Existential Abuse of Readers in Samuel Beckett’s Malone Dies

Malone Dies marks the point where Samuel Beckett foremost turns to “metaphysical destruction” of “untrue self,” and Derrida’s critique of the notion of “self-presence” of the subject. In this article, I examine Beckett’s literary absurdities to his readers’ concerns of “abuse” through them. For this investigation Malone Dies posits a stream of conflicting “linguistic nihilism” to the concerns of deconstructing “untrue self,” arguably, which will reflect how abuse of Beckettian readers is stimulated. In this context, abuse is specific forms of emotional tensions aroused by the readers encounter with the Beckettian intersubjectivity. Direct support to private firms - evidence on effectiveness

Governments use a variety of instruments to provide direct support to private enterprises. These include the provision of finance (subsidized and/or directed credit) and business development services (management and marketing advice to small businesses, agricultural extension services, support for enterprise level training and support to technology development). These interventions are distinct from those that support enterprises indirectly by establishing a policy and legal environment conducive to enterprise development. How effective have these direct support schemes been? This paper attempts to provide some answers to this question by surveying the available literature on the effectiveness of direct support interventions. Where available, impact evaluations suggest that the performance has been mixed at best. The evidence indicates that active intervention does not work unless the basic environment for private sector development is sound. The Disease Pattern and Utilisation of Health Care Services in Pakistan

Health is an important aspect of human life. In general terms, better health status of individuals reflects reduced illnesses, low level of morbidity, and less burden of disease in a given population. It is widely recognised that improved health not only lowers mortality, morbidity and level of fertility, but also contributes to increased productivity and regular school attendance of children as a result of fewer work days lost due to illness, which in turn have implications for economic and social well-being of the population at large. Hence investing in health is vital for promoting human resource development and economic growth in a country [World Bank (1993)]. A view of Pakistan’s health profile indicates that the sector has expanded considerably in terms of physical infrastructure and its manpower in both the public and private sector. This has contributed to some improvement in selected health status indicators over the years. However, the public health care delivery system has been inadequate in meeting the needs of the fast growing population and in filtering down its benefits to the gross-root level. As such, Pakistan still has one of the highest rates of infant and child mortality, total fertility and maternal mortality when compared with many other countries in the Asian region [UNDP (2000)]. Due to low priority given to social sector development in the past and low budgetary allocations made to the health sector, the evidence shows that mortality and morbidity indices have not reduced to the desired level and large gaps remain in the quality of care indicators, especially in rural areas [Federal Bureau of Statistics (2000)]. High levels of infant and child mortality and fertility in Pakistan point towards the fact that health and illness problems are severe for young children and mothers.

Using Rb-TnSeq to Analyze Natural Variation in Saccharomyces cerevisiae

One of the main challenges in biology today is the characterization of millions of genes of unknown function being continuously identified in sequencing studies. Transposon mutagenesis is a technique that has been widely used for annotating gene function and has now been combined with next-generation sequencing (Tn-Seq) to assess mutant fitness on a genome wide basis. However, Tn-Seq approaches are often constrained by laborious library preparation protocols which limit the number of organisms or conditions that can be assessed. Random bar code transposon-site sequencing (RB-TnSeq), is a transposon sequencing technique that streamlines library preparation and increases the throughput of mutant fitness profiling by incorporating random DNA barcodes into Tn5 Transposons. Rb-TnSeq has been successfully used for high throughput mutant fitness assays in diverse bacterial species. However, this technique is yet to be applied to a eukaryotic model organism. The goal of this study is to develop tools that allow the construction of barcoded mutant libraries in saccharomyces cerevisiae and describes methods for producing barcoded mutant libraries using a plasmid based or transposome based approach. These library construction protocols can prove to be powerful tools for studying gene function in S. cerevisiae on a genome wide basis
