    Grain and malt milling energies in sorghum and their relationships with extract and diastatic power

    Twenty three sorghum genotypes were shown to vary widely for a range of grain and malt quality characters. Grain milling energy did not give an indication of the likely malting performance of sorghum samples. There were, however, close relationships between malt milling energy and both % extract and diastatic power, if samples with unusually high or low grain nitrogen contents were excluded from the population under study. Rapid screening of very small malt samples for milling energy and diastatic power could form the basis for malting quality selection in early generations of sorghum breeding programme

    The mammalian respiratory tract is densely innervated by autonomic and sensory nerves around airways and blood vessels. Subsets of these nerves contain a number of putative neurotransmitter peptides, such as substance P and calcitonin gene-related peptide (CGRP) in sensory nerves and vasoactive intestinal polypeptide (VIP), possibly serving autonomic functions. CGRP is also found in endocrine cells in rat airway epithelium. These peptides are all pharmacologically potent effectors of bronchial and vascular smooth muscle and bronchial secretion. Their functions in vivo are less well established. We have therefore examined the effects of inhaled acrolein, a sensory irritant, on three pulmonary neuropeptides: CGRP, substance P, and VIP. Groups of rats (n = 3 each) were exposed for 10 min to acrolein in air (Ct = 510, 1858, and 5693 mg.min/m3) or to air alone. Fifteen minutes later they were killed (pentabarbitone IP) and their respiratory tracts were dissected and fixed in 0.4% p-benzoquinone solution. Cryostat sections were stained by indirect immunofluorescence for a general nerve marker (PGP 9.5) and neuropeptides. The acrolein-treated animals had a dose-related decrease in tracheal substance P- and CGRP-immunoreactive nerve fibers compared with controls. No change was seen in total nerve fiber distribution and number (PGP 9.5) or VIP immunoreactivity, nor in CGRP-immunoreactive epithelial endocrine cells. It is concluded that the rat tracheal peptidergic nerves are a sensitive indicator of inhaled irritant substances. Their reduced immunoreactivity may be because of a release of sensory neuropeptides that could play a role in the physiological response to irritant or toxic compounds

    Adaptation strategies and approaches for forested watersheds

    Intentional climate adaptation planning for ecosystems has become a necessary part of the job for natural resource managers and natural resource professionals in this era of non-stationarity. One of the major challenges in adapting ecosystems to climate change is in the translation of broad adaptation concepts to specific, tangible actions. Addressing management goals and values while considering the long-term risks associated with local climate change can make forested watershed management plans more robust to uncertainty and changing conditions. We provide a menu of tiered adaptation strategies, which we developed with a focus on forests of the Midwest and Northeastern U.S., as part of a flexible framework to support the integration of climate change considerations into forested watershed management and conservation activities. This menu encapsulates ideas from the literature into statements that signify climate adaptation intention and provide examples of associated tactics to help ground the concepts in specific actions. Finally, we describe two demonstration projects, shared through the Northern Institute of Applied Climate Science’s Climate Change Response Framework, that have used this Forested Watershed Adaptation Menu and Adaptation Workbook in project-level planning

    The use of rapid screening tests to compare changes during malting in sorghum

    Eight sorghum cultivars were malted and samples were taken after steeping and after each of 5 days germination. In genotypes with low levels of grain nitrogen, up to 35% of the nitrogen solubilisation during the malting process occurred in the steeping phase. After 5 days germination, extracts ranged from very low to moderate levels and correlated highly with diastase and half-grain mash results. In addition, genotypes producing higher extracts showed greater reductions in milling energy between 2 and 5 days germination. During malting, there was considerable genotype X day interaction for several quality characters, suggesting variation in rates of development as well as final quantitie

    Beyond planning tools: Experiential learning in climate adaptation planning and practices

    In the past decade, several dedicated tools have been developed to help natural resources professionals integrate climate science into their planning and implementation; however, it is unclear how often these tools lead to on-the-ground climate adaptation. Here, we describe a training approach that we developed to help managers effectively plan to execute intentional, climate-informed actions. This training approach was developed through the Climate Change Response Framework (CCRF) and uses active and focused work time and peer-to-peer interaction to overcome observed barriers to using adaptation planning tools. We evaluate the effectiveness of this approach by examining participant evaluations and outlining the progress of natural resources projects that have participated in our trainings. We outline a case study that describes how this training approach can lead to place and context-based climate-informed action. Finally, we describe best practices based on our experience for engaging natural resources professionals and helping them increase their comfort with climate-informed planning

    Methicillin sensitive Staphylococcus aureus producing Panton-Valentine leukocidin toxin in Trinidad & Tobago: a case report

    <p>Abstract</p> <p>Introduction</p> <p>Certain <it>Staphylococcus aureus </it>strains produce Panton-Valentine leukocidin, a toxin that lyses white blood cells causing extensive tissue necrosis and chronic, recurrent or severe infection. This report documents a confirmed case of methicillin-sensitive <it>Staphylococcus aureus </it>strain harboring Panton-Valentine leukocidin genes from Trinidad and Tobago. To the best of our knowledge, this is the first time that such a case has been identified and reported from this country.</p> <p>Case presentation</p> <p>A 13-year-old Trinidadian boy of African descent presented with upper respiratory symptoms and gastroenteritis-like syptoms. About two weeks later he was re-admitted to our hospital complaining of pain and weakness affecting his left leg, where he had received an intramuscular injection of an anti-emetic drug. He deteriorated and developed septic arthritis, necrotizing fasciitis and septic shock with acute respiratory distress syndrome, leading to death within 48 hours of admission despite intensive care treatment. The infection was caused by <it>S. aureus</it>. Bacterial isolates from specimens recovered from our patient before and after his death were analyzed using microarray DNA analysis and <it>spa </it>typing, and the results revealed that the <it>S. aureus </it>isolates belonged to clonal complex 8, were methicillin-susceptible and positive for Panton-Valentine leukocidin. An autopsy revealed multi-organ failure and histological tissue stains of several organs were also performed and showed involvement of his lungs, liver, kidneys and thymus, which showed Hassal's corpuscles.</p> <p>Conclusion</p> <p>Rapid identification of Panton-Valentine leukocidin in methicillin-sensitive <it>S. aureus </it>isolates causing severe infections is necessary so as not to miss their potentially devastating consequences. Early feedback from the clinical laboratories is crucial.</p

    Association mapping of malting quality traits in UK spring and winter barley cultivar collections

    Key Message: Historical malting quality data was collated from UK national and recommended list trial data and used in a GWAS. 25 QTL were identified, with the majority from spring barley cultivar sets. Abstract: In Europe, the most economically significant use of barley is the production of malt for use in the brewing and distilling industries. As such, selection for traits related to malting quality is of great commercial interest. In order to study the genetic basis of variation for malting quality traits in UK cultivars, a historical set of trial data was collated from national and recommended list trials from the period 1988 to 2016. This data was used to estimate variety means for 20 quality related traits in 451 spring barley cultivars, and 407 winter cultivars. Genotypes for these cultivars were generated using iSelect 9k and 50k genotyping platforms, and a genome wide association scan performed to identify malting quality quantitative trait loci (QTL). 24 QTL were identified in spring barley cultivars, and 2 from the winter set. A number of these correspond to known malting quality related genes but the remainder represents novel genetic variation that is accessible to breeders for the genetic improvement of new cultivars.Mark E.
