    Infections associated with eating seed sprouts: an international concern.

    Recent outbreaks of Salmonella and Escherichia coli O157:H7 infections associated with raw seed sprouts have occurred in several countries. Subjective evaluations indicate that pathogens can exceed 107 per gram of sprouts produced from inoculated seeds during sprout production without adversely affecting appearance. Treating seeds and sprouts with chlorinated water or other disinfectants fails to eliminate the pathogens. A comprehensive approach based on good manufacturing practices and principles of hazard analysis and critical control points can reduce the risk of sprout-associated disease. Until effective measures to prevent sprout-associated illness are identified, persons who wish to reduce their risk of foodborne illness from raw sprouts are advised not to eat them; in particular, persons at high risk for severe complications of infections with Salmonella or E. coli O157:H7, such as the elderly, children, and those with compromised immune systems, should not eat raw sprouts

    Broadly targeted human cytomegalovirus-specific CD4+ and CD8+ T cells dominate the memory compartments of exposed subjects

    Human cytomegalovirus (HCMV) infections of immunocompetent hosts are characterized by a dynamic, life-long interaction in which host immune responses, particularly of T cells, restrain viral replication and prevent disease but do not eliminate the virus or preclude transmission. Because HCMV is among the largest and most complex of known viruses, the T cell resources committed to maintaining this balance have never been characterized completely. Here, using cytokine flow cytometry and 13,687 overlapping 15mer peptides comprising 213 HCMV open reading frames (ORFs), we found that 151 HCMV ORFs were immunogenic for CD4+ and/or CD8+ T cells, and that ORF immunogenicity was influenced only modestly by ORF expression kinetics and function. We further documented that total HCMV-specific T cell responses in seropositive subjects were enormous, comprising on average ∼10% of both the CD4+ and CD8+ memory compartments in blood, whereas cross-reactive recognition of HCMV proteins in seronegative individuals was limited to CD8+ T cells and was rare. These data provide the first glimpse of the total human T cell response to a complex infectious agent and will provide insight into the rules governing immunodominance and cross-reactivity in complex viral infections of humans

    The binary system of the spinning-top Be star Achernar

    Achernar, the closest and brightest classical Be star, presents rotational flattening, gravity darkening, occasional emission lines due to a gaseous disk, and an extended polar wind. It is also a member of a close binary system with an early A-type dwarf companion. We aim to determine the orbital parameters of the Achernar system and to estimate the physical properties of the components. We monitored the relative position of Achernar B using a broad range of high angular resolution instruments of the VLT/VLTI (VISIR, NACO, SPHERE, AMBER, PIONIER, GRAVITY, and MATISSE) over a period of 13 years (2006-2019). These astrometric observations are complemented with a series of more than 700 optical spectra for the period from 2003 to 2016. We determine that Achernar B orbits the Be star on a seven-year period, eccentric orbit (e = 0.7255 +/- 0.0014) which brings the two stars within 2 au at periastron. The mass of the Be star is found to be mA = 6.0 +/- 0.6 Msun for a secondary mass of mB = 2.0 +/- 0.1 Msun. We find a good agreement of the parameters of Achernar A with the evolutionary model of a critically rotating star of 6.4 Msun at an age of 63 million years. We also identify a resolved comoving low-mass star, which leads us to propose that Achernar is a member of the Tucana-Horologium moving group. Achernar A is presently in a short-lived phase of its evolution following the turn-off, during which its geometrical flattening ratio is the most extreme. Considering the orbital parameters, no significant interaction occurred between the two components, demonstrating that Be stars may form through a direct, single-star evolution path without mass transfer. Since component A will enter the instability strip in a few hundred thousand years, Achernar appears to be a promising progenitor of the Cepheid binary systems.Comment: 27 pages, Astronomy & Astrophysics, in press.     Multi-task learning for subthalamic nucleus identification in deep brain stimulation

    Deep brain stimulation (DBS) of Subthalamic nucleus (STN) is the most successful treatment for advanced Parkinson’s disease. Localization of the STN through Microelectrode recordings (MER) is a key step during the surgery. However, it is a complex task even for a skilled neurosurgeon. Different researchers have developed methodologies for processing and classification of MER signals to locate the STN. Previous works employ the classical paradigm of supervised classification, assuming independence between patients. The aim of this paper is to introduce a patient-dependent learning scenario, where the predictive ability for STN identification at the level of a particular patient, can be used to improve the accuracy for STN identification in other patients. Our inspiration is the multi-task learning framework, that has been receiving increasing interest within the machine learning community in the last few years. To this end, we employ the multi-task Gaussian processes framework that exhibits state of the art performance in multi-task learning problems. In our context, we assume that each patient undergoing DBS is a different task, and we refer to the method as multi-patient learning. We show that the multi-patient learning framework improves the accuracy in the identification of STN in a range from 4.1 to 7.7%, compared to the usual patient-independent setup, for two different datasets. Given that MER are non stationary and noisy signals. Traditional approaches in machine learning fail to recognize accurately the STN during DBS. By contrast in our proposed method, we properly exploit correlations between patients with similar diseases, obtaining an additional information. This information allows to improve the accuracy not only for locating STN for DBS but also for other biomedical signal classification problems

    Alfalfa Seed Decontamination in Salmonella Outbreak

    Based on in vitro data, the U.S. Food and Drug Administration recommends chemical disinfection of raw sprout seeds to reduce enteric pathogens contaminating the seed coats. However, little is known about the effectiveness of decontamination at preventing human disease. In 1999, an outbreak of Salmonella enterica serotype Mbandaka occurred in Oregon, Washington, Idaho, and California. Based on epidemiologic and pulsed-field gel electrophoresis evidence from 87 confirmed cases, the outbreak was linked to contaminated alfalfa seeds grown in California’s Imperial Valley. Trace-back and trace-forward investigations identified a single lot of seeds used by five sprout growers during the outbreak period. Cases of salmonellosis were linked with two sprout growers who had not employed chemical disinfection; no cases were linked to three sprout growers who used disinfection. This natural experiment provides empiric evidence that chemical disinfection can reduce the human risk for disease posed by contaminated seed sprouts
