Evaluating the Applicability of the Fokker-Planck Equation in Polymer Translocation: A Brownian Dynamics Study
Brownian dynamics (BD) simulations are used to study the translocation
dynamics of a coarse-grained polymer through a cylindrical nanopore. We
consider the case of short polymers, with a polymer length, N, in the range
N=21-61. The rate of translocation is controlled by a tunable friction
coefficient, gamma_{0p}, for monomers inside the nanopore. In the case of
unforced translocation, the mean translocation time scales with polymer length
N as ~ (N-N_p)^alpha, where N_p is the average number of monomers in the
nanopore. The exponent approaches the value alpha=2 when the pore friction is
sufficiently high, in accord with the prediction for the case of the
quasi-static regime where pore friction dominates. In the case of forced
translocation, the polymer chain is stretched and compressed on the cis and
trans sides, respectively, for low gamma_{0p}. However, the chain approaches
conformational quasi-equilibrium for sufficiently large gamma_{0p}. In this
limit the observed scaling of with driving force and chain length
supports the FP prediction that is proportional to N/f_d for sufficiently
strong driving force. Monte Carlo simulations are used to calculate
translocation free energy functions for the system. The free energies are used
with the Fokker-Planck equation to calculate translocation time distributions.
At sufficiently high gamma_{0p}, the predicted distributions are in excellent
agreement with those calculated from the BD simulations. Thus, the FP equation
provides a valid description of translocation dynamics for sufficiently high
pore friction for the range of polymer lengths considered here. Increasing N
will require a corresponding increase in pore friction to maintain the validity
of the FP approach. Outside the regime of low N and high pore friction, the
polymer is out of equilibrium, and the FP approach is not valid.Comment: 13 pages, 11 figure
Recent H-alpha results on pulsar B2224+65's bow-shock nebula, the "Guitar"
We used the 4 m Discovery Channel Telescope (DCT) at Lowell observatory in 2014 to observe the Guitar Nebula, an Hα bow-shock nebula around the high-velocity radio pulsar B2224+65. Since the nebula`s discovery in 1992, the structure of the bow-shock has undergone significant dynamical changes. We have observed the limb structure, targeting the "body" and "neck" of the guitar. Comparing the DCT observations to 1995 observations with the Palomar 200-inch Hale telescope, we found changes in both spatial structure and surface brightness in the tip, head, and body of the nebula
Chemotropic guidance facilitates axonal regeneration and synapse formation after spinal cord injury.
A principal objective of spinal cord injury (SCI) research is the restoration of axonal connectivity to denervated targets. We tested the hypothesis that chemotropic mechanisms would guide regenerating spinal cord axons to appropriate brainstem targets. We subjected rats to cervical level 1 (C1) lesions and combinatorial treatments to elicit axonal bridging into and beyond lesion sites. Lentiviral vectors expressing neurotrophin-3 (NT-3) were then injected into an appropriate brainstem target, the nucleus gracilis, and an inappropriate target, the reticular formation. NT-3 expression in the correct target led to reinnervation of the nucleus gracilis in a dose-related fashion, whereas NT-3 expression in the reticular formation led to mistargeting of regenerating axons. Axons regenerating into the nucleus gracilis formed axodendritic synapses containing rounded vesicles, reflective of pre-injury synaptic architecture. Thus, we report for the first time, to the best of our knowledge, the reinnervation of brainstem targets after SCI and an essential role for chemotropic axon guidance in target selection
Angular versus radial correlation effects on momentum distributions of light two-electron ions
We investigate different correlation mechanisms for two-electron systems and
compare their respective effects on various electron distributions. The
simplicity of the wave functions used allows for the derivation of closed-form
analytical expressions for all electron distributions. Among other features, it
is shown that angular and radial correlation mechanisms have opposite effects

Presentation and Outcomes After Medical and Surgical Treatment Versus Medical Treatment Alone of Spontaneous Infectious Spondylodiscitis: A Systematic Literature Review and Meta-Analysis.
Study Design: Systematic literature review.
