    Mining anti-hypertensive peptides in animal food through deep learning: a case study of gastrointestinal digestive products of royal jelly

    To shorten the complex and time-consuming process of the identification method of the traditional food angiotensin-I-converting enzyme (ACE-I) inhibitory peptides, we propose AHTPeptideFusion based on a segmented fusion with the protein language model and deep learning. The statistical analysis found that hydrophobic amino acids, N-terminal valine is a dominant amino acid in the activity of ACE-I inhibitory peptides. In 12 machine learning (ML) algorithms, the transformer outperformed the other 11 models, with the best performance in predicting short and medium peptides. In the external dataset, AHTPeptideFusion fused by transformer and random forest (RF) showed excellent performance (accuracy > 0.9) in predicting ACE-I inhibitory peptides with lengths ranging from 2 to 15 amino acid residues and different activity distributions, and the reliability and accuracy of AHTPeptideFusion was demonstrated by synthetic peptide and ACE-I inhibition experiments. In addition, hydrogen bonding and electrostatic interaction between 4 synthetic peptides and active residues of ACE-I were found by molecular docking. To further explore the ACE-I inhibitory peptides from animal-derived foods, we established an automated pipeline consisting of the trinity of proteomics, virtual enzymatic digestion and AHTPeptideFusion, and tapped the ACE-I inhibitory peptide released from royal jelly after digestion in the gastrointestinal tract. In conclusion, this computational pipeline will become a powerful screening tool for active peptides from animal-derived foods, which can help food scientists accelerate the mining and design of active peptides from animal-derived foods. Overall, AHTPeptideFusion will be a powerful ACE-I inhibitor peptide prediction tool, it can help food scientists accelerate the mining and design of ACE-I inhibitory peptides

    Solving an airport ground service task assignment problem with an exact algorithm.

    In this paper, an airport ground service task assignment problem is studied. A task represents a service, which must be performed by one or multiple ground crew of a shift with required qualification/proficiency within a prescribed time period. For every assigned task, define "task priority" times "task duration" as the "benefit" generated. The objective is to maximize the summation of "benefit" for all the assigned tasks. The problem is modeled as an integer linear programming problem with mathematical formulation. A branch-and-price algorithm is proposed for solving the problem instances to optimality. To expedite the column generation process, an acceleration strategy is proposed. The computational results show that our proposed branch-and-price algorithm is capable of solving large-sized instances and the acceleration strategy is quite effective in reducing the computational time. Moreover, the impact of changing various characteristics of tasks and shifts on the performance of the algorithm is studied in detail with supporting computational experiments. In particular, the impact of reducing the qualifications is significant with 20.82% improvement in the objective value

    Transcriptome analysis reveals association of carotenoid metabolism pathway with fruit color in melon

    Abstract Flesh color is an important quality of melon (Cucumis melo L.) and is determined mainly by carotenoid content, awarding them with colors, aromas, and nutrients. enhancing the nutritional and health benefits of fruits and vegetables for humans. In this study, we performed transcriptomic analysis of two melon inbred line “B-14” (orange-flesh) and “B-6” (white-flesh) at three developmental stages. We observed that the β-carotene content of inbred line “B-6” (14.232 μg/g) was significantly lower than that of inbred line “B-14” (0.534 μg/g). RNA-sequencing and quantitative reverse transcription PCR analyses were performed to identify differentially expressed genes (DEGs) between the two inbred lines at different stages; the DEGs were analyzed using the Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes databases (KEGG). We identified 33 structural DEGs in different developmental periods of the two lines that were related to carotenoid metabolism. Among them, PSY, Z-ISO, ZDS, CRTISO, CCD4, VDE1, and NCED2 were highly correlated with carotenoid content. Thus, this study provides a basis for molecular mechanism of carotenoid biosynthesis and flesh color in melon fruit

    Drying characteristics and processing parameters for microwave-vacuum drying of kiwifruit (Actinidia deliciosa) slices

