    Conservation of rare and valuable books in our country has a long tradition. Since 2001, work on identification and state recording of book monuments is one of the priority directions of State Cultural Policy of Russia. Functions of organizational and scientific-methodological center are being performed by the Russian State Library.By now there is created scientific-methodological and organizational basis of the system of state registration of book monuments. There are adopted legal rules governing the state registration of book monuments, however its implementation involves significant legal and organizational problems. For further development of system of state recording of book monuments and solution of the existing problems, at the Government support there is created the All-Russian Corpus of Book Monuments, there is continued scientific and applied research on its identification and description, implemented and improved the technology of automated record-keeping, there is executed training of personnel capable to implement system approach in work with book monuments.Еятельность по сохранению редких и ценных книг в нашей стране имеет давние традиции. С 2001 г. работа по выявлению и государственному учету книжных памятников является одним из приоритетных направлений государственной культурной политики России. Функции организационного и научно-методического центра выполняет Российская государственная библиотека. К настоящему времени созданы научно-методические и организационные основы системы государственной регистрации книжных памятников. Приняты юридические нормы, определяющие порядок государственной регистрации книжных памятников, однако их исполнение сопряжено со значительными юридическими и организационными проблемами. Для дальнейшего развития системы их государственного учета при поддержке государства создается Общероссийский свод книжных памятников, продолжаются научные и прикладные разработки по вопросам их выявления и описания, внедряется и совершенствуется технология автоматизированного учета, ведется профессиональная подготовка кадров, способных реализовать системный подход в работе с книжными памятниками

    There is presented an overview on the 10th All-Russian Meeting of the Heads of Information Services for Culture and Art “Libraries in the information support for realization of state cultural policy”, held on 3—4 October, 2017, and organized by the Russian State Library in the partnership with the Saint-Petersburg State Institute of Culture. The central theme of the discussion was the development of information involvement of the regional libraries in the implementation of state cultural policy, enhancement of the scientific information activity of libraries in the sphere of culture and art, and increase of their innovation capacity. Within the frames of the meeting there was held a webinar on the topic “Modern information resources on culture and art”; there also took place the award ceremony of the winners of the 6th All-Russian competition for the best e-publication on culture and art, which was attended by 63 state and municipal libraries from 44 regions of Russia. At the special meeting of libraries — participants of the Russian system of scientific and information support in the sphere of culture (Rosinformkultura) there was announced on the adoption of a new edition of the Regulations on Rosinformkultura, and there was elected its Board. The Final document calls to promote the development of electronic information resources on culture and art, to ensure the relevance of information and its accessibility on the Internet, and actively participate in the projects of Rosinformkultura.Представлен обзор X Всероссийского совещания руководителей служб информации по культуре и искусству «Библиотеки в информационном обеспечении реализации государственной культурной политики», которое состоялось 3—4 октября 2017 г., и было организовано Российской государственной библиотекой в партнерстве с Санкт-Петербургским государственным институтом культуры. Центральной темой совещания стали вопросы развития информационного участия региональных библиотек в реализации государственной культурной политики, совершенствования научно-информационной деятельности библиотек в сфере культуры и искусства, повышения их инновационной активности. В рамках совещания был проведен вебинар на тему «Современные информационные ресурсы по культуре и искусству», состоялось торжественное награждение победителей и лауреатов VI Всероссийского смотра-конкурса на лучшее электронное издание по культуре и искусству, в котором участвовали 63 государственные и муниципальные библиотеки из 44 регионов России. На специальном заседании библиотек — участников Российской системы научно-информационного обеспечения сферы культуры (Росинформкультуры)было объявлено о принятии новой редакции Положения о Росинформкультуре и был избран ее Совет. В Итоговом документе заключен призыв способствовать развитию электронных информационных ресурсов по культуре и искусству, обеспечивать актуальность информации и ее доступность в Интернете, активнее участвовать в проектах Росинформкультуры

    Measurement of Calcium Dissociation Rates from Troponin C in Rigor Skeletal Myofibrils

    Ca2+ dissociation from the regulatory domain of troponin C may influence the rate of striated muscle relaxation. However, Ca2+ dissociation from troponin C has not been measured within the geometric and stoichiometric constraints of the muscle fiber. Here we report the rates of Ca2+ dissociation from the N-terminal regulatory and C-terminal structural domains of fluorescent troponin C constructs reconstituted into rabbit rigor psoas myofibrils using stopped-flow technology. Chicken skeletal troponin C fluorescently labeled at Cys 101, troponin CIAEDANS, reported Ca2+ dissociation exclusively from the structural domain of troponin C at ∼0.37, 0.06, and 0.07/s in isolation, in the presence of troponin I and in myofibrils at 15°C, respectively. Ca2+ dissociation from the regulatory domain was observed utilizing fluorescently labeled troponin C containing the T54C and C101S mutations. Troponin CMIANST54C,C101S reported Ca2+ dissociation exclusively from the regulatory domain of troponin C at >1000, 8.8, and 15/s in isolation, in the presence of troponin I and in myofibrils at 15°C, respectively. Interestingly, troponin CIAANST54C,C101S reported a biphasic fluorescence change upon Ca2+ dissociation from the N- and C-terminal domains of troponin C with rates that were similar to those reported by troponin CMIANST54C,C101S and troponin CIAEDANS at all levels of the troponin C systems. Furthermore, the rate of Ca2+ dissociation from troponin C in the myofibrils was similar to the rate of Ca2+ dissociation measured from the troponin C-troponin I complexes. Since the rate of Ca2+ dissociation from the regulatory domain of TnC in myofibrils is similar to the rate of skeletal muscle relaxation, Ca2+ dissociation from troponin C may influence relaxation

