    Climate Change and Energy Security - Obama’s Historic Challenge

    It was a long 35 years ago that Richard Nixon proclaimed, Our national goal should be to meet our own energy needs without depending on any foreign sources. Even now that goal of energy security remains elusive although it has occupied an attention almost continuously. Under the sharp increase in oil prices until last year, the past several years have made a huge impact on public awareness and have contributed to a growing concern about energy independence. At the same time, a big difference has come about in attitudes toward climate change. In December 1997, when tough negotiators agreed to the Kyoto Protocol, the United States Senate was at odds with the international community, voting 95-0 against the accord. Now the cultural landscape is full of cover articles in major media publications on such topics as powerful hurricanes, 100 year storms, disappearing species, as well as Al Gores book and film An Inconvenient Truth and his Nobel Peace Prize. Global warming has arrived at center stage as a major concern for economics and national security. Governors are taking dynamic steps to fight climate change, major companies are working for strategic measures, venture capitalists are pouring money into alternative energy, military specialists are engaged by the global security dangers of climate change, and civil societies are demonstrating against global warming.Climate change, energy security

    Coupling between pore formation and phase separation in charged lipid membranes

    We investigated the effect of charge on the membrane morphology of giant unilamellar vesicles (GUVs) composed of various mixtures containing charged lipids. We observed the membrane morphologies by fluorescent and confocal laser microscopy in lipid mixtures consisting of a neutral unsaturated lipid [dioleoylphosphatidylcholine (DOPC)], a neutral saturated lipid [dipalmitoylphosphatidylcholine (DPPC)], a charged unsaturated lipid [dioleoylphosphatidylglycerol (DOPG()^{\scriptsize{(-)}})], a charged saturated lipid [dipalmitoylphosphatidylglycerol (DPPG()^{\scriptsize{(-)}})], and cholesterol (Chol). In binary mixtures of neutral DOPC/DPPC and charged DOPC/DPPG()^{\scriptsize{(-)}}, spherical vesicles were formed. On the other hand, pore formation was often observed with GUVs consisting of DOPG()^{\scriptsize{(-)}} and DPPC. In a DPPC/DPPG()^{\scriptsize{(-)}}/Chol ternary mixture, pore-formed vesicles were also frequently observed. The percentage of pore-formed vesicles increased with the DPPG()^{\scriptsize{(-)}} concentration. Moreover, when the head group charges of charged lipids were screened by the addition of salt, pore-formed vesicles were suppressed in both the binary and ternary charged lipid mixtures. We discuss the mechanisms of pore formation in charged lipid mixtures and the relationship between phase separation and the membrane morphology. Finally, we reproduce the results seen in experimental systems by using coarse-grained molecular dynamics simulations.Comment: 34 pages, 10 figure

According to the results obtained from studies on several animal models of gastrointestinal inflammation, PPARγ has been implicated in the regulation of the immune response, particularly inflammation control, and has gained importance as a potential therapeutic target in the management of gastrointestinal inflammation.     Neutrophil-Dependent Oxidative Stress in Ulcerative Colitis

    Neutrophil accumulation within epithelial crypts and in the intestinal mucosa directly correlates with clinical disease activity and epithelial injury in ulcerative colitis (UC). Current advances have defined the mechanisms by which neutrophils are activated or migrate across mucosal epithelia. A better understanding of this process will likely provide new insights into novel treatment strategies for UC. Especially, activated neutrophils produce reactive oxygen and nitrogen species within intestinal mucosa, which induce oxidative stress. In clinically, we have succeeded to develop a novel granulocytes adsorptive apheresis therapy for UC. In this article, we discuss current advances to define the role of neutrophils-dependent oxidative stress in UC

    Heme oxygenase-1: a novel therapeutic target for gastrointestinal diseases

    Heme oxygenase-1 (HO-1) is the rate-limiting enzyme in the catabolism of heme, followed by production of biliverdin, free iron and carbon monoxide (CO). HO-1 is a stress-responsive protein induced by various oxidative agents. Recent studies demonstrate that the expression of HO-1 in response to different inflammatory mediators may contribute to the resolution of inflammation and has protective effects in several organs against oxidative injury. Although the mechanism underlying the anti-inflammatory actions of HO-1 remains poorly defined, both CO and biliverdin/bilirubin have been implicated in this response. In the gastrointestinal tract, HO-1 is shown to be transcriptionally induced in response to oxidative stress, preconditioning and acute inflammation. Recent studies suggest that the induction of HO-1 expression plays a critical protective role in intestinal damage models induced by ischemia-reperfusion, indomethacin, lipopolysaccharide-associated sepsis, trinitrobenzene sulfonic acid, and dextran sulfate sodium, indicating that activation of HO-1 may act as an endogenous defensive mechanism to reduce inflammation and tissue injury in the gastrointestinal tract. In addition, CO derived from HO-1 is shown to be involved in the regulation in gastro-intestinal motility. These in vitro and in vivo data suggest that HO-1 may be a novel therapeutic target in patients with gastrointestinal diseases

