    Geographical gradient of mean age of dengue haemorrhagic fever patients in northern Thailand

    Dengue haemorrhagic fever (DHF) is caused by dengue virus transmitted by Aedes mosquitoes; mean age of patients varies temporally and geographically. Variability in age of patients may be due to differences in transmission intensity or demographic structure. To compare these two hypotheses, the mean age of DHF patients from 90 districts in northern Thailand (1994ā€“1996, 2002ā€“2004) was regressed against (i) Aedes abundance or (ii) demographic variables (birthrate, average age) of the district. We also developed software to quantify direction and strength of geographical gradients of these variables. We found that, after adjusting for socioeconomics, climate, spatial autocorrelation, the mean age of patients was correlated only with Aedes abundance. The geographical gradient of mean age of patients originated from entomological, climate, and socioeconomic gradients. Vector abundance was a stronger determinant of mean age of patients than demographic variables, in northern Thailand

    Identification of mosquito repellent odours from Ocimum forskolei

    <p>Abstract</p> <p>Background</p> <p>Native mosquito repellent plants have a good potential for integrated mosquito control in local settings. <it>Ocimum forskolei</it>, Lamiaceae, is used in Eritrea as a spatial mosquito repellent inside houses, either through crushing fresh plants or burning dry plants. We verified whether active repellent compounds could be identified using gas-chromatography coupled electroantennogram recordings (GC-EAD) with headspace extracts of crushed plants.</p> <p>Results</p> <p>EAD active compounds included (R)-(-)-linalool, (S)-(+)-1-octen-3-ol, trans-caryophyllene, naphthalene, methyl salicylate, (R)-(-)-Ī±-copaene, methyl cinnamate and (E)-ocimene. Of these compounds (R)-(-)-linalool, methyl cinnamate and methyl salicylate reduced landing of female <it>Aedes aegypti </it>on human skin-odor baited tubes. The latter two are novel mosquito repellent compounds.</p> <p>Conclusions</p> <p>The identification of mosquito repellent compounds contributes to deciphering the mechanisms underlying repulsion, supporting the rational design of novel repellents. The three mosquito repellent compounds identified in this study are structurally dissimilar, which may indicate involvement of different sensory neurons in repulsion. Repulsion may well be enhanced through combining different repellent plants (or their synthetic mimics), and can be a locally sustainable part in mosquito control efforts.</p

    Ethnobotanical study of some of mosquito repellent plants in north-eastern Tanzania

    The use of plant repellents against nuisance biting insects is common and its potential for malaria vector control requires evaluation in areas with different level of malaria endemicity. The essential oils of Ocimum suave and Ocimum kilimandscharicum were evaluated against malaria vectors in north-eastern Tanzania. An ethnobotanical study was conducted at Moshi in Kilimanjaro region north-eastern Tanzania, through interviews, to investigate the range of species of plants used as insect repellents. Also, bioassays were used to evaluate the protective potential of selected plants extracts against mosquitoes. The plant species mostly used as repellent at night are: fresh or smoke of the leaves of O. suave and O. kilimandscharicum (Lamiaceae), Azadirachta indica (Meliaceae), Eucalyptus globules (Myrtaceae) and Lantana camara (Verbenaceae). The most popular repellents were O. kilimandscharicum (OK) and O. suave (OS) used by 67% out of 120 households interviewed. Bioassay of essential oils of the two Ocimum plants was compared with citronella and DEET to study the repellence and feeding inhibition of untreated and treated arms of volunteers. Using filter papers impregnated with Ocimum extracts, knockdown effects and mortality was investigated on malaria mosquito Anopheles arabiensis and Anopheles gambiae, including a nuisance mosquito, Culex quinquefasciatus. High biting protection (83% to 91%) and feeding inhibition (71.2% to 92.5%) was observed against three species of mosquitoes. Likewise the extracts of Ocimum plants induced KD90 of longer time in mosquitoes than citronella, a standard botanical repellent. Mortality induced by standard dosage of 30 mg/m2 on filter papers, scored after 24 hours was 47.3% for OK and 57% for OS, compared with 67.7% for citronella. The use of whole plants and their products as insect repellents is common among village communities of north-eastern Tanzania and the results indicate that the use of O. suave and O. kilimandscharicum as a repellent would be beneficial in reducing vector biting. The widespread use of this approach has a potential to complement other control measures

    New Introductions, Spread of Existing Matrilines, and High Rates of Pyrethroid Resistance Result in Chronic Infestations of Bed Bugs (Cimex lectularius L.) in Lower-Income Housing

    Infestations of the common bed bug (Cimex lectularius L.) have increased substantially in the United States in the past 10-15 years. The housing authority in Harrisonburg, Virginia, conducts heat-treatments after bed bugs are detected in a lower-income housing complex, by treating each infested unit at 60Ā°C for 4-6 hours. However, a high frequency of recurrent infestations called into question the efficacy of this strategy. Genetic analysis using Bayesian clustering of polymorphic microsatellite loci from 123 bed bugs collected from 23 units from May 2012 to April 2013 in one building indicated that (a) 16/21 (73%) infestations were genetically similar, suggesting ineffective heat-treatments or reintroductions from within the building or from a common external source, followed by local spread of existing populations; and (b) up to 5 of the infestations represented new genotypes, indicating that 5 new populations were introduced into this building in one year, assuming they were not missed in earlier screens. There was little to no gene flow among the 8 genetic clusters identified in the building. Bed bugs in the U.S. often possess one or both point mutations in the voltage-gated sodium channel, termed knockdown resistance (kdr), from valine to leucine (V419L) and leucine to isoleucine (L925I) that confer target-site resistance against pyrethroid insecticides. We found that 48/121 (40%) bed bugs were homozygous for both kdr mutations (L419/I925), and a further 59% possessed at least one of the kdr mutations. We conclude that ineffective heat treatments, new introductions, reintroductions and local spread, and an exceptionally high frequency of pyrethroid resistance are responsible for chronic infestations in lower-income housing. Because heat treatments fail to protect from reintroductions, and pesticide use has not decreased the frequency of infestations, preventing new introductions and early detection are the most effective strategies to avoid bed bug infestations in multistory apartment buildings

    Geographical structure of dengue transmission and its determinants in Thailand

    Expansion of dengue has been attributed to urbanization. To test this concept, we examined dengue transmission intensities in Thailand. We used the inverse of mean age of dengue haemorrhagic fever (DHF) cases as a surrogate of dengue transmission intensity (or force of infection). The transmission intensity in Bangkok decreased rapidly since the mid-1990s, to levels that are currently lower than in other regions. Regression analysis revealed that transmission intensity is highest in the Northeastern rural region, mainly due to scarcity of private water wells. Private wells reduce the need for household water containers, the major breeding sites for vectors. Cumulatively, these results show that urbanization is not necessarily associated with intense dengue transmission in Thailand. Paradoxically, the DHF incidence in Bangkok has surpassed other regions despite declines in transmission intensity. This finding implies the existence of endemic stability (i.e. low incidence of a clinical illness in spite of high transmission intensity)
