    Manure and lagoon nutrients from dairies using flush systems

    Nine primar,y lagoons and solids storage basins were sampled on Kansas dairies using flush systems. These samples were analyzed for nutrient content of wastewater and sand manure. The manure moisture content in the storage basins averaged 81%. The average totals of nitrogen, phosphate, and potash were 3450, 1345, and 1420 mg/L, respectively, for flushing systems. The average totals of nitrogen, phosphate, and potash in the lagoon samples were 816, 337, and 1134 mg/L, respectively, for dairies using recycled water for flushing alleys. These data and previously reported data indicate that lagoon effluent and manure removed from basins must be managed differently between dairies using flush versus scrape systems

    From microanalysis to supercontinents: insights from the Rio Apa Terrane into the Mesoproterozoic SW Amazonian Craton evolution during Rodinia assembly

    First published: 10 November 2021Deciphering the tectono-metamorphic evolution of Precambrian terranes can be difficult due to reworking by later superimposed events. Whole-rock elemental and isotopic geochemistry and zircon U–Pb geochronology are often employed in those studies, but these approaches are often not sensitive to the presence of multiple events and medium-grade metamorphic episodes. The Rio Apa Terrane (RAT), an allochthonous fragment of the Amazonian Craton, is a crustal block with a well-characterized evolution but with no detailed thermal constraints for its tectono-metamorphic evolution. In contrast to previous studies, we show the existence of four tectono-metamorphic events at c. 1780 Ma, c. 1625 Ma, c. 1420- 1340 Ma and c. 1300-1200 Ma on the basis of apatite, titanite and rutile U–Pb–REE, in-situ white-mica Rb-Sr and in-situ garnet Lu-Hf geochronology combined with mineral chemistry and phase-equilibria modelling. The c. 1780 Ma event is recorded in the basement of the Western domain, representing an extensional event coeval with the development of its Eastern domain in response to the retreat stage of the accretionary system. This is followed by juxtaposition of the Western and Eastern domains along a major crustal boundary at ca. 1625 Ma, which is defined by the magnetic profiles and zircon U–Pb-Hf data across the boundary. The third and fourth events correspond to progressive high-pressure/medium temperature (HP/MT) metamorphism, characterized by an anticlockwise P-T path, suggesting a convergent-to-collisional tectonic setting. The RAT was accreted to the adjoining Paraguá Terrane at c. 1420-1340 Ma under an isobaric P-T evolution spanning ~530 to 600 °C and ~10.0 kbar. Subsequently, the combined Rio Apa and Paraguá terranes collided with the SW Amazonian Craton at c. 1300-1200 Ma, reaching P-T conditions of ~560-580 °C and ~10.9- 11.7 kbar during crustal thickening. This study reveals for the first time the existence of a HP/MT metamorphic evolution related to the growth of the SW Amazonian Craton as part of an accretionary orogenic system during Rodinia assembly in the Paleo- to Mesoproterozoic.Bruno V.     Fast automated cell phenotype image classification

    BACKGROUND: The genomic revolution has led to rapid growth in sequencing of genes and proteins, and attention is now turning to the function of the encoded proteins. In this respect, microscope imaging of a protein's sub-cellular localisation is proving invaluable, and recent advances in automated fluorescent microscopy allow protein localisations to be imaged in high throughput. Hence there is a need for large scale automated computational techniques to efficiently quantify, distinguish and classify sub-cellular images. While image statistics have proved highly successful in distinguishing localisation, commonly used measures suffer from being relatively slow to compute, and often require cells to be individually selected from experimental images, thus limiting both throughput and the range of potential applications. Here we introduce threshold adjacency statistics, the essence which is to threshold the image and to count the number of above threshold pixels with a given number of above threshold pixels adjacent. These novel measures are shown to distinguish and classify images of distinct sub-cellular localization with high speed and accuracy without image cropping. RESULTS: Threshold adjacency statistics are applied to classification of protein sub-cellular localization images. They are tested on two image sets (available for download), one for which fluorescently tagged proteins are endogenously expressed in 10 sub-cellular locations, and another for which proteins are transfected into 11 locations. For each image set, a support vector machine was trained and tested. Classification accuracies of 94.4% and 86.6% are obtained on the endogenous and transfected sets, respectively. Threshold adjacency statistics are found to provide comparable or higher accuracy than other commonly used statistics while being an order of magnitude faster to calculate. Further, threshold adjacency statistics in combination with Haralick measures give accuracies of 98.2% and 93.2% on the endogenous and transfected sets, respectively. CONCLUSION: Threshold adjacency statistics have the potential to greatly extend the scale and range of applications of image statistics in computational image analysis. They remove the need for cropping of individual cells from images, and are an order of magnitude faster to calculate than other commonly used statistics while providing comparable or better classification accuracy, both essential requirements for application to large-scale approaches