    Involvement of Nitric Oxide in a Rat Model of Carrageenin-Induced Pleurisy

    Some evidence indicates that nitric oxide (NO) contributes to inflammation, while other evidence supports the opposite conclusion. To clarify the role of NO in inflammation, we studied carrageenin-induced pleurisy in rats treated with an NO donor (NOC-18), a substrate for NO formation (L-arginine), and/or an NO synthase inhibitor (S-(2-aminoethyl) isothiourea or NG-nitro-L-arginine). We assessed inflammatory cell migration, nitrite/nitrate values, lipid peroxidation and pro-inflammatory mediators. NOC-18 and L-arginine reduced the migration of inflammatory cells and edema, lowered oxidative stress, and normalized antioxidant enzyme activities. NO synthase inhibitors increased the exudate formation and inflammatory cell number, contributed to oxidative stress, induced an oxidant/antioxidant imbalance by maintaining high O2−, and enhanced the production of pro-inflammatory mediators. L-arginine and NOC-18 reversed the proinflammatory effects of NO synthase inhibitors, perhaps by reducing the expression of adhesion molecules on endothelial cells. Thus, our results indicate that NO is involved in blunting—not enhancing—the inflammatory response

    Possible ability of bovine follicular fluid to attract migrating bull spermatozoa

    AimTo examine the potential of bovine follicular fluid (BFF) to attract bull spermatozoa. MethodsThe ability of the BFF to attract bull sperm was evaluated by observing changes in sperm migration after being placed in a cross-column chamber. The movement parameters of the heads and flagella of the sperm that were attracted to the BFF were analyzed by using the Computer Assisted Sperm Analysis system. ResultsIt was observed that 61.6% of the bull sperm migrated toward the BFF when the BFF was used at a concentration of 0.1%, but 67.2% of the sperm did not migrate toward the BFF at a concentration of 10%. Relatively larger numbers of both precapacitated and postcapacitated bull sperm migrated toward the BFF (0.1%). The ability of the 0.1% BFF to attract sperm probably affected both the normal artificial insemination (AI) fertility sperm and the poor AI fertility spermatozoa. The flagellar curvilinear ratio of the sperm winding to the 0.1% BFF was significantly higher than that of the prewinding sperm. ConclusionThese results could suggest that BFF potentially attracts bull sperm at a certain concentration, irrespective of the capacitation status of the sperm. Although the mechanism by which this attraction occurs remains unclear, these data imply that it could be related to BFF-dependent changes in the sperm flagellar curvilinear ratio.ArticleREPRODUCTIVE MEDICINE AND BIOLOGY.16(2):133-138(2017)journal articl

    Primary Colonic Signet Ring Cell Carcinoma Presenting Carcinocythemia: An Autopsy Case

    Primary colorectal signet ring cell carcinoma (SRCC) is a rare but distinctive type of mucin-producing adenocarcinoma of the large intestine with still controversial clinicopathological features and prognosis. We encountered primary colonic SRCC in a 51-year-old Japanese man with extensive bone metastasis ultimately leading to carcinocythemia before the initiation of chemotherapy and surgical intervention. Three days before death, besides progressive disseminated intravascular coagulation that had been present on admission, hematological examination showed sudden leukocytosis with nonhematopoietic cells that subsequently turned out to be signet ring cells (SRCs). Carcinocythemia, the presence of circulating cancer cells in peripheral blood, is considered to be a rare but an ominous phenomenon occurring in the advanced stage of certain types of cancers, particularly mammary lobular carcinoma. It can be assumed that carcinoma cells lacking intercellular cohesiveness and polarized cell membrane organization, including SRCs as well as lobular carcinoma cells, can readily get access to the peripheral circulation; however, to our knowledge, this is the first report of primary colorectal SRCC that presented carcinocythemia. Extensive bone metastatic sites, in the present case, may have functioned as a reservoir of circulating SRCs