    This study experimented on microwave-vacuum (MWV) drying of kiwifruit (Actinidia deliciosa) slices as a way to improve drying productivity and the quality of dried fruit. The effects of microwave power density, sample thickness and vacuum degree on the drying characteristics of kiwifruit slices were studied. The results showed that these three factors had significant effects on the vitamin C and chlorophyll contents and on the rehydration capacity of dried kiwifruit slices, as well as the total drying time. The processing parameters of MWV drying of kiwifruit slices were optimized by using Box-Behnken design with a quadratic regression model built by response surface methodology. The optimum processing parameters were experimentally determined as microwave power density at 7.7 W, sample thickness of 6.0 mm and MWV drying for 11.5 min at a vacuum degree of -90 kPa. High-quality dried kiwifruit slices can be created under these conditions using this drying process

    Preparation and Characterization of Coating Based on Protein Nanofibers and Polyphenol and Application for Salted Duck Egg Yolks

    Salted duck egg yolk (SDEY) is one of the traditional pickled egg products in Asian countries, which suffers from the weight loss and deterioration of texture characteristics during storage. To better maintain the texture of SDEY, an edible coating based on whey protein isolate nanofibers (WPNFs) with glycerol (Gly) as a plasticizer and incorporating carvacrol (CA) as an antimicrobial agent was developed. Whey protein isolate (WPI, 5%) was used to self-assemble into WPNFs at 80 °C for 10 h. The particle size, zeta-potential and microstructure of WPNFs–CA emulsion were investigated to evaluate the distribution. Results proved that WPNFs–CA emulsion had smaller particle size and better distribution than WPI–CA emulsion. WPNFs–CA/Gly edible coating was then prepared based on WPNFs–CA emulsion. The WPNFs–CA/Gly edible coating exhibited higher antibacterial activity while the WPNFs–CA/Gly film had smooth and continuous surfaces and better transmittance compared with other samples. Furthermore, weight losses and textural properties changes of SDEYs with WPNFs–CA/Gly coating were evaluated. Results proved that salted duck egg yolks with WPNFs–CA/Gly coating exhibited lower weight losses. Textural properties were significantly improved by the WPNFs–CA/Gly coating on SDEYs than those uncoated samples. It was noted that the egg yolks coated with the WPNFs–CA/Gly coating had the lowest hardness increase rate (18.22%). Hence, WPNF-based coatings may have a good development prospect in the food industry

    Small Molecules Promote the Rapid Generation of Dental Epithelial Cells from Human-Induced Pluripotent Stem Cells

    Human-induced pluripotent stem cells (hiPSCs) offer a promising source for generating dental epithelial (DE) cells. Whereas the existing differentiation protocols were time-consuming and relied heavily on growth factors, herein, we developed a three-step protocol to convert hiPSCs into DE cells in 8 days. In the first phase, hiPSCs were differentiated into non-neural ectoderm using SU5402 (an FGF signaling inhibitor). The second phase involved differentiating non-neural ectoderm into pan-placodal ectoderm and simultaneously inducing the formation of oral ectoderm (OE) using LDN193189 (a BMP signaling inhibitor) and purmorphamine (a SHH signaling activator). In the final phase, OE cells were differentiated into DE through the application of Purmorphamine, XAV939 (a WNT signaling inhibitor), and BMP4. qRT-PCR and immunostaining were performed to examine the expression of lineage-specific markers. ARS staining was performed to evaluate the formation of the mineralization nodule. The expression of PITX2, SP6, and AMBN, the emergence of mineralization nodules, and the enhanced expression of AMBN and AMELX in spheroid culture implied the generation of DE cells. This study delineates the developmental signaling pathways and uses small molecules to streamline the induction of hiPSCs into DE cells. Our findings present a simplified and quicker method for generating DE cells, contributing valuable insights for dental regeneration and dental disease research

    Novel Edible Coating with Antioxidant and Antimicrobial Activities Based on Whey Protein Isolate Nanofibrils and Carvacrol and Its Application on Fresh-Cut Cheese

