Common Versus Unique Variance Across Measures of Worry and Rumination: Predictive Utility and Mediational Models for Anxiety and Depression
Repetitive negative thinking (RNT) has been identified as a transdiagnostic construct. However, diagnosis- specific questionnaires have traditionally been used to measure RNT across emotional disorders, and thus the degree to which they assess shared versus unique aspects of RNT is unclear. Furthermore, the degree to which shared versus unique variance across these measures contributes to the prediction of anxiety and depression symptoms is yet to be fully understood. This study had three aims. First, confirmatory factor analysis was used to test the degree to which two common, diagnosis-specific questionnaires assess common versus unique variance in RNT. One questionnaire measured worry whereas the other measured two aspects of rumination (brooding, reflection). Second, the contribution of the shared and unique variance in predicting symptoms of anxiety and depression was determined. Third, the role of shared and unique variance in mediating the relationships between the vulnerability factor of negative affectivity and symptoms of anxiety and depression was assessed
Comparative Study on the Therapeutic Potential of Neurally Differentiated Stem Cells in a Mouse Model of Multiple Sclerosis
Background: Transplantation of neural stem cells (NSCs) is a promising novel approach to the treatment of neuroinflammatory diseases such as multiple sclerosis (MS). NSCs can be derived from primary central nervous system (CNS) tissue or obtained by neural differentiation of embryonic stem (ES) cells, the latter having the advantage of readily providing an unlimited number of cells for therapeutic purposes. Using a mouse model of MS, we evaluated the therapeutic potential of NSCs derived from ES cells by two different neural differentiation protocols that utilized adherent culture conditions and compared their effect to primary NSCs derived from the subventricular zone (SVZ). Methodology/Principal Findings: The proliferation and secretion of pro-inflammatory cytokines by antigen-stimulated splenocytes was reduced in the presence of SVZ-NSCs, while ES cell-derived NSCs exerted differential immunosuppressive effects. Surprisingly, intravenously injected NSCs displayed no significant therapeutic impact on clinical and pathological disease outcomes in mice with experimental autoimmune encephalomyelitis (EAE) induced by recombinant myelin oligodendrocyte glycoprotein, independent of the cell source. Studies tracking the biodistribution of transplanted ES cellderived NSCs revealed that these cells were unable to traffic to the CNS or peripheral lymphoid tissues, consistent with the lack of cell surface homing molecules. Attenuation of peripheral immune responses could only be achieved through multiple high doses of NSCs administered intraperitoneally, which led to some neuroprotective effects within the CNS
Perforocycloides nathalieae new genus and species, an unusual Silurian cyclocystoid (Echinodermata) from Anticosti Island, Québec, Canada
Cyclocystoids are a poorly known, rare, extinct class of bi-facially flattened, disc shaped echinoderms, ranging from the Middle Ordovician to the Early Carboniferous. Articulated cyclocystoids are relatively common in the Ordovician but are rarer in younger strata. Here we describe Perforocycloides nathaliae new genus and species, from the early Silurian of Anticosti Island, Québec, Canada, the first articulated cyclocystoid from the Silurian of North America. This taxon is distinguished from other cyclocystoids by the number of variably sized marginal ossicles, the lack of interseptal plates, and the novelty of pores located in the distal part of the sutures between adjacent marginals on the dorsal surface. These dorsal intermarginal sutural pores led to canals which penetrated the contiguous area of the lateral surface of the marginals and emerged on the ventral surface between the cupules of adjacent marginals. These dorsal intermarginal sutural pores/canals appear to be unique to Perforocycloides and whilst their function is speculative, they provided some form of communication between the dorsal disc and the distal side of the ventral marginals/cupules. Perforocycloides most closely resembles the Ordovician–Silurian genus Zygocycloides, suggesting that this genus may have diversified more widely during the Silurian than previously reported. A review of global Silurian cyclocystoid distribution suggests taxa were geographically confined and that greatest diversity appears to have been located within Baltica. However, it also demonstrates our current limited knowledge.
