    Biologization of potato cultivation technology

    In biologized agricultural production, along with minimized fertilizers, growth activators are widely used. The article analyzes experimental data on potato cultivation using such preparations. The purpose of this study was to study the effect of copper nanoparticles and liquid-phase biopreparation on the phenology and yield of potatoes in the conditions of lysimetric experiment. The research results showed that pre-sowing treatment of seed tubers with growth activators had a positive effect on linear indicators, heat resistance of plants, increasing it by 16.7-50.0% compared to the control, as well as yield (the increase was 8.6-54.8 t/ha). It is shown that the best effect is obtained by the combined use of copper nanoparticles and 1% liquid-phase biological product

    At present, the fact that all unforeseen negative consequences of anthropogenic human activity significantly affect the biochemical regime of the natural environment is obvious. Therefore, agriculture should be aimed at reducing the release of chemicals from the biological cycle. Field observations were conducted to study the effect of detoxification techniques on the accumulation of heavy metals in soil. Six experiment variants were developed, using detoxicants with various combinations of them. The results of this research indicate that the organic matter of manure binds soluble metal salts into organometallic complexes, and fertilizer phosphates convert them into sparingly soluble compounds. Thus, they improve (heal) the soil contaminated by heavy metals

    The article concentrates on immigration and emigration impact on EU Member State economy and analyzes the migration situation in Poland. If the purpose of immigration is to obtain a residency and then integration into society, thus the increase in population will be adopted by the national economy in a short period. There also won’t be problems with circular migration, since the workforce rotation will be within the agreed limits. However, temporary migration will lead either to unacceptably low living conditions of temporarily employed foreigners, to an increase in social tension, or to an inefficient use of social infrastructureEl artículo se centra en el impacto de la inmigración y la emigración en la economía de los Estados miembros de la UE y analiza la situación de la migración en Polonia. Si el propósito de la inmigración es obtener una residencia y luego la integración en la sociedad, el aumento de la población será adoptado por la economía nacional en un corto período.     Social Ideal and its Implementation in the Philosophy of S. L. Frank

    The paper examines the social philosophy of a prominent Russian thinker, Semen Frank with the aim of identifying its relations with general philosophical ideas, as well as with religious foundations of his thought. For these purposes, authors examine the basic concepts of Frank’s social philosophy in their interaction and connection. Based on the research, authors conclude that Frank built his social theory on the basis of the philosophy all-unity that gives primacy to the spiritual aspects of the social body over the material and empirical ones. The basic social concepts he develops are related with the two fundamental modes of the reality – ideal and empirical.El artículo examina la filosofía social de un destacado pensador ruso, Semen Frank, con el objetivo de identificar sus relaciones con ideas filosóficas generales, así como con los fundamentos religiosos de su pensamiento. Para estos fines, los autores examinan los conceptos básicos de la filosofía social de Frank en su interacción y conexión. Con base en la investigación, los autores concluyen que Frank construyó su teoría social sobre la base de la filosofía de la unidad total que da primacía a los aspectos espirituales del cuerpo social sobre bases materiales y empíricas. Los conceptos sociales básicos que desarrolla están relacionados con los dos modos fundamentales de la realidad: ideal y empírico

    Clinical aspects of TP53 gene inactivation in diffuse large B-cell lymphoma

    Abstract Background The knowledge about specific mechanisms generating TP53 dysfunction in diffuse large B-cell lymphoma is limited. The aim of the current study was to comprehensively explore TP53 gene variability resulting from somatic mutations, promoter methylation, and allelic imbalance in tumorous tissue of diffuse large B-cell lymphoma (DLBCL). Methods DNA samples from 74 patients with DLBCL were used. Genomic DNA was isolated from paraffin blocks of lymph nodes or from extranodal biopsies of tumors by the phenol–chloroform extraction method with guanidine. Analysis of coding sequences of the TP53 gene was based on Sanger’s direct sequencing method. The methylation status of the TP53 promoter was analyzed using by methylation-specific PCR on bisulfite-converted DNA. Assessment of the detected mutations was carried out in the IARC TP53 Database and the TP53 UMD mutation database of human cancer. Results The mutations in regions coding for the DNA-binding domain were prevalent (95%). In the analyzed sample of patients, codons 275, 155, 272, and 212 were hotspots of mutations in the TP53 gene. In addition, functionally significant intron mutations (IVS6-36G > C and IVS5 + 43G > T) were detected. Instances of TP53 promoter methylation were observed only in a few samples of diffuse large B-cell lymphoma tissue. Furthermore, loss of heterozygosity was revealed only in the subgroup of patients with altered status of the gene (mutations were detected in five patients and promoter methylation in one case). Conclusions Thus, the results suggest that there are two sequential events in the formation of diffuse large B-cell lymphoma in at least some cases. The first event is mutation or methylation of the TP53 promoter, leading to appearance of a cell with increased risk of malignant transformation. The second event is the loss of an intact allele of the gene; this change is necessary for tumorigenesis. We identified TP53 mutation patterns in a Russian cohort of patients with de novo DLBCL who were treated with R-CHOP and R-CHOP-like regimens and confirmed that TP53 mutation status is a valuable prognostic biomarker

    Uridine phosphorylase catalyzes the phosphorolysis of ribonucleosides, with the nitrogenous base and ribose 1-phosphate as products. Additionally, it catalyzes the reverse reaction of the synthesis of ribonucleosides from ribose 1-phosphate and a nitrogenous base. However, the enzyme does not catalyze the synthesis of nucleosides when the substrate is a nitrogenous base substituted at the 6-­position, such as 6-methyluracil (6-MU). In order to explain this fact, it is essential to investigate the three-dimensional structure of the complex of 6-MU with uridine phosphorylase. 6-MU is a pharmaceutical agent that improves tissue nutrition and enhances cell regeneration by normalization of nucleotide exchange in humans. 6-MU is used for the treatment of diseases of the gastrointestinal tract, including infectious diseases. Here, procedures to obtain the uridine phosphorylase from the pathogenic bacterium Vibrio cholerae (VchUPh), purification of this enzyme, crystallization of the complex of VchUPh with 6-MU, and X-ray data collection and preliminary X-ray analysis of the VchUPh-6-MU complex at atomic resolution are reported

    A high-resolution structure of the complex of Vibrio cholerae uridine phosphorylase (VchUPh) with its physiological ligand thymidine is important in order to determine the mechanism of the substrate specificity of the enzyme and for the rational design of pharmacological modulators. Here, the expression and purification of VchUPh and the crystallization of its complex with thymidine are reported. Conditions for crystallization were determined with an automated Cartesian Dispensing System using The Classics, MbClass and MbClass II Suites crystallization kits. Crystals of the VchUPh–thymidine complex (of dimensions ∼200–350 µm) were grown by the sitting-drop vapour-diffusion method in ∼7 d at 291 K. The crystallization solution consisted of 1.5 µl VchUPh (15 mg ml(−1)), 1 µl 0.1 M thymidine and 1.5 µl reservoir solution [15%(w/v) PEG 4000, 0.2 M MgCl(2).6H(2)O in 0.1 M Tris–HCl pH 8.5]. The crystals diffracted to 2.12 Å resolution and belonged to space group P2(1) (No. 4), with unit-cell parameters a = 91.80, b = 95.91, c = 91.89 Å, β = 119.96°. The Matthews coefficient was calculated as 2.18 Å(3) Da(−1); the corresponding solvent content was 43.74%
