Existence theorems for a crystal surface model involving the p-Laplace operator
The manufacturing of crystal films lies at the heart of modern
nanotechnology. How to accurately predict the motion of a crystal surface is of
fundamental importance. Many continuum models have been developed for this
purpose, including a number of PDE models, which are often obtained as the
continuum limit of a family of kinetic Monte Carlo models of crystal surface
relaxation that includes both the solid-on-solid and discrete Gaussian models.
In this paper we offer an analytical perspective into some of these models. To
be specific, we study the existence of a weak solution to the boundary value
problem for the equation - \Delta e^{-\mbox{div}\left(|\nabla u|^{p-2}\nabla
u\right)}+au=f, where are given numbers and is a given
function. This problem is derived from a crystal surface model proposed by
J.L.~Marzuola and J.~Weare (2013 Physical Review, E 88, 032403). The
mathematical challenge is due to the fact that the principal term in our
equation is an exponential function of a p-Laplacian. Existence of a

Ustekinumab treats psoriasis by suppressing RORC and T-box but its suppression of GATA restrains its efficacy
Psoriasis is a T-cell mediated disease that involves IL-23/Th17 and IL-12/Th1 axes. Ustekinumab, a fully human monoclonal antibody targeting the p40 subunit of both IL-12 and IL-23, has proven to be efficient and safe for treating patients with psoriasis. Yet, there have been no reports with human skin/blood samples that would elucidate the molecular mechanisms by which ustekinumab calms psoriasis skin lesions. To investigate the efficacy and molecular pathway (RORC, t-BOX and GATA) of ustekinumab in treating patients with psoriasis skin lesions. A total of 30 patients with psoriasis were randomized into placebo group and treatment group. PASI of each patient was calculated at 0, 12 and 24 weeks post-treatment. The mRNA levels of RORC, t-BOX and GATA in peripheral blood mononuclear cells separated from patientsâ whole blood were analyzed using qPCR. Decreased mRNA of RORC, t-BOX and GATA were observed after continuous injections, indicating that ustekinumab exerts its effect by interacting with these molecules; while no significant difference in foxp3 mRNA levels were found between placebo group and treatment group
Analysis of Agreement on Traditional Chinese Medical Diagnostics for Many Practitioners
In Traditional Chinese Medicine (TCM) diagnostics, it is an important issue to study the degree of agreement among several distinct practitioners. In order to study the reliability of TCM diagnostics, we have to design an experiment to simultaneously deal with both of the cases when the data is ordinal and when there are many TCM practitioners. In this study, we consider a reliability measure called âKrippendorff's alphaâ to investigate the agreement of tongue diagnostics in TCM. Besides, since it is not easy to obtain a large data set with patients rated simultaneously by many TCM practitioners, we use the renowned âbootstrappingâ to obtain a 95% confidence interval for the Krippendorff's alpha. The estimated Krippendorff's alpha for the agreement among ten physicians that discerned fifteen randomly chosen patients is 0.7343, and the 95% bootstrapping confidence interval for the true alpha coefficient is [0.6570, 0.7349]. A combined targeted mutation analysis of IRF6 gene would be useful in the first screening of oral facial clefts
BACKGROUND: Interferon Regulatory Factor 6 (IRF6) is a member of the IRF family of transcription factors. It has been suggested to be an important contributor to orofacial development since mutations of the IRF6 gene has been found in Van der Woude (VWS) and popliteal pterygium syndromes (PPS), two disorders that can present with isolated cleft lip and palate. The association between IRF6 gene and cleft lip and palate has also been independently replicated in many populations. METHODS: We screened a total of 155 Taiwanese patients with cleft lip with or without cleft palate (CL/P); 31 syndromic (including 19 VWS families), 44 non-syndromic families with at least two affected members, and 80 non-syndromic patients through a combined targeted, polymerase chain reaction (PCR)-based mutation analysis for the entire coding regions of IRF6 gene. RESULTS: We found 11 mutations in 57.89% (11/19) of the VWS patients and no IRF6 mutation in 44 of the non-syndromic multiplex families and 80 non-syndromic oral cleft patients. In this IRF6 gene screening, five of these mutations (c.290 A>G, p.Tyr97Cys; c.360-375 16Â bp deletion, p.Gln120HisfsX24; c.411_412 insA, p.Glu136fsX3; c.871 A>C, p.Thr291Pro; c.969Â G>A, and p.Trp323X) have not been reported in the literature previously. Exon deletion was not detected in this series of IRF6 gene screening. CONCLUSIONS: Our results confirm the crucial role of IRF6 in the VWS patients and further work is needed to explore for its function in the non-syndromic oral cleft with vary clinical features
Unexpected sawtooth artifact in beat-to-beat pulse transit time measured from patient monitor data
Object: It is increasingly popular to collect as much data as possible in the
hospital setting from clinical monitors for research purposes. However, in this
setup the data calibration issue is often not discussed and, rather, implicitly
assumed, while the clinical monitors might not be designed for the data
analysis purpose. We hypothesize that this calibration issue for a secondary
analysis may become an important source of artifacts in patient monitor data.
We test an off-the-shelf integrated photoplethysmography (PPG) and
electrocardiogram (ECG) monitoring device for its ability to yield a reliable
pulse transit time (PTT) signal. Approach: This is a retrospective clinical
study using two databases: one containing 35 subjects who underwent
laparoscopic cholecystectomy, another containing 22 subjects who underwent
spontaneous breathing test in the intensive care unit. All data sets include
recordings of PPG and ECG using a commonly deployed patient monitor. We
calculated the PTT signal offline. Main Results: We report a novel constant
oscillatory pattern in the PTT signal and identify this pattern as a sawtooth
artifact. We apply an approach based on the de-shape method to visualize,
quantify and validate this sawtooth artifact. Significance: The PPG and ECG
signals not designed for the PTT evaluation may contain unwanted artifacts. The
PTT signal should be calibrated before analysis to avoid erroneous
interpretation of its physiological meaning
