    MITOCHONDRIAL DNA POLYMORPHISMS AND FERTILITY IN BEEF CATTLE

    Two regions of mitochondrial DNA, D-loop and ND-5 were characterized using polymerase chain reaction – restriction fragment length polymorphism (PCR-RFLP) involving 422 beef cattle of Hereford and composite breeds from Wokalup’s research station. ANOVA models (model I, II) were used to estimate associations between molecular haplotypes and quantitative traits. The phenotypic data used were records on calving rate, defined as the mean number of live calves born over four years, while the genotypic data used were the result of PCR-RFLP analysis in both regions of mitochondrial DNA using 7 restriction enzymes. The results of the present study have provided evidence that mitochondrial polymorphisms in the D-loop and ND-5 regions are associated significantly with fertility. This is the first report of a correlation between mitochondrial polymorphism in D-loop and ND-5 on fertility in beef cattle. Key words: PCR-RFLP, bovine mitochondrial DNA, D-loop, ND-5

All rights reserved.It is often known, from modelling studies, that a certain mode of climate tipping (of the oceanic thermohaline circulation, for example) is governed by an underlying fold bifurcation. For such a case, we present a scheme of analysis that determines the best stochastic fit to the existing data. This provides the evolution rate of the effective control parameter, the variation of the stability coefficient and the path itself and its tipping point. By assessing the actual effective level of noise in the available time series, we are then able to make probability estimates of the time of tipping. This new technique is applied, first, to the output of a computer simulation for the end of greenhouse Earth about 34 million years ago when the climate tipped from a tropical state into an icehouse state with ice caps. Second, we use the algorithms to give probabilistic tipping estimates for the end of the most recent glaciation of the Earth using actual geological ice-core data

This Theme Issue addresses two practical problems that make even prediction of the statistical properties of the climate, when treated as the attractor of a chaotic system (the weather), so challenging. The first is that even for the most detailed models, these statistical properties of the attractor show systematic biases. The second is that the attractor may undergo sudden large-scale changes on a time scale that is fast compared with the gradual change of the forcing (the so-called climate tipping)

Using techniques from bifurcation theory, recent work looks for a slowing down of the intrinsic transient responses, which is predicted to occur before an instability is encountered. This is done, for example, by determining the short-term autocorrelation coefficient ARC(1) in a sliding window of the time-series: this stability coefficient should increase to unity at tipping. Such studies have been made both on climatic computer models and on real paleoclimate data preceding ancient tipping events. The latter employ reconstituted time-series provided by ice cores, sediments, etc., and seek to establish whether the actual tipping could have been accurately predicted in advance. One such example is the end of the Younger Dryas event, about 11 500 years ago, when the Arctic warmed by 7°C in 50 yrs. A second gives an excellent prediction for the end of "greenhouse" Earth about 34 million years ago when the climate tipped from a tropical state into an icehouse state, using data from tropical Pacific sediment cores. This prediction science is very young, but some encouraging results are already being obtained. Future analyses will clearly need to embrace both real data from improved monitoring instruments, and simulation data generated from increasingly sophisticated predictive models

    Loop-mediated isothermal amplification (LAMP) method for rapid detection of Trypanosoma brucei rhodesiense

    Loop-mediated isothermal amplification (LAMP) of DNA is a novel technique that rapidly amplifies target DNA under isothermal conditions. In the present study, a LAMP test was designed from the serum resistance-associated (SRA) gene of Trypanosoma brucei rhodesiense, the cause of the acute form of African sleeping sickness, and used to detect parasite DNA from processed and heat-treated infected blood samples. The SRA gene is specific to T. b. rhodesiense and has been shown to confer resistance to lysis by normal human serum. The assay was performed at 62°C for 1 h, using six primers that recognised eight targets. The template was varying concentrations of trypanosome DNA and supernatant from heat-treated infected blood samples. The resulting amplicons were detected using SYTO-9 fluorescence dye in a real-time thermocycler, visual observation after the addition of SYBR Green I, and gel electrophoresis. DNA amplification was detected within 35 min. The SRA LAMP test had an unequivocal detection limit of one pg of purified DNA (equivalent to 10 trypanosomes/ml) and 0.1 pg (1 trypanosome/ml) using heat-treated buffy coat, while the detection limit for conventional SRA PCR was ∼1,000 trypanosomes/ml. The expected LAMP amplicon was confirmed through restriction enzyme RsaI digestion, identical melt curves, and sequence analysis. The reproducibility of the SRA LAMP assay using water bath and heat-processed template, and the ease in results readout show great potential for the diagnosis of T. b. rhodesiense in endemic regions

