Urban regeneration and tenure mix: Exploring the dynamics of neighbour interactions
This paper draws on in-depth research on the nature and intensity of neighbour relations in an area in Peckham, London, which underwent urban regeneration aimed at improving the urban environment and increasing the area’s tenure mix. Drawing on the literature on neighbouring, the paper explores residents’ perceptions and attitudes towards their neighbours and the dynamics of their routine interactions. Despite findings pointing towards a general atmosphere of cordiality and solidarity, interactions were casual, coexisted with prejudiced views towards certain groups and areas, and were viewed by residents as part of their everyday social practices of community. As a result, very little else was exchanged between tenures, putting into question some policy assumptions that the increased physical proximity between housing tenures can potentially lead to instrumental interaction that can benefit low-income households in social housing. Reflecting on these findings, the paper discusses some implications that have relevance for policy and research
Influência de polimorfismos em genes de citocinas e de receptores de citocinas na resposta ao tratamento e no grau do dano hepático em pacientes portadores de hepatite C crônica
Resumo: O vírus da hepatite C (HCV) é um patógeno capaz de causar infecção crônica em cerca de dois terços dos indivíduos infectados, conseqüência de suas habilidades em driblar tanto a imunidade inata quanto a adquirida. A fibrose é resultado de injúrias repetitivas causadas aos hepatócitos decorrente da infecção pelo HCV e da ação da resposta imunológica, que levam a uma falha no processo regenerativo e deposição de uma quantidade abundante de matriz extracelular. Citocinas regulam a resposta inflamatória à injúria interferindo também na fibrogênese. O protocolo padrão de tratamento para hepatite C crônica é baseado na combinação de interferon-alfa convencional ou peguilado e ribavirina, administrados por 24 ou 48 semanas, respectivamente. Polimorfismos de um único nucleotídeo (SNPs) localizados em regiões reguladoras/codificadoras de genes de citocinas poderiam contribuir para a fibrogênese hepática, bem como influenciar a resposta ao tratamento, pois interferem na expressão e secreção de citocinas, importantes proteínas que participam da resposta imunológica contra o HCV. Portanto, o objetivo do presente trabalho foi tipar 22 SNPs localizados em 13 genes de citocinas/receptores de citocinas visando avaliar a influência das variantes polimórficas no grau de dano hepático e na resposta ao tratamento em pacientes brasileiros cronicamente infectados apenas pelo genótipo 1 do HCV. Variantes polimórficas para as posições TNFA-308, IL6-174, IL6nt565 e IL4RA+1902 estão associadas ao grau de dano hepático. Variantes polimórficas para as posições IL10-819, IL10-592, IL1A-889, IL1B+3962, IL1R1 pst1 1970 e IL4RA+1902 estão associadas à resposta ao tratamento. Concluímos que polimorfismos em genes de citocinas/receptores de citocinas parecem estar influenciando o dano hepático, bem como a resposta ao tratamento nos pacientes estudados

Femme 7.83 - words give me Wings, comes from my look on the contemporary, coming from Poetry. This work deconstructs the pain of thorns that cross the body without an organ that feels so much, feels sorry for a search for erasure of what is imposed on us, to exist and become a woman, the marks of a body, in its most sensitive poetics is a political body, which constructs a new discourse every erasing and renewing the marks, to discover itself in resonance with the Earth, to blossom in its identity. This paper shows my career as a student in the Arts course, the process of preparation of the work, the exhibition in 2018, the relationship with philosophy, literature and the phallus of Feminism in Performance. The good meetings, and the result of these meetings, of a body that asks for work can be said that it was necessary to be the work, starting from a poetry, in its potency, the work is to affect, and to whom to leave affect, in this sensitive world, in our time, now. It was not the purpose of the work to make a psychoanalytical analysis, but rather to search for the paths of Art, with a theoretical framework guided by Deleuze, which expands horizons with the “Anti-Oedipus”, “Elektra”, through which I could have more perception of the body, the affections. I also draw the work of Simone de Beauvoir, “Second sex”, which is existential, Hegelian, deals with a transcendent body. The process of the work produced here is not therapeutic and its result is to reproduce reflections and discussions around the work and in what it affects us.

