    No variation of physical performance and perceived exertion after adrenal gland stimulation by synthetic ACTH (Synacthen®) in cyclists

    There is anecdotal evidence that athletes use the banned substance Synacthen® because of its perceived benefit with its associated rise in cortisol. To test the performance-enhancing effects of Synacthen®, eight trained cyclists completed two, 2-day exercise sessions separated by 7-10days. On the first day of each 2-day exercise session, subjects received either Synacthen® (0.25mg, TX) or placebo (PLA) injection. Performance was assessed by a 20-km time trial (TT) after a 90-min fatigue period on day 1 and without the fatiguing protocol on day 2. Plasma androgens and ACTH concentrations were measured during the exercise bouts as well as the rate of perceived exertion (RPE). Spot urines were analyzed for androgens and glucocorticoids quantification. Basal plasma hormones did not differ significantly between PLA and TX groups before and 24h after the IM injection (P>0.05). After TX injection, ACTH peaked at 30min and hormone profiles were significantly different compared to the PLA trial (P<0.001). RPE increased significantly in both groups as the exercise sessions progressed (P<0.001) but was not influenced by treatment. The time to completion of the TT was not affected on both days by Synacthen® treatment. In the present study, a single IM injection of synthetic ACTH did not improve either acute or subsequent cycling performance and did not influence perceived exertion.     The Rapidly Flaring Afterglow of the Very Bright and Energetic GRB 070125

    We report on multi-wavelength observations, ranging from the X-ray to radio wave bands, of the IPN-localized gamma-ray burst GRB 070125. Spectroscopic observations reveal the presence of absorption lines due to O I, Si II, and C IV, implying a likely redshift of z = 1.547. The well-sampled light curves, in particular from 0.5 to 4 days after the burst, suggest a jet break at 3.7 days, corresponding to a jet opening angle of ~7.0 degrees, and implying an intrinsic GRB energy in the 1 - 10,000 keV band of around E = (6.3 - 6.9)x 10^(51) erg (based on the fluences measured by the gamma-ray detectors of the IPN network). GRB 070125 is among the brightest afterglows observed to date. The spectral energy distribution implies a host extinction of Av < 0.9 mag. Two rebrightening episodes are observed, one with excellent time coverage, showing an increase in flux of 56% in ~8000 seconds. The evolution of the afterglow light curve is achromatic at all times. Late-time observations of the afterglow do not show evidence for emission from an underlying host galaxy or supernova. Any host galaxy would be subluminous, consistent with current GRB host-galaxy samples. Evidence for strong Mg II absorption features is not found, which is perhaps surprising in view of the relatively high redshift of this burst and the high likelihood for such features along GRB-selected lines of sight.Comment: 50 pages, 9 figures, 5 tables Accepted to the Astrophysical Journa

    HST and Optical Data Reveal White Dwarf Cooling, Spin and Periodicities in GW Librae 3-4 Years after Outburst

    Since the large amplitude 2007 outburst which heated its accreting, pulsating white dwarf, the dwarf nova system GW Librae has been cooling to its quiescent temperature. Our Hubble Space Telescope ultraviolet spectra combined with ground-based optical coverage during the 3rd and 4th year after outburst show that the fluxes and temperatures are still higher than quiescence (T=19,700K and 17,300K vs 16,000K pre-outburst for a log g=8.7 and d=100 pc). The K{wd} of 7.6+/-0.8 km/s determined from the CI1463 absorption line, as well as the gravitational redshift implies a white dwarf mass of 0.79+/-0.08 Msun. The widths of the UV lines imply a white dwarf rotation velocity vsin i of 40 km/s and a spin period of 209 s (for an inclination of 11 deg and a white dwarf radius of 7x10^{8} cm). Light curves produced from the UV spectra in both years show a prominent multiplet near 290 s, with higher amplitude in the UV compared to the optical, and increased amplitude in 2011 vs 2010. As the presence of this set of periods is intermittent in the optical on weekly timescales, it is unclear how this relates to the non-radial pulsations evident during quiescence.Comment: 36 pages, 15 figures; Astrophysical Journal, 2012, 75

    Galex and optical observations of GW librae during the long decline from superoutburst

    The prototype of accreting, pulsating white dwarfs (GW Lib) underwent a large amplitude dwarf nova outburst in 2007. We used ultraviolet data from Galaxy Evolution Explorer and ground-based optical photometry and spectroscopy to follow GW Lib for three years following this outburst. Several variations are apparent during this interval. The optical shows a superhump modulation in the months following outburst, while a 19 minute quasi-periodic modulation lasting for several months is apparent in the year after outburst. A long timescale (about 4 hr) modulation first appears in the UV a year after outburst and increases in amplitude in the following years. This variation also appears in the optical two years after outburst but is not in phase with the UV. The pre-outburst pulsations are not yet visible after three years, likely indicating the white dwarf has not returned to its quiescent state

    Cortisol urinary metabolites in dogs with hypercortisolism, congestive heart failure and healthy dogs