    Chloramidine/Bisindolylmaleimide-I-Mediated Inhibition of Exosome and Microvesicle Release and Enhanced Efficacy of Cancer Chemotherapy

    Microvesicle (MV) release from tumour cells influences drug retention, contributing to cancer drug resistance. Strategically regulating MV release may increase drug retention within cancer cells and allow for lower doses of chemotherapeutic drugs. The contribution of exosomes to drug retention still remains unknown. Potential exosome and MV (EMV) biogenesis inhibitors, tested on human prostate cancer (PC3) cells for their capacity to inhibit EMV release, were also tested on PC3 and MCF-7 (breast cancer) cells for improving chemotherapy. Agents inhibiting EMV release most significantly, whilst maintaining cell viability, were chloramidine (Cl-amidine; 50 µM) and bisindolylmaleimide-I (10 µM). Apoptosis mediated by the chemotherapy drug 5-fluorouracil (5-FU) was significantly enhanced in PC3 cells in the presence of both these EMV inhibitors, resulting in a 62% (Cl-amidine + 5-FU) and 59% (bisindolylmaleimide-I + 5-FU) decrease in numbers of viable PC3 cells compared to 5-FU alone after 24 h. For MCF-7 cells, there were similar increased reductions of viable cells compared to 5-FU treatment alone ranging from 67% (Cl-amidine + 5-FU) to 58% (bisindolylmaleimide-I + 5-FU). Using combinatory treatment, the two EMV inhibitors further reduced the number of viable cancer cells tested. Neither inhibitor affected cell viability. Combining selected EMV inhibitors may pose as a novel strategy to enhance the efficacy of chemotherapeutic drug-mediated apoptosis

    Carryover of scanning behaviour affects upright face recognition differently to inverted face recognition.

    Face perception is characterized by a distinct scanpath. While eye movements are considered functional, there has not been direct evidence that disrupting this scanpath affects face recognition performance. The present experiment investigated the influence of an irrelevant letter-search task (with letter strings arranged horizontally, vertically, or randomly) on the subsequent scanning strategies in processing upright and inverted famous faces. Participants’ response time to identify the face and the direction of their eye movements were recorded. The orientation of the letter search influenced saccadic direction when viewing the face images, such that a direct carryover-effect was observed. Following a vertically oriented letter-search task, the recognition of famous faces was slower and less accurate for upright faces, and faster for inverted faces. These results extend the carryover findings of Thompson and Crundall into a novel domain. Crucially they also indicate that upright and inverted faces are better processed by different eye movements, highlighting the importance of scanpaths in face recognition

    The Meat Standards Australia Index indicates beef carcass quality

    A simple index that reflects the potential eating quality of beef carcasses is very important for producer feedback. The Meat Standards Australia (MSA) Index reflects variation in carcass quality due to factors that are influenced by producers (hot carcass weight, rib fat depth, hump height, marbling and ossification scores along with milk fed veal category, direct or saleyard consignment, hormonal growth promotant status and sex). In addition, processor impacts on meat quality are standardised so that the MSA Index could be compared across time, breed and geographical regions. Hence, the MSA Index was calculated using achilles hung carcasses, aged for 5 days postmortem. Muscle pH can be impacted by production, transport, lairage or processing factors, hence the MSA Index assumes a constant pH of 5.6 and loin temperature of 7 o C for all carcasses. To quantify the cut weight distribution of the 39 MSA cuts in the carcass, 40 Angus steers were sourced from the low (n=13), high (n=15) and myostatin (n=12) muscling selection lines. The left side of each carcass was processed down to the 39 trimmed MSA cuts. There was no difference in MSA cut distribution between the low and high muscling lines (P>0.05), although there were differences with nine cuts from the myostatin line (P<0.05). There was no difference in the MSA Index calculated using actual muscle percentages and using the average from the low and high muscling lines (R 2 =0.99). Different cooking methods impacted via a constant offset between eating quality and carcass input traits (R 2 =1). The MSA Index calculated for the four most commercially important cuts was highly related to the index calculated using all 39 MSA cuts (R 2 =0.98), whilst the accuracy was lower for an index calculated using the striploin (R 2 =0.82). Therefore, the MSA Index was calculated as the sum of the 39 eating quality scores predicted at 5 days ageing, based on their most common cooking method, weighted by the proportions of the individual cut relative to total weight of all cuts. The MSA Index provides producers with a tool to assess the impact of management and genetic changes on the predicted eating quality of the carcass. The MSA Index could also be utilised for benchmarking and to track eating quality trends at farm, supply chain, regional, state or national levels