    Crucial Physical Dependencies of the Core-Collapse Supernova Mechanism

    We explore with self-consistent 2D F{\sc{ornax}} simulations the dependence of the outcome of collapse on many-body corrections to neutrino-nucleon cross sections, the nucleon-nucleon bremsstrahlung rate, electron capture on heavy nuclei, pre-collapse seed perturbations, and inelastic neutrino-electron and neutrino-nucleon scattering. Importantly, proximity to criticality amplifies the role of even small changes in the neutrino-matter couplings, and such changes can together add to produce outsized effects. When close to the critical condition the cumulative result of a few small effects (including seeds) that individually have only modest consequence can convert an anemic into a robust explosion, or even a dud into a blast. Such sensitivity is not seen in one dimension and may explain the apparent heterogeneity in the outcomes of detailed simulations performed internationally. A natural conclusion is that the different groups collectively are closer to a realistic understanding of the mechanism of core-collapse supernovae than might have seemed apparent.Comment: 25 pages; 10 figure

    Diagnosis of aortic graft infection : a case definition by the management of aortic graft infection collaboration (MAGIC)

    Objective/Background The management of aortic graft infection (AGI) is highly complex and in the absence of a universally accepted case definition and evidence-based guidelines, clinical approaches and outcomes vary widely. The objective was to define precise criteria for diagnosing AGI. Methods A process of expert review and consensus, involving formal collaboration between vascular surgeons, infection specialists, and radiologists from several English National Health Service hospital Trusts with large vascular services (Management of Aortic Graft Infection Collaboration [MAGIC]), produced the definition. Results Diagnostic criteria from three categories were classified as major or minor. It is proposed that AGI should be suspected if a single major criterion or two or more minor criteria from different categories are present. AGI is diagnosed if there is one major plus any criterion (major or minor) from another category. (i) Clinical/surgical major criteria comprise intraoperative identification of pus around a graft and situations where direct communication between the prosthesis and a nonsterile site exists, including fistulae, exposed grafts in open wounds, and deployment of an endovascular stent-graft into an infected field (e.g., mycotic aneurysm); minor criteria are localized AGI features or fever ≥38°C, where AGI is the most likely cause. (ii) Radiological major criteria comprise increasing perigraft gas volume on serial computed tomography (CT) imaging or perigraft gas or fluid (≥7 weeks and ≥3 months, respectively) postimplantation; minor criteria include other CT features or evidence from alternative imaging techniques. (iii) Laboratory major criteria comprise isolation of microorganisms from percutaneous aspirates of perigraft fluid, explanted grafts, and other intraoperative specimens; minor criteria are positive blood cultures or elevated inflammatory indices with no alternative source. Conclusion This AGI definition potentially offers a practical and consistent diagnostic standard, essential for comparing clinical management strategies, trial design, and developing evidence-based guidelines. It requires validation that is planned in a multicenter, clinical service database supported by the Vascular Society of Great Britain & Ireland

