    Reconstruction of Winter Temperature since the 1830s in Kawanishi Based on Historical Weather Documents

    In the present study, we estimated the winter temperature variations from 1830/31 to 2008/09 in the town of Kawanishi in the southern part of Yamagata prefecture using daily weather reports and observa tions documented in an old personal diary. Using historical weather reports, we computed the snowfall rate (percentage ratio of snowfall days to the total number of precipitation days) for each winter during the study period. Then, mean winter temperatures in Kawanishi were estimated using a simple linear regression analysis based on the relationship between the mean winter temperature and snowfall rate, which was computed from historical weather reports. The results of this analysis showed that there were several warm winters in the late 19th century. The warm winters were observed in a period extending from the late 1840s to the early 1850s, and a period in the late 1860s. The results also showed that the temperatures in those warm winters were similar to the average winter temperature for the period 1970/71-1999/2000. By comparing the time series of estimated temperatures to those of long-term historical instrumental temperature data since the late 19th century, we found that the variation in the estimated temperature correlated well with that of the instrumental temperature data. This indicates that the estimated results in the present study are highly reliable

    Reactions of oxalyl chloride with 1,2-cycloalkanediols in the presence of triethylamine

    The relationship between the product patterns and the configurations of 1,2-cycloheptane- and 1,2-cyclooctanediols 9 in the cyclocondensations with oxalyl chloride in the presence of tricthylamine at 0°C has been shown analogous to that obtained for 1,2-disubstituted acyclic ethylene glycols 1: cis-1,2-cyclooctanediol (9f) produced the cyclic oxalate 14f as the major product, while trans-1,2-cycloheptanediol (9e) and trans-1,2-cyclooctanediol (9g) formed the cyclic carbonates 12e, g as the major products. On the other hand, the cyclic oxalates 14a-d were formed as the major products from 1,2-cyclopentane- and 1,2-cyclohexanediols regardless of the configuration. These results can be accounted for by assuming the boat-like transition states for cyclizations of the half esters of comparatively rigid five- and six-membered diols 9a-d. The cyclic oxalates 14a, c may be directly formed through the resulting tetrahedral intermediates from cis-diols (9a, c), and the cyclic carbonates 12a, c as the minor products after ring inversion of the tetrahedral intermediates. The tetrahedral intermediates from the trans-isomers 9b,d cannot undergo ring inversion, producing no traces of the cyclic carbonates 12b,d. © 2002 Pharmaceutical Society of Japan

    Efficient synthesis and hydrolysis of cyclic oxalate esters of glycols

    Based on the mechanism postulated for the formation of the cyclic carbonates 3 in the reactions of glycols 1 with oxalyl chloride in the presence of triethylamine, we present here three efficient syntheses of the cyclic oxalates 2 of various glycols 1 by controlling the formation of 3: replacement of the base by pyridine markedly diminishes yields of 3 in all reactions, realizing dramatic reversals of the product ratios in the reactions with the (R*,R*)-compounds 1g-i, q, r and pinacol (1k); although considerable amounts of the oxalate polymers are formed in the reactions with some (R*,S*)-glycols, this drawback can be removed by the use of 2,4,6-collidine instead of pyridine; 1,1′-oxalyldiimidazole is useful for the synthesis of two selected cyclic oxalates 2e, f. The cyclic oxalates 2 other than trisubstituted and tetrasubstituted ones were found to be very reactive: kinetic studies on the hydrolysis of 1,4-dioxane-2,3-dione (2a) as well as its mono- and some selected 5,6-disubstituted derivatives 2 have revealed that they undergo hydrolysis 260-1500 times more rapidly than diethyl oxalate (12) in acetate buffer-acetonitrile (pH 5.69) at 25°C. Although the cyclic oxalate 2l from cis-1,2-cyclopentanediol (1l) was 1.5 times more reactive than 2a, it has been shown with other substrates that increasing number of the alkyl substituents decreases the rate of hydrolysis. On the contrary, the phenyl group was found to have somewhat accelerative effect. © 2002 Pharmaceutical Society of Japan

    Reconstruction of July Temperature Variations since the 1830s in Kawanishi Based on Historical Weather Documents

    In the present study, we estimated the July maximum temperature variations from 1830 to 2011 in the town of Kawanishi in the southern part of Yamagata prefecture using daily weather reports and observa tions documented in an old personal diary. Using historical weather reports, we computed the number of "fine-weather" days for each July during the study period. Then, July maximum temperatures in Kawanishi were estimated using a simple linear regression analysis based on the relationship between the July maximum temperature and number of "fine-weather" days, computed from historical weather reports. By com paring the time series of estimated temperatures with those of historical instrumental temperature data in the late 19th century, we found that the variation in the estimated temperature correlated well with that of the instrumental temperature data. This indicates that the estimated results in the present study are highly reliable. The results of this analysis showed that there were cool periods in the 1830s, 1860s, 1900s, and a period extending from the 1980s to the early 1990s. Those cool periods in the 1830s, 1860s, and 1900s coincided with poor rice harvests and severe famines in northern Japan. Warm summers were observed in the 1850s, a period extending from the 1870s to 1880s, and 1920s. We found that temperatures in the early 1850s were similar to those of warm summers in the late 20th century