    Catecholaminergic polymorphic ventricular tachycardia (CPVT) is a familial arrhythmogenic syndrome characterized by sudden death. There are several genetic forms of CPVT associated with mutations in genes encoding the cardiac ryanodine receptor (RyR2) and its auxiliary proteins including calsequestrin (CASQ2) and calmodulin (CaM). It has been suggested that impairment of the ability of RyR2 to stay closed (ie, refractory) during diastole may be a common mechanism for these diseases.     Disease-Related Cardiac Troponins Alter Thin Filament Ca2+ Association and Dissociation Rates

    The contractile response of the heart can be altered by disease-related protein modifications to numerous contractile proteins. By utilizing an IAANS labeled fluorescent troponin C, , we examined the effects of ten disease-related troponin modifications on the Ca2+ binding properties of the troponin complex and the reconstituted thin filament. The selected modifications are associated with a broad range of cardiac diseases: three subtypes of familial cardiomyopathies (dilated, hypertrophic and restrictive) and ischemia-reperfusion injury. Consistent with previous studies, the majority of the protein modifications had no effect on the Ca2+ binding properties of the isolated troponin complex. However, when incorporated into the thin filament, dilated cardiomyopathy mutations desensitized (up to 3.3-fold), while hypertrophic and restrictive cardiomyopathy mutations, and ischemia-induced truncation of troponin I, sensitized the thin filament to Ca2+ (up to 6.3-fold). Kinetically, the dilated cardiomyopathy mutations increased the rate of Ca2+ dissociation from the thin filament (up to 2.5-fold), while the hypertrophic and restrictive cardiomyopathy mutations, and the ischemia-induced truncation of troponin I decreased the rate (up to 2-fold). The protein modifications also increased (up to 5.4-fold) or decreased (up to 2.5-fold) the apparent rate of Ca2+ association to the thin filament. Thus, the disease-related protein modifications alter Ca2+ binding by influencing both the association and dissociation rates of thin filament Ca2+ exchange. These alterations in Ca2+ exchange kinetics influenced the response of the thin filament to artificial Ca2+ transients generated in a stopped-flow apparatus. Troponin C may act as a hub, sensing physiological and pathological stimuli to modulate the Ca2+-binding properties of the thin filament and influence the contractile performance of the heart

    The article is devoted to the modern period of research activity of the Russian State Library ‒ one of the Russian Federation’s national libraries, which, in accordance with the current legislation, are research institutions in library science, bibliography and book studies, and methodological, scientific-information and cultural centers of federal importance, participating in development and implementation of the federal policy in the field of librarianship. The article presents the main directions and subjects of the RSL’s research work in 2015–2018. In addition to traditional areas (the history, theory and practice of librarianship, the disclosure of collections, and the study of monuments of book culture), there have also been developed some research projects aimed at solving current issues of library development in the digital environment (the formation and organization of access to electronic information resources, the creation of digital services, etc.). The article demonstrates major publications of the RSL employees, representing the research results (monographs and articles, catalogues, bibliographic indexes, manuals and reference books). The article analyzes the innovations in organizing scientific activities and the achieved results (including scientometric characteristics), which confirm the status of the Russian State Library as a leading research center in the field of library science, bibliography and book studies.В статье освещён новейший период научно-исследовательской деятельности Российской государственной библиотеки ‒ одной из национальных библиотек Российской Федерации, которые в соответствии с действующим законодательством являются научно-исследовательскими учреждениями в таких сферах, как библиотековедение, библиографоведение и книговедение; методическими, научно-информационными и культурными центрами федерального значения; участвуют в разработке и реализации федеральной политики в области библиотечного дела. Представлены основные направления и тематика научно-исследовательской работы РГБ в 2015‒2018 гг. Кроме традиционных направлений (история, теория и практика библиотечного дела, раскрытие фондов и изучение памятников книжной культуры) развивались исследовательские проекты, направленные на решение актуальных проблем библиотечного развития в цифровой среде (формирование электронных информационных ресурсов и доступ к ним, создание цифровых сервисов и т.д.). Представлены основные публикации сотрудников РГБ о результатах проведённых исследований. Проанализированы новации в организации научной деятельности и результаты их внедрения, которые подтверждают статус РГБ как ведущего научного центра в области библиотековедения, библиографоведения и книговедения. Подчёркнуто, что результаты исследований публикуются в статусных научных изданиях, что обеспечивает библиотеке авторитет в академической среде и высокие наукометрические показатели
