Preparation of mono-substituted malonic acid half oxyesters (SMAHOs).
The use of mono-substituted malonic acid half oxyesters (SMAHOs) has been hampered by the sporadic references describing their preparation. An evaluation of different approaches has been achieved, allowing to define the best strategies to introduce diversity on both the malonic position and the ester function. A classical alkylation step of a malonate by an alkyl halide followed by a monosaponification gave access to reagents bearing different substituents at the malonic position, including functionalized derivatives. On the other hand, the development of a monoesterification step of a substituted malonic acid derivative proved to be the best entry for diversity at the ester function, rather than the use of an intermediate Meldrum acid. Both these transformations are characterized by their simplicity and efficiency, allowing a straightforward access to SMAHOs from cheap starting materials
DefectâDriven Magnetization Configuration of Isolated Linear Assemblies of Iron Oxide Nanoparticles
International audienceThe magnetization state of one-dimensional magnetic nanoparticle chains plays a key role for a wide range of applications ranging from diagnosis and therapy in medicine to actuators, sensors and quantum recording media. The interplay between the exact particle orientation and the magnetic anisotropy is in turn crucial for controlling the overall magnetization state with high precision. Here, we report on a three-dimensional description of the magnetic structure of one-NP-wide chains. In this aim, we combined two complementary experimental techniques, magnetic force microscopy (MFM) and electronic holography (EH) which are sensitive to out-of-plane and in-plane magnetization components, respectively. We extended our approach to micromagnetic simulations which provided results in good agreement with MFM and EH. The findings are at variance with the known results on unidirectional nanoparticle assemblies, and show that magnetization is rarely strictly collinear to the chain axis. The magnetic structure of one-NP-wide chains can be interpreted as head-to-head magnetic domain structures with off-axis magnetization components, which is very sensitive to morphological defects in the chain structure such as minute size variation of NPs, tiny misalignment of NPs and/or crystal orientation with respect to easy magnetization axis
Nanotrench for nano and microparticle electrical interconnects
We present a simple and versatile patterning procedure for the reliable and reproducible fabrication of high aspect ratio (10 4 ) electrical interconnects that have separation distances down to 20 nm and lengths of several hundreds of microns. The process uses standard optical lithography techniques and allows parallel processing of many junctions, making it easily scalable and industrially relevant. We demonstrate the suitability of these nanotrenches as electrical interconnects for addressing micro and nanoparticles by realizing several circuits with integrated species. Furthermore, low impedance metal-metal low contacts are shown to be obtained when trapping a single metal-coated microsphere in the gap, emphasizing the intrinsic good electrical conductivity of the interconnects, even though a wet process is used. Highly resistive magnetite-based nanoparticles networks also demonstrate the advantage of the high aspect ratio of the nanotrenches for providing access to electrical properties of highly resistive materials, with leakage current levels below 1 pA. © 2010 IOP Publishing Ltd
Epitope-specific humoral responses to human cytomegalovirus glycoprotein-B vaccine with MF59: Anti-AD2 levels correlate with protection from viremia
The human cytomegalovirus (HCMV) virion envelope protein glycoprotein B (gB) is essential for viral entry and represents a major target for humoral responses following infection. Previously, a phase-2 placebo-controlled clinical trial conducted in solid organ transplant candidates demonstrated that vaccination with gB plus MF59 adjuvant significantly increased gB ELISA antibody levels whose titer correlated directly with protection against post-transplant viremia. The aim of the current study was to investigate in more detail this protective humoral response in vaccinated seropositive transplant recipients. We focussed on four key antigenic domains (AD) of gB; AD1, AD2, AD4 and AD5 measuring antibody levels in patient sera and correlating these with post-transplant HCMV viremia. Vaccination of seropositive patients significantly boosted pre-existing antibody levels against the immunodominant region AD1 as well as against AD2, AD4 and AD5. A decreased incidence of viremia correlated with higher antibody titers against AD2 but not with antibody titers against the other three ADs. Overall, these data support the hypothesis that antibodies against AD2 are a major component of the immune protection of seropositives seen following vaccination with gB/MF59 vaccine and identify a correlate of protective immunity in allograft patients

Stability and thermoelectric performance of doped higher manganese silicide materials solidi fied by RGS (ribbon growth on substrate) synthesis
Large scale deployment of thermoelectric devices requires that the thermoelectric materials
have stable electrical, thermal and mechanical properties under the conditions of operation. In this
study we examine the high temperature stability of higher manganese silicide (HMS) materials
prepared by the RGS (ribbon growth on substrate) technique. In particular we characterize the effect of
element substitution on the structural and electrical changes occurring at the hot side of temperatures