    Nonadrenal diseases (NAD), including congestive heart failure (CHF), can affect the conversion of cortisone to cortisol favoring the production of cortisol's urinary downstream metabolites 5α/5β-tetrahydrocortisol (THF) relative to tetrahydrocortisone (THE). We hypothesized that healthy dogs would have lower urinary levels of cortisol, cortisone, THF, and THE than dogs with hypercortisolism (HC) or CHF, and the latter would have higher urinary levels of THF and lower THE than dogs with HC. Four, 9, and 8 dogs with HC, CHF, and normal health, respectively, were included in a pilot prospective cross-sectional study. A single morning voided urine sample was analyzed for urinary cortisol metabolites by liquid chromatography–mass spectrometry. The percentages of conjugated urinary metabolites were significantly higher in dogs with CHF than in healthy dogs (p=0.001), and not different in HC dogs (p=0.07). Log-transformed urine cortisol metabolites–to–creatinine ratios in healthy dogs were significantly lower than the 2 other groups (p<0.001). The urinary free THE:THF ratio was significantly higher (p<0.001) than the urinary total and conjugated THE:THF ratios. Health status did not affect the total, conjugated, and free THE:THF ratios (p=0.61). Additional studies are needed to investigate differences in cortisol metabolites between dogs with HC and NAD to accurately discriminate between the groups

    Short Communication Effects of periconceptional undernutrition on maternal taurine concentrations in sheep

    Abstract Taurine has an important role in numerous physiological processes, including many aspects of fetal development such as development of the pancreas and brain, and requirements increase during pregnancy. Periconceptional undernutrition has long-term effects on pancreas and brain function of the offspring, but the effects on maternal taurine economy are unknown. We, therefore, studied the effects of different periods of periconceptional undernutrition on maternal plasma and urine taurine concentrations before and during pregnancy. Four groups of singleton-bearing ewes were studied (n 10-11): controls fed ad libitum, and groups undernourished from 60 d before until mating (PreC), from 2 d before mating until 30 d after mating (PostC) or from 60 d before until 30 d after mating (PreĂľPostC). In PreC ewes, plasma taurine concentrations remained at control levels for the first 30 d, and then decreased through the remainder of undernutrition, but recovered by 30 d after mating; urinary taurine excretion was low at mating, but recovered similarly. In PostC ewes, plasma taurine concentrations recovered after 2 weeks despite ongoing undernutrition; urinary taurine excretion had recovered by 30 d after mating. PreĂľ PostC ewes followed the same pattern as PreC for the first 60 d, but plasma taurine concentrations and urinary excretion recovered slowly, and did not reach the control levels until 97 d. These data suggest that different periods of mild periconceptional undernutrition in sheep have different but substantial effects on maternal taurine homoeostasis. These effects may be one mechanism by which maternal periconceptional undernutrition alters development of the offspring with implications for adult health

    Maternal periconceptional undernutrition (PCUN) affected fetal pancreatic maturation in late gestation lambs and impaired glucose tolerance in 10-month-old sheep. To examine the importance of the timing of maternal undernutrition around conception, a further cohort was born to PCUN ewes [undernourished for 61 d before conception (PreC), 30 d after conception (PostC), or 61 d before until 30 d after conception (PrePostC)], or normally fed ewes (Control) (n = 15–20/group). We compared glucose tolerance, insulin secretion, and sensitivity at 36 months of age. We also examined protein expression of insulin signalling proteins in muscle from these animals and in muscle from a fetal cohort (132 d of gestation; n = 7–10/ group). Adult PostC and PrePostC sheep had higher glucose area under the curve than Controls (P = 0.07 and P = 0.02, respectively), whereas PreC sheep were similar to Controls (P = 0.97). PostC and PrePostC had reduced first-phase insulin secretion compared with Control (P = 0.03 and P = 0.02, respectively). PreC was similar to Control (P = 0.12). Skeletal muscle SLC2A4 protein expression in PostC and PrePostC was increased 19%–58% in fetuses (P = 0.004), but decreased 39%–43% in adult sheep (P = 0.003) compared with Controls. Consistent with this, protein kinase C zeta (PKCζ) protein expression tended to be increased in fetal (P = 0.09) and reduced in adult (P = 0.07) offspring of all PCUN ewes compared with Controls. Maternal PCUN alters several aspects of offspring glucose homeostasis into adulthood.     Altered Dairy Protein Intake Does Not Alter Circulatory Branched Chain Amino Acids in Healthy Adults: A Randomized Controlled Trial

    Dairy, as a major component of a high protein diet, is a critical dietary source of branched chain amino acids (BCAA), which are biomarkers of health and diseases. While BCAA are known to be key stimulators of protein synthesis, elevated circulatory BCAA is an independent risk factor for type 2 diabetes mellitus. This study examined the impact of altered dairy intake on plasma BCAA and their potential relationship to insulin sensitivity. Healthy adults (n = 102) were randomized to receive dietary advice to reduce, maintain, or increase habitual dairy intake for 1 month. Food intake was recorded with food frequency questionnaires. Self-reported protein intake from dairy was reported to be reduced (&minus;14.6 &plusmn; 3.0 g/day), maintained (&minus;4.0 &plusmn; 2.0 g/day) or increased (+13.8 &plusmn; 4.1 g/day) according to group allocation. No significant alterations in circulating free amino acids (AA), including BCAA, were measured. Insulin sensitivity, as assessed by homeostatic model assessment-insulin resistance (HOMA-IR), was also unaltered. A significant change in dairy protein intake showed no significant effect on fasting circulatory BCAA and insulin sensitivity in healthy populations