Este trabalho desconstrói a dor de espinhos que atravessam o corpo “sem órgãos”, que sente tanto, sente muito, por uma busca de apagamento do que nos é imposto, do existir e tornar-se mulher, as marcas de um corpo, em sua poética mais sensível, é corpo político, que constrói um novo discurso, a cada apagar, e renovando as marcas, se descobrindo em ressonância com a Terra, desabrochando em sua identidade. Neste sentido, este Trabalho de Conclusão de Curso, mostra meu percurso como aluna no curso de Artes, o processo de preparação da obra, a exposição em 2018, relação com a filosofia, a literatura, e o Feminismo em Performance. Os bons encontros, e do resultado desses encontros, de um corpo que pede obra, pode-se dizer que houve a necessidade dele ser a obra, partindo de um Devir poesia, em sua potência, a obra é para afetar, e a quem se deixar afetar, neste mundo sensível, nesse nosso tempo agora. Não foi o objetivo de o trabalho fazer uma análise psicanalítica, mas sim trazer pesquisas pelos caminhos da Arte, com um referencial teórico pautado por Deleuze, que expande os horizontes, com o “Anti-Édipo”, “Elektra”, por meio do qual pude ter mais percepção do corpo imanente, e dos afetos. Lanço mão também da obra de Simone de Beauvoir, “Segundo sexo”, que é existencial, hegeliana (Ernst Hegel, filósofo do século XIX), trata-se de um corpo transcendente, de um devir outro. O processo da obra aqui produzida, não é terapêutico, seu resultado é reproduzir reflexões, e discussões em torno da obra e no que ela nos afeta.

Bioinformatic analysis of promoters from Ashbya gossypii
Ashbya gossypii is a filamentous fungus that produces large quantities of riboflavin, a living factory that generates nearly half of this vitamins world supply. The fungus can grow on industrial wastes and can produce other value-added compounds including recombinant proteins, single cell oil, nucleosides, folic acid and organoleptic compounds. At the genome level, 95% of the genes have homologues in Saccharomyces cerevisiae (90% in syntenic positions). Many molecular tools used
in yeast are functional in both organisms, including the autonomously replicating sequences.
Fine-tuning heterologous metabolic pathways requires the availability of promoters with a wide
range of activity. Endogenous promoters provide the main regulatory elements for gene expression control, however, there is a limited range of well characterised promoters available for metabolic engineering of A. gossypii. Repeated copies of promoters for genome editing purposes increases the probability of homologous recombination and causes strain instability, what is disadvantageous when extensive pathway engineering is performed.
The identification of transcription factor (TF) binding sites (TFBSs) in promoters is important for rational design of regulatory crosstalk in metabolic engineering strategies. TFs usually bind to degenerated sequences that can be identified using bioinformatics tools. The MEME algorithm can be used to mine DNA motifs from upstream sequences of co-expressed or co-regulated genes. The obtained motifs can further be compared to TFBSs in databases for S. cerevisiae (i.e. JASPAR, TRANSFAC, YEASTRACT). This procedure allowed the identification of 4 DNA motifs, 8
matching putative TFs, and TATA-box as important elements for high level gene expression.
Thus, the bioinformatic approach used in this study, in intergenic regions from A. gossypii, can be also performed in sequences of other organisms with the aim of identifying potential candidate motifs for subsequent experimental characterization and allowing future construction of hybrid semi-synthetic promoters.info:eu-repo/semantics/publishedVersio
New genome-editing tools for Ashbya gossypii
Ashbya gossypii is a splendid riboflavin biofactory that suppresses nearly half of the worldwide
demand for vitamin B2. The availability of its genome sequence allowed a better understanding
of this fungus metabolism and the development of targeted genome editing tools. Such
progresses allowed increasing the vitamin B2 natural production levels, and to expand the
potential of this fungus towards the production of nucleosides, folic acid, single cell oil and
fragrances [1,2].
Now, with a broadening range of potential genetic targets, modular and flexible pathway
assembly tools are more convenient than ever. There is also an urgent need for a larger set of
well characterized promoters for metabolic flux optimization towards improved production of
target products. As such, this work aimed at developing a modular, flexible and fast pathway
assembly platform for targeted chromosomal integration in A. gossypii that offers a wide range
of expression strengths (promoters).
The developed pathway assembly platform is in an early-stage, designed to easily allow fast
cloning and exchange of selection markers, promoters, genes and terminators. As endogenous
promoters provide the required regulatory elements for gene expression, several
transcriptomic datasets were scanned for medium to high strength promoters. A set of uni- and
bi-directional promoters was selected, and each promoter cloned into promoter-probes for
characterization in terms of expression strength and regulation, a task that is under way using
multiple reporter systems. Preliminary results revealed that the bi-directional promoter
AgCCW12/HOG1p drives moderate gene expression through the HOG1p side and strong
expression through the CCW12p side. The datasets were also used for transcription factor
binding motif mining, in order to find important regulation elements that confer strong gene
expression values. Five transcription factor binding motifs were identified as strong targets for
promoter engineering. This information can hereafter enable the rational design of
synthetic/semi-synthetic minimal promoters.
With the current genetic engineering turnaround time to genetically modify A. gossypii being a
limiting factor to establish this fungus as a state-of-the-art microbial factory, this pathway assembly platform has great relevance. With a growing number of promoters characterized, it