    The stable fly, Stomoxys calcitrans, is a major blood-feeding pest of livestock that has near worldwide distribution, causing an annual cost of over $2 billion for control and product loss in the USA alone. Control of these flies has been limited to increased sanitary management practices and insecticide application for suppressing larval stages. Few genetic and molecular resources are available to help in developing novel methods for controlling stable flies. This study examines stable fly biology by utilizing a combination of high-quality genome sequencing and RNA-Seq analyses targeting multiple developmental stages and tissues. In conjunction, 1600 genes were manually curated to characterize genetic features related to stable fly reproduction, vector host interactions, host-microbe dynamics, and putative targets for control. Most notable was characterization of genes associated with reproduction and identification of expanded gene families with functional associations to vision, chemosensation, immunity, and metabolic detoxification pathways. The combined sequencing, assembly, and curation of the male stable fly genome followed by RNA-Seq and downstream analyses provide insights necessary to understand the biology of this important pest. These resources and new data will provide the groundwork for expanding the tools available to control stable fly infestations. The close relationship of Stomoxys to other blood-feeding (horn flies and Glossina) and non-blood-feeding flies (house flies, medflies, Drosophila) will facilitate understanding of the evolutionary processes associated with development of blood feeding among the Cyclorrhapha

    Clinical indicators for common paediatric conditions: processes, provenance and products of the CareTrack Kids study

    BACKGROUND:In order to determine the extent to which care delivered to children is appropriate (in line with evidence-based care and/or clinical practice guidelines (CPGs)) in Australia, we developed a set of clinical indicators for 21 common paediatric medical conditions for use across a range of primary, secondary and tertiary healthcare practice facilities. METHODS:Clinical indicators were extracted from recommendations found through systematic searches of national and international guidelines, and formatted with explicit criteria for inclusion, exclusion, time frame and setting. Experts reviewed the indicators using a multi-round modified Delphi process and collaborative online wiki to develop consensus on what constituted appropriate care. RESULTS:From 121 clinical practice guidelines, 1098 recommendations were used to draft 451 proposed appropriateness indicators. In total, 61 experts (n = 24 internal reviewers, n = 37 external reviewers) reviewed these indicators over 40 weeks. A final set of 234 indicators resulted, from which 597 indicator items were derived suitable for medical record audit. Most indicator items were geared towards capturing information about under-use in healthcare (n = 551, 92%) across emergency department (n = 457, 77%), hospital (n = 450, 75%) and general practice (n = 434, 73%) healthcare facilities, and based on consensus level recommendations (n = 451, 76%). The main reason for rejecting indicators was 'feasibility' (likely to be able to be used for determining compliance with 'appropriate care' from medical record audit). CONCLUSION:A set of indicators was developed for the appropriateness of care for 21 paediatric conditions. We describe the processes (methods), provenance (origins and evolution of indicators) and products (indicator characteristics) of creating clinical indicators within the context of Australian healthcare settings. Developing consensus on clinical appropriateness indicators using a Delphi approach and collaborative online wiki has methodological utility.     Bounds on the possible evolution of the Gravitational Constant from Cosmological Type-Ia Supernovae

    Recent high-redshift Type Ia supernovae results can be used to set new bounds on a possible variation of the gravitational constant GG. If the local value of GG at the space-time location of distant supernovae is different, it would change both the kinetic energy release and the amount of 56^{56}Ni synthesized in the supernova outburst. Both effects are related to a change in the Chandrasekhar mass MChG3/2M_{Ch} \propto G^{-3/2}. In addition, the integrated variation of GG with time would also affect the cosmic evolution and therefore the luminosity distance relation. We show that the later effect in the magnitudes of Type Ia supernovae is typically several times smaller than the change produced by the corresponding variation of the Chandrasekhar mass. We investigate in a consistent way how a varying GG could modify the Hubble diagram of Type Ia supernovae and how these results can be used to set upper bounds to a hypothetical variation of GG. We find G/G_0 \la 1.1 and G'/G \la 10^{-11} yr^{-1} at redshifts z0.5z\simeq 0.5. These new bounds extend the currently available constrains on the evolution of GG all the way from solar and stellar distances to typical scales of Gpc/Gyr, i.e. by more than 15 orders of magnitudes in time and distance.Comment: 9 pages, 4 figures, Phys.     Speeding up the spatial adiabatic passage of matter waves in optical microtraps by optimal control

    We numerically investigate the performance of atomic transport in optical microtraps via the so called spatial adiabatic passage technique. Our analysis is carried out by means of optimal control methods, which enable us to determine suitable transport control pulses. We investigate the ultimate limits of the optimal control in speeding up the transport process in a triple well configuration for both a single atomic wave packet and a Bose-Einstein condensate within a regime of experimental parameters achievable with current optical technology.Comment: 17 pages, 14 figure
