Cytochrome oxidase subunit VI of Trypanosoma brucei is imported without a cleaved presequence and is developmentally regulated at both RNA and protein levels
Mitochondrial respiration in the African trypanosome undergoes dramatic developmental stage regulation. This requires co-ordinated control of components encoded by both the nuclear genome and the kinetoplast, the unusual mitochondrial genome of these parasites. As a model for understanding the co-ordination of these genomes, we have examined the regulation and mitochondrial import of a nuclear-encoded component of the cytochrome oxidase complex, cytochrome oxidase subunit VI (COXVI). By generating transgenic trypanosomes expressing intact or mutant forms of this protein, we demonstrate that COXVI is not imported using a conventional cleaved presequence and show that sequences at the N-terminus of the protein are necessary for correct mitochondrial sorting. Analyses of endogenous and transgenic COXVI mRNA and protein expression in parasites undergoing developmental stage differentiation demonstrates a temporal order of control involving regulation in the abundance of, first, mRNA and then protein. This represents the first dissection of the regulation and import of a nuclear-encoded protein into the cytochrome oxidase complex in these organisms, which were among the earliest eukaryotes to possess a mitochondrion
Geniculo-Cortical Projection Diversity Revealed within the Mouse Visual Thalamus

Recent investigations are beginning to reveal regional complexity in mouse dLGN. Using local injections of retrograde tracers into V1 of adult and neonatal mice, we examined the developing organisation of geniculate projection columns: the population of dLGN-V1 projection neurons that converge in cortex. Serial sectioning of the dLGN enabled the distribution of labelled projection neurons to be reconstructed and collated within a common standardised space. This enabled us to determine: the organisation of cells within the dLGN-V1 projection columns; their internal organisation (topology); and their order relative to V1 (topography). Here, we report parameters of projection columns that are highly variable in young animals and refined in the adult, exhibiting profiles consistent with shell and core zones of the dLGN. Additionally, such profiles are disrupted in adult animals with reduced correlated spontaneous activity during development. Assessing the variability between groups with partial least squares regression suggests that 4?6 cryptic lamina may exist along the length of the projection column. Our findings further spotlight the diversity of the mouse dLGN?an increasingly important model system for understanding the pre-cortical organisation and processing of visual information. Influences of human activities on seabird populations in the North Sea
This paper describes the North Sea's seabird resource, which is of great international importance. It reviews the significance of human activities on seabirds; these are persecution, fishing, eutrophication, chemical pollution, introduction of alien predators, rising sea level, and oil pollution. The paper speculates on future trends. Some suggestions to reduce human pressures, to use seabirds as monitors of the marine environment and to conserve their populations more effectively are made
A quantitative method for evaluating the importance of marine areas for conservation of birds
Objective criteria are needed for ranking marine sites when examining candidate areas for protection measures. We suggest a Marine Classification Criterion (MCC) which allows the application of the widely used Ramsar 1% criterion for wetlands for seabirds with clustered distribution in offshore habitats. The maximum size of an area considered to be internationally important has not been defined by the Ramsar Convention. Terrestrial and coastal sites generally have obvious hydrological or physical boundaries, whereas such boundaries are less obvious at sea. The smallest unit which would pass the demands set by the MCC is 1% of the bio-geographic population of a particular species concentrated in an area (site) supporting a density exceeding a value equivalent of four times the average density of the species in the investigated regional sea. The effect of choosing smaller or larger reference densities is tested. The results indicate that the chosen threshold density is a suitable requirement for the inclusion of the most important areas for seabird species with at least 25% of their bio-geographic population occurring in the studied regions of the North Sea and the Baltic Sea. The test cases indicate that provided the MCC is based on geo-statistical analyses of un-biased survey data the boundaries of areas holding large concentrations of seabirds can be estimated with confidence. The MCC could be used to identify concentrations of seabirds and other marine animals of conservation priority and to rank marine areas by their cumulative importance to different species

Ecosystem objectives to support the UK vision for the marine environment
European Maritime States already have commitments to protect species and habitats and maintain quality standards in coastal and offshore waters. These are a direct response to environmental legislation in Europe and commitments made to biodiversity conservation in OSPAR and at the World Summit on Sustainable Development. An integrated approach to management requires that these are consistent with the requirements for sustainable development, and include wider social considerations and active stakeholder participation. This review describes a hierarchical framework that incorporates the marine objectives and delivery statements of ecological, social and economic sectors. The framework leads from the UK's guiding principles for sustainable development, through visionary statements and strategic goals for high level delivery, to operational objectives and statements of action which deliver management. Parts of this hierarchy can already be populated for the UK, especially those at the higher levels. At the operational level, however, there is less clarity. The review shows that, despite some gaps, existing commitments for ecological components of the ecosystem are transparent and generally conform to this framework, due largely to high profile government funding of environmental protection and science and a single national vision for the marine environment. Specific objectives for six components of the ecosystem were developed; benthic habitats, seabirds and mammals, phytoplankton and zooplankton, fish, and physical/chemical quality of the water and atmosphere. The objectives included some that avoided limits and others that aimed to achieve targets, and for management to be effective it will be important to have a common understanding of how these can together be interpreted and made operational. In a review of 13 social and economic sectors, few provided a clear breakdown of objectives leading from a high level vision or a sustainable development principle. Six sectors did not have high level government targets for production or other measures of sectoral performance. Such commercial operations generally determine their own operational delivery targets based on market forces. Recent proposals in Europe to create an integrated framework for social, economic and environmental activities will need to carefully balance the development of a strong and competitive marine economy with existing international obligations to environmental protection