Objectives: The aims of this study were to (1) describe the clinical features, disabilities, and incidence of neurologic deficits of pyogenic spondylodiscitis prior to treatment and (2) compare the functional outcomes between patients who underwent medical treatment alone or in combination with surgery for pyogenic spondylodiscitis.
Methods: A systematic literature review was performed using PubMed according to PRISMA guidelines. No year restriction was put in place. Statistical analysis of pooled data, when documented in the original report (ie, number of patients with desired variable and number of patients evaluated), was conducted to determine the most common presenting symptoms, incidence of pre- and postoperative neurologic deficits, associated comorbidities, infectious pathogens, approach for surgery when performed, and duration of hospitalization. Outcomes data, including return to work status, resolution of back pain, and functional recovery were also pooled among all studies and surgery-specific studies alone. Meta-analysis of studies with subgroup analysis of pain-free outcome in surgical and medical patients was performed.
Results: Fifty of 1286 studies were included, comprising 4173 patients undergoing either medical treatment alone or in combination with surgery. Back pain was the most common presenting symptom, reported in 91% of patients. Neurologic deficit was noted in 31% of patients.
Conclusion: Medical management remains first-line treatment of infectious pyogenic spondylodiscitis. Surgery may be indicated for progressive pain, persistent infection on imaging, deformity or neurologic deficits. If surgery is required, reported literature shows potential for significant pain reduction, improved neurologic function and a high number of patients returning to a normal functional/work status
Epigenetic Modifications of the PGC-1α Promoter during Exercise Induced Expression in Mice
The transcriptional coactivator, PGC-1α, is known for its role in mitochondrial biogenesis. Although originally thought to exist as a single protein isoform, recent studies have identified additional promoters which produce multiple mRNA transcripts. One of these promoters (promoter B), approximately 13.7kb upstream of the canonical PGC-1α promoter (promoter A), yields alternative transcripts present at levels much lower than the canonical PGC-1α mRNA transcript. In skeletal muscle, exercise resulted in a substantial, rapid increase of mRNA of these alternative PGC-1α transcripts. Although the β2-adrenergic receptor was identified as a signaling pathway that activates transcription from PGC-1α promoter B, it is not yet known what molecular changes occur to facilitate PGC-1α promoter B activation following exercise. We sought to determine whether epigenetic modifications were involved in this exercise response in mouse skeletal muscle. We found that DNA hydroxymethylation correlated to increased basal mRNA levels from PGC-1α promoter A, but that DNA methylation appeared to play no role in the exercise-induced activation of PGC-1α promoter B. The level of the activating histone mark H3K4me3 increased with exercise 2–4 fold across PGC- 1α promoter B, but remained unaltered past the canonical PGC-1α transcriptional start site. Together, these data show that epigenetic modifications partially explain exercise-induced changes in the skeletal muscle mRNA levels of PGC-1α isoforms
A Subaru Weak Lensing Survey I: Cluster Candidates and Spectroscopic Verification
We present the results of an ongoing weak lensing survey conducted with the
Subaru telescope whose initial goal is to locate and study the distribution of
shear-selected structures or halos. Using a Suprime-cam imaging survey spanning
21.82 square degree, we present a catalog of 100 candidate halos located from
lensing convergence maps. Our sample is reliably drawn from that subset of our
survey area, (totaling 16.72 square degree) uncontaminated by bright stars and
edge effects and limited at a convergence signal to noise ratio of 3.69. To
validate the sample detailed spectroscopic measures have been made for 26
candidates using the Subaru multi-object spectrograph, FOCAS. All are confirmed
as clusters of galaxies but two arise as the superposition of multiple clusters
viewed along the line of sight. Including data available in the literature and
an ongoing Keck spectroscopic campaign, a total of 41 halos now have reliable
redshifts. For one of our survey fields, the XMM LSS (Pierre et al. 2004)
field, we compare our lensing-selected halo catalog with its X-ray equivalent.
Of 15 halos detected in the XMM-LSS field, 10 match with published X-ray
selected clusters and a further 2 are newly-detected and spectroscopically
confirmed in this work. Although three halos have not yet been confirmed, the
high success rate within the XMM-LSS field (12/15) confirms that weak lensing
provides a reliable method for constructing cluster catalogs, irrespective of
the nature of the constituent galaxies or the intracluster medium.Comment: To appear in ApJ, High resolution preprint available at

Racial Differences in the Effect of Granulocyte Macrophage Colony-Stimulating Factor on Improved Walking Distance in Peripheral Artery Disease: The PROPEL Randomized Clinical Trial.
