Phosphatidyl inositol-phospholipase C in squid photoreceptor membrane is activated by stable metarhodopsin via GTP-binding protein, Gq
AbstractPhosphatidyl inositol-phospholipase C (PI-PLC) in squid retina was studied by immunoblotting and its activites were determined using [3H]phosphatidyl inositol bisphosphate ([3H]PIP2) as substrate. PI-PLC activity was found mostly in soluble fraction when the retina homogenate was treated with 400 mM KCl, but was associated with rhabdomal membranes under low salt conditions (20 mM Hepes). A protein with apparent molecular mass of 130 kD was recognized by an antibody against PLCβ4/norp A in both 400 mM KCl soluble and rhabdomal membrane fractions. A 42 kD protein recognized by antibody against the C-terminus of Gqα was also present in these two fractions. GTPγS stimulated only the PI-PLC activity associated with membrane and was magnesium dependent. PI-PLC activity was found to be (i) highly dependent upon calcium concentrations, (ii) enhanced by GTP but not by other nucleotides, and (iii) significantly stimulated by light at lower concentrations of GTPγS. The stimulation by light was still observed when irradiated membrane was incubated at 10°C for 10 min and then mixed with GTPγS. Teaching Strategy for the Interactive Lesson Using the Internet - As an Example of a German Class for Intermediate Students
For six years I have taught Intermediate German at several universities using homepages from the Internet as teaching materials. During these six years the Internet spread quickly. The quantity of information sources available on the Internet is increasing day by day, and their quality is improving. I can say with confidence that the average level of computer literacy among present-day students shows a remarkable advance on the past. Now, there is also none of the mental distance between studens and the Internet that existed even just a few years ago. In such a situation the Internet can be utilized in drawing learners up to the next level targeted for them. Here I introduce two examples implemented with the presetn writer\u27s students in 2001-2002: teh posting of students results on the Internet and a project in which each students established contact with the webmaster of a German language homepage. Both are based in the interactive aspect of the Internet. In order to teach students who can manipulate a computer comparatively freely, an appropriate new strategy must be devised. In this report I want to offer one suggestion for such a strategy
Enhanced anti-HIV-1 activity of CC-chemokine LD78β, a non-allelic variant of MIP-1α/LD78α
AbstractWe compared the anti-HIV-1 activity of CC-chemokine LD78β with that of MIP-1α, another CC-chemokine which shows 94% sequence homology with LD78β. Despite its close similarity to MIP-1α, the anti-HIV-1 activity of LD78β appeared to be nearly 10 times higher than that of MIP-1α. Mutagenesis of MIP-1α showed that the N-terminal additional tetrapeptide, which was present in LD78β and absent in MIP-1α, is responsible for enhanced anti-HIV-1 activity. The N-terminal structure-function relationship of LD78β described here will be of value in understanding the chemokine-receptor interactions and designing anti-HIV-1 compounds based on LD78β
