    Association study of the vesicular monoamine transporter 1 (VMAT1) gene with schizophrenia in a Japanese population

    BACKGROUND: Vesicular monoamine transporters (VMATs) mediate accumulation of monoamines such as serotonin, dopamine, adrenaline, and noradrenaline from the cytoplasm into storage organelles. The VMAT1 (alternatively solute carrier family 18: SLC18A1) regulates such biogenic amines in neuroendocrine systems. The VMAT1 gene maps to chromosome 8p21.3, a locus with strong evidence of linkage with schizophrenia. A recent study reported that a non-synonymous single nucleotide polymorphism (SNP) of the gene (Pro4Thr) was associated with schizophrenia. METHODS: We attempted to replicate this finding in a Japanese sample of 354 schizophrenics and 365 controls. In addition, we examined 3 other non-synonymous SNPs (Thr98Ser, Thr136Ile, and Val392Leu). Genotyping was performed by the TaqMan allelic discrimination assay. RESULTS: There was no significant difference in genotype or allele distribution of the three SNPs of Pro4Thr, Thr136Ile, or Val392Leu between patients and controls. There was, however, a significant difference in genotype and allele distributions for the Thr98Ser polymorphism between the two groups (P = 0.01 for genotype and allele). When sexes were examined separately, significant differences were observed in females (P = 0.006 for genotype, P = 0.003 for allele), but not in males. The Thr98 allele was more common in female patients than in female controls (odds ratio 1.69, 95% CI 1.19–2.40, P = 0.003). Haplotype-based analyses also provided evidence for a significant association in females. CONCLUSION: We failed to replicate the previously reported association of Pro4Thr of the VMAT1 gene with schizophrenia. However, we obtained evidence for a possible role of the Thr98Ser in giving susceptibility to schizophrenia in women

    Canonical versus non-canonical transsynaptic signaling of neuroligin 3 tunes development of sociality in mice

    社会性の発達を調節する新たな機構を発見. 京都大学プレスリリース. 2021-03-26.Neuroligin 3 (NLGN3) and neurexins (NRXNs) constitute a canonical transsynaptic cell-adhesion pair, which has been implicated in autism. In autism spectrum disorder (ASD) development of sociality can be impaired. However, the molecular mechanism underlying NLGN3-mediated social development is unclear. Here, we identify non-canonical interactions between NLGN3 and protein tyrosine phosphatase δ (PTPδ) splice variants, competing with NRXN binding. NLGN3-PTPδ complex structure revealed a splicing-dependent interaction mode and competition mechanism between PTPδ and NRXNs. Mice carrying a NLGN3 mutation that selectively impairs NLGN3-NRXN interaction show increased sociability, whereas mice where the NLGN3-PTPδ interaction is impaired exhibit impaired social behavior and enhanced motor learning, with imbalance in excitatory/inhibitory synaptic protein expressions, as reported in the Nlgn3 R451C autism model. At neuronal level, the autism-related Nlgn3 R451C mutation causes selective impairment in the non-canonical pathway. Our findings suggest that canonical and non-canonical NLGN3 pathways compete and regulate the development of sociality

    Measurement of submaximal aerobic power in children

    PWC150 in 935 school children whose ages were ranged from 6 to 17 yrs, was measured using a computarized bicycle ergometer. The absolute values of boys increased with age, while thase of girls increased in like mannar until 11 yrs, but maintained the almost same level over 11 yrs. Sex difference was observed in the relative value of PWC150 to body weight. It was concluded that PWC150 was available for easy assessment of submaximal aerobic power in children although further researches were required for obtaining the reliable norm

    Post-intervention Status in Patients With Refractory Myasthenia Gravis Treated With Eculizumab During REGAIN and Its Open-Label Extension

    OBJECTIVE: To evaluate whether eculizumab helps patients with anti-acetylcholine receptor-positive (AChR+) refractory generalized myasthenia gravis (gMG) achieve the Myasthenia Gravis Foundation of America (MGFA) post-intervention status of minimal manifestations (MM), we assessed patients' status throughout REGAIN (Safety and Efficacy of Eculizumab in AChR+ Refractory Generalized Myasthenia Gravis) and its open-label extension. METHODS: Patients who completed the REGAIN randomized controlled trial and continued into the open-label extension were included in this tertiary endpoint analysis. Patients were assessed for the MGFA post-intervention status of improved, unchanged, worse, MM, and pharmacologic remission at defined time points during REGAIN and through week 130 of the open-label study. RESULTS: A total of 117 patients completed REGAIN and continued into the open-label study (eculizumab/eculizumab: 56; placebo/eculizumab: 61). At week 26 of REGAIN, more eculizumab-treated patients than placebo-treated patients achieved a status of improved (60.7% vs 41.7%) or MM (25.0% vs 13.3%; common OR: 2.3; 95% CI: 1.1-4.5). After 130 weeks of eculizumab treatment, 88.0% of patients achieved improved status and 57.3% of patients achieved MM status. The safety profile of eculizumab was consistent with its known profile and no new safety signals were detected. CONCLUSION: Eculizumab led to rapid and sustained achievement of MM in patients with AChR+ refractory gMG. These findings support the use of eculizumab in this previously difficult-to-treat patient population. CLINICALTRIALSGOV IDENTIFIER: REGAIN, NCT01997229; REGAIN open-label extension, NCT02301624.     Evaluation of Wearable Cameras for Monitoring and Analyzing Calf Behavior: A Preliminary Study

    Understanding cattle behavior is important for discerning their health and management status. However, manual observations of cattle are time-consuming and labor-intensive. Moreover, during manual observations, the presence or position of a human observer may alter the normal behavior of the cattle. Wearable cameras are small and lightweight; therefore, they do not disturb cattle behavior when attached to their bodies. Thus, this study aimed to evaluate the suitability of wearable cameras for monitoring and analyzing cattle behavior. From December 18 to 27, 2017, this study used four 2-month-old, group-housed Holstein calves at the Field Science Center of the Obihiro University of Agriculture and Veterinary Medicine, Japan. Calf behavior was recorded every 30 s using a wearable camera (HX-A1H, Panasonic, Japan) from 10:00 to 15:30 and observed directly from 11:00 to 12:00 and 14:00 to 15:00. In addition, the same observer viewed the camera recordings corresponding to the direct observation periods, and the results were compared. The correlation coefficients of all behavioral data from direct and wearable camera video observations were significant (p < 0.01). We conclude that wearable cameras are suitable for observing calf behavior, particularly their posture (standing or lying), as well as their ruminating and feeding behaviors

    Awareness Communications by Entertaining Toy Doll Agents

    In this paper, we propose a sensor-doll system that provides multiple users at remote locations with an awareness communication channel. A doll is used as the interface agent of the local user, and this agent is connected to a remote doll by local and/or wide area networks. The doll sends out information on the local ambient activities and user’s intentional interactions to the remote agent and, at the same time, displays the received remote activities by adapting its presentation to the local context. Musical sound expression is used to display the remote awareness, mixing the local response and remote activities. Music also provides an entertaining and sympathetic intimacy with the doll and eventually the remote user. The design and implementation of the networked sensordoll, equipped with various tactile sensors and a PC, are described in detail. We also discuss issues of awareness communication and give preliminary experimental results