of thermoelectric devices relevant to this material (600°C). Only by using suitable substitution (4%
vanadium at the Mn site) can we obtain temperature-independent structural parameters in the range
20°C - 600°C, a condition that results in stable electrical properties. Additionally, we show that 4%
vanadium substitution at the Mn site offers the best thermoelectric figure of merit among the different
compositions reported here with ZTmax=0.52, a value comparable to the state of the art for HMS
materials. Our analysis suggests that ionized impurity scattering is responsible for the better
performance of this material
Design of Fe3âxO4 raspberry decorated graphene nanocomposites with high performances in lithium-ion battery
Fe3âxO4 raspberry shaped nanostructures/graphene nanocomposites were synthesized by a one-step polyol-solvothermal method to be tested as electrode materials for Li-ion battery (LIB). Indeed, Fe3âxO4 raspberry shaped nanostructures consist of original oriented aggregates of Fe3âxO4 magnetite nanocrystals, ensuring a low oxidation state of magnetite and a hollow and porous structure, which has been easily combined with graphene sheets. The resulting nanocomposite powder displays a very homogeneous spatial distribution of Fe3âxO4 nanostructures at the surface of the graphene sheets. These original nanostructures and their strong interaction with the graphene sheets resulted in very small capacity fading upon Li+ ion intercalation. Reversible capacity, as high as 660 mAh/g, makes this material promising for anode in Li-ion batteries application
Two dimensional dipolar coupling in monolayers of silver and gold nanoparticles on a dielectric substrate
The dimensionality of assembled nanoparticles plays an important role in their optical and magnetic properties, via dipolar effects and the interaction with their environment. In this work we develop a methodology for distinguishing between two (2D) and three (3D) dimensional collective interactions on the surface plasmon resonance of assembled metal nanoparticles. Towards that goal, we elaborate different sets of Au and Ag nanoparticles as suspensions, random 3D arrangements and well organized 2D arrays. Then we model their scattering cross-section using effective field methods in dimension n, including interparticle as well as particle-substrate dipolar interactions. For this modelling, two effective field medium approaches are employed, taking into account the filling factors of the assemblies. Our results are important for realizing photonic amplifier devices
Extending the clinical spectrum of X-linked Tonne-Kalscheuer syndrome (TOKAS):new insights from the fetal perspective
INTRODUCTION:Â Tonne-Kalscheuer syndrome (TOKAS) is a recessive X-linked multiple congenital anomaly disorder caused by RLIM variations. Of the 41 patients reported, only 7 antenatal cases were described.METHOD:Â After the antenatal diagnosis of TOKAS by exome analysis in a family followed for over 35 years because of multiple congenital anomalies in five male fetuses, a call for collaboration was made, resulting in a cohort of 11 previously unpublished cases.RESULTS:Â We present a TOKAS antenatal cohort, describing 11 new cases in 6 French families. We report a high frequency of diaphragmatic hernia (9 of 11), differences in sex development (10 of 11) and various visceral malformations. We report some recurrent dysmorphic features, but also pontocerebellar hypoplasia, pre-auricular skin tags and olfactory bulb abnormalities previously unreported in the literature. Although no clear genotype-phenotype correlation has yet emerged, we show that a recurrent p.(Arg611Cys) variant accounts for 66% of fetal TOKAS cases. We also report two new likely pathogenic variants in RLIM, outside of the two previously known mutational hotspots.CONCLUSION:Â Overall, we present the first fetal cohort of TOKAS, describe the clinical features that made it a recognisable syndrome at fetopathological examination, and extend the phenotypical spectrum and the known genotype of this rare disorder.</p
Haploinsufficiency of ARFGEF1 is associated with developmental delay, intellectual disability, and epilepsy with variable expressivity
ADP ribosylation factor guanine nucleotide exchange factors (ARFGEFs) are a family of proteins implicated in cellular trafficking between the Golgi apparatus and the plasma membrane through vesicle formation. Among them is ARFGEF1/BIG1, a protein involved in axon elongation, neurite development, and polarization processes. ARFGEF1 has been previously suggested as a candidate gene for different types of epilepsies, although its implication in human disease has not been well characterized. International data sharing, in silico predictions, and in vitro assays with minigene study, western blot analyses, and RNA sequencing. We identified 13 individuals with heterozygous likely pathogenic variants in ARFGEF1. These individuals displayed congruent clinical features of developmental delay, behavioral problems, abnormal findings on brain magnetic resonance image (MRI), and epilepsy for almost half of them. While nearly half of the cohort carried de novo variants, at least 40% of variants were inherited from mildly affected parents who were clinically re-evaluated by reverse phenotyping. Our in silico predictions and in vitro assays support the contention that ARFGEF1-related conditions are caused by haploinsufficiency, and are transmitted in an autosomal dominant fashion with variable expressivity. We provide evidence that loss-of-function variants in ARFGEF1 are implicated in sporadic and familial cases of developmental delay with or without epilepsy
