vortices in (2+1)d Conformal Fluids

    We study isolated, stationary, axially symmetric vortex solutions in (2+1)-dimensional viscous conformal fluids. The equations describing them can be brought to the form of three coupled first order ODEs for the radial and rotational velocities and the temperature. They have a rich space of solutions characterized by the radial energy and angular momentum fluxes. We do a detailed study of the phases in the one-parameter family of solutions with no energy flux. This parameter is the product of the asymptotic vorticity and temperature. When it is large, the radial fluid velocity reaches the speed of light at a finite inner radius. When it is below a critical value, the velocity is everywhere bounded, but at the origin there is a discontinuity.     Studies on Medium Grade Ilmenite-Depleted Beach Placer deposit of Chavakkad Ponnani Coast, Kerala

    In the present paper, characterization, beneficiation and further value addition studies carried out with samples collected from unexploited Chavakkad ponnani (CP) area of Kerala coast have been reported. Characterization studies involve size and chemical analyses, sink and float studies and optical microscopy. total 21 samples were collected from 18 km. stretch of CP area at almost 1 km. apart. While sink and float studies and optical microscopy were carried on with all individual samples, detail characterization and beneficiation studies were undertaken with composite samples-one prepared from all 21 samples and the second from few selected samples. Pre-concentration followed by high tension separation and magnetic separation yielded product containing more than 95% THM with around 90% recovery. Further value addition studies with generated ilmenite concentrate was found to produce synthetic rutile, through reduction followed by oxidation and leaching, containing more than 94% TiO2