    The composition and properties of edible coatings (ECs) will significantly influence their effects of food preservation. For the first time, whey protein isolates nanofibers (WPNFs), as a novel material with high hydrophobicity and antioxidant activity, combined with carvacrol (CA) as an antimicrobial agent and glycerol (Gly) as a plasticizer, was used to prepare edible coating (WPNFs-CA/Gly) for preserving fresh-cut Cheddar cheese. The prepared WPNFs and ECs emulsions have been investigated with transmission electron microscopy. Furthermore, the antioxidant activity of ECs emulsions, antimicrobial activity of edible films, and the physical properties of edible films, such as micromorphology, thickness, transparency, and moisture content, have also been evaluated. The weight losses and physical characteristics of both coated and uncoated fresh-cut Cheddar cheese samples have been assessed during storage. The DPPH free radical scavenging rate of WPNFs-CA/Gly emulsion was up to 67.89% and the reducing power was 0.821, which was higher than that of WPI-CA/Gly emulsions. The antimicrobial activity of WPNFs-CA/Gly films was nearly 2.0-fold higher than that of WPNFs/Gly films for the presence of CA. The WPNFs-CA/Gly films had smooth and continuous surfaces, and the transparency reached 49.7% and the moisture content was 26.0%, which was better than that of WPI-CA/Gly films. Furthermore, Cheddar cheese with WPNFs-CA/Gly coatings has shown lower weight losses (15.23%) and better textural properties than those uncoated samples. This in-depth study has provided a valuable and noteworthy approach about the novel edible coating material

    Discrimination and Prediction of Lonicerae japonicae Flos and Lonicerae Flos and Their Related Prescriptions by Attenuated Total Reflectance Fourier Transform Infrared Spectroscopy Combined with Multivariate Statistical Analysis

    LJF and LF are commonly used in Chinese patent drugs. In the Chinese Pharmacopoeia, LJF and LF once belonged to the same source. However, since 2005, the two species have been listed separately. Therefore, they are often misused, and medicinal materials are indiscriminately put in their related prescriptions in China. In this work, firstly, we established a model for discriminating LJF and LF using ATR-FTIR combined with multivariate statistical analysis. The spectra data were further preprocessed and combined with spectral filter transformations and normalization methods. These pretreated data were used to establish pattern recognition models with PLS-DA, RF, and SVM. Results demonstrated that the RF model was the optimal model, and the overall classification accuracy for LJF and LF samples reached 98.86%. Then, the established model was applied in the discrimination of their related prescriptions. Interestingly, the results show good accuracy and applicability. The RF model for discriminating the related prescriptions containing LJF or LF had an accuracy of 100%. Our results suggest that this method is a rapid and effective tool for the successful discrimination of LJF and LF and their related prescriptions

    Fish Gill Inspired Crossflow for Efficient and Continuous Collection of Spilled Oil

    Developing an effective system to clean up large-scale oil spills is of great significance due to their contribution to severe environmental pollution and destruction. Superwetting membranes have been widely studied for oil/water separation. The separation, however, adopts a gravity-driven approach that is inefficient and discontinuous due to quick fouling of the membrane by oil. Herein, inspired by the crossflow filtration behavior in fish gills, we propose a crossflow approach via a hydrophilic, tilted gradient membrane for spilled oil collection. In crossflow collection, as the oil/water flows parallel to the hydrophilic membrane surface, water is gradually filtered through the pores, while oil is repelled, transported, and finally collected for storage. Owing to the selective gating behavior of the water-sealed gradient membrane, the large pores at the bottom with high water flux favor fast water filtration, while the small pores at the top with strong oil repellency allow easy oil transportation. In addition, the gradient membrane exhibits excellent antifouling properties due to the protection of the water layer. Therefore, this bioinspired crossflow approach enables highly efficient and continuous spilled oil collection, which is very promising for the cleanup of large-scale oil spills