    Sphingosine 1-Phosphate (S1P) in the Peritoneal Fluid Skews M2 Macrophage and Contributes to the Development of Endometriosis

    Sphingosine 1-phosphate (S1P), an inflammatory mediator, is abundantly contained in red blood cells and platelets. We hypothesized that the S1P concentration in the peritoneal cavity would increase especially during the menstrual phase due to the reflux of menstrual blood, and investigated the S1P concentration in the human peritoneal fluid (PF) from 14 non-endometriosis and 19 endometriosis patients. Although the relatively small number of samples requires caution in interpreting the results, S1P concentration in the PF during the menstrual phase was predominantly increased compared to the non-menstrual phase, regardless of the presence or absence of endometriosis. During the non-menstrual phase, patients with endometriosis showed a significant increase in S1P concentration compared to controls. In vitro experiments using human intra-peritoneal macrophages (MΦ) showed that S1P stimulation biased them toward an M2MΦ-dominant condition and increased the expression of IL-6 and COX-2. An in vivo study showed that administration of S1P increased the size of the endometriotic-like lesion in a mouse model of endometriosis

In particular, studies of gene expression dynamics and regulation landscape in COVID-19 infected individuals are limited. Here, we report on a thorough analysis of whole blood RNA-seq data from 465 genotyped samples from the Japan COVID-19 Task Force, including 359 severe and 106 non-severe COVID-19 cases. We discover 1169 putative causal expression quantitative trait loci (eQTLs) including 34 possible colocalizations with biobank fine-mapping results of hematopoietic traits in a Japanese population, 1549 putative causal splice QTLs (sQTLs; e.g. two independent sQTLs at TOR1AIP1), as well as biologically interpretable trans-eQTL examples (e.g., REST and STING1), all fine-mapped at single variant resolution. We perform differential gene expression analysis to elucidate 198 genes with increased expression in severe COVID-19 cases and enriched for innate immune-related functions. Finally, we evaluate the limited but non-zero effect of COVID-19 phenotype on eQTL discovery, and highlight the presence of COVID-19 severity-interaction eQTLs (ieQTLs; e.g., CLEC4C and MYBL2). Our study provides a comprehensive catalog of whole blood regulatory variants in Japanese, as well as a reference for transcriptional landscapes in response to COVID-19 infection

    DOCK2 is involved in the host genetics and biology of severe COVID-19

Here we conducted a genome-wide association study (GWAS) involving 2, 393 cases of COVID-19 in a cohort of Japanese individuals collected during the initial waves of the pandemic, with 3, 289 unaffected controls. We identified a variant on chromosome 5 at 5q35 (rs60200309-A), close to the dedicator of cytokinesis 2 gene (DOCK2), which was associated with severe COVID-19 in patients less than 65 years of age. This risk allele was prevalent in East Asian individuals but rare in Europeans, highlighting the value of genome-wide association studies in non-European populations. RNA-sequencing analysis of 473 bulk peripheral blood samples identified decreased expression of DOCK2 associated with the risk allele in these younger patients. DOCK2 expression was suppressed in patients with severe cases of COVID-19. Single-cell RNA-sequencing analysis (n = 61 individuals) identified cell-type-specific downregulation of DOCK2 and a COVID-19-specific decreasing effect of the risk allele on DOCK2 expression in non-classical monocytes. Immunohistochemistry of lung specimens from patients with severe COVID-19 pneumonia showed suppressed DOCK2 expression. Moreover, inhibition of DOCK2 function with CPYPP increased the severity of pneumonia in a Syrian hamster model of SARS-CoV-2 infection, characterized by weight loss, lung oedema, enhanced viral loads, impaired macrophage recruitment and dysregulated type I interferon responses. We conclude that DOCK2 has an important role in the host immune response to SARS-CoV-2 infection and the development of severe COVID-19, and could be further explored as a potential biomarker and/or therapeutic target

    ALK-Positive Squamous Cell Carcinoma Dramatically Responded to Alectinib

    Anaplastic lymphoma kinase (ALK) rearrangement is usually observed in patients with adenocarcinoma. Herein, we report a case of squamous cell carcinoma (SCC) with ALK rearrangement treated with alectinib. The patient was a 73-year-old woman without a smoking history. She consulted us with nonproductive cough and loss of appetite. Computed tomography scan revealed a mass in the left lower lobe of the lung. According to the pathological examinations, we diagnosed the tumor as SCC. Because the patient had never smoked, we searched for driver mutations and found that the tumor harbored ALK rearrangement. We began treatment with alectinib, and the tumor remarkably reduced in volume. No severe adverse events were observed. Although there are only few reports of SCC with ALK rearrangement, this case implies that clinicians should consider searching for driver mutations in patients with SCC when there are atypical findings or characteristics