Background The effects of race on response to medical therapy in people with peripheral artery disease ( PAD ) are unknown. Methods and Results In the PROPEL (Progenitor Cell Release Plus Exercise to Improve Functional Performance in PAD) Trial, PAD participants were randomized to 1 of 4 groups for 6Â months: supervised treadmill exercise+granulocyte-macrophage colony-stimulating factor ( GM - CSF ) (Group 1), exercise+placebo (Group 2), attention control+ GM - CSF (Group 3), or attention control+placebo (Group 4). Change in 6-minute walk distance was measured at 12- and 26-week follow-up. In these exploratory analyses, groups receiving GM - CSF (Groups 1 and 3), placebo (Groups 2 and 4), exercise (Groups 1 and 2), and attention control (Groups 2 and 4) were combined, maximizing statistical power for studying the effects of race on response to interventions. Of 210 PAD participants, 141 (67%) were black and 64 (30%) were white. Among whites, GM - CSF improved 6-minute walk distance by +22.0Â m (95% CI : -4.5, +48.5, P=0.103) at 12 weeks and +44.4Â m (95% CI : +6.9, +82.0, P=0.020) at 26 weeks, compared with placebo. Among black participants, there was no effect of GM - CSF on 6-minute walk distance at 12-week ( P=0.26) or 26-week (-5.0Â m [-27.5, +17.5, P=0.66]) follow-up, compared with placebo. There was an interaction of race on the effect of GM - CSF on 6-minute walk change at 26-week follow-up ( P=0.018). Exercise improved 6-minute walk distance in black ( P=0.006) and white ( P=0.034) participants without interaction. Conclusions GM - CSF improved 6-minute walk distance in whites with PAD but had no effect in black participants. Further study is needed to confirm racial differences in GM - CSF efficacy in PAD . Prospects for local co-governance
British local authorities and their partners are increasingly developing new ways of working together with local communities. The nature of this co-working, however, is complex, multi-faceted and little understood. This article argues for greater clarity of thinking on the topic, by analysing this co-working as a form of political co-governance, and drawing attention in particular to issues of scale and democracy. Using evidence from a study of 43 local authority areas, 16 authorities are identified where co-governance is practised, following three main types of approach: service-influencing, service-delivering and parish council developing. It is concluded that strengthening political co-governance is essential for a healthy democracy
Halo Substructure and the Power Spectrum
In this proceeding, we present the results of a semi-analytic study of CDM
substructure as a function of the primordial power spectrum. We apply our
method to several tilted models in the LCDM framework with n=0.85-1.1,
sigma_8=0.65-1.2 when COBE normalized. We also study a more extreme, warm dark
matter-like spectrum that is sharply truncated below a scale of 10^10 h^-1
Msun. We show that the mass fraction of halo substructure is not a strong
function of spectral slope, so it likely will be difficult to constrain tilt
using flux ratios of gravitationally lensed quasars. On the positive side, all
of our CDM-type models yield projected mass fractions in good agreement with
strong lensing estimates: f \sim 1.5% at M \sim 10^8 Msun. The truncated model
produces a significantly smaller fraction, f \lsim 0.3%, suggesting that warm
dark matter-like spectra may be distinguished from CDM spectra using lensing.
We also discuss the issue of dwarf satellite abundances, with emphasis on the
cosmological dependence of the map between the observed central velocity
dispersion of Milky Way satellites and the maximum circular velocities of their
host halos. In agreement with earlier work, we find that standard LCDM
over-predicts the estimated count of Milky Way satellites at fixed Vmax by an
order of magnitude, but tilted models do better because subhalos are less
concentrated. Interestingly, under the assumption that dwarfs have isotropic
velocity dispersion tensors, models with significantly tilted spectra (n \lsim
0.85, sigma_8 \lsim 0.7) may under-predict the number of large Milky Way
