    Soil mineral nitrogen benefits derived from legumes and comparisons of the apparent recovery of legume or fertiliser nitrogen by wheat

    Nitrogen (N) contributed by legumes is an important component of N supply to subsequent cereal crops, yet few Australian grain-growers routinely monitor soil mineral N before applying N fertiliser. Soil and crop N data from 16 dryland experiments conducted in eastern Australia from 1989–2016 were examined to explore the possibility of developing simple predictive relationships to assist farmer decision-making. In each experiment, legume crops were harvested for grain or brown-manured (BM, terminated before maturity with herbicide), and wheat, barley or canola were grown. Soil mineral N measured immediately before sowing wheat in the following year was significantly higher (P < 0.05) after 31 of the 33 legume pre-cropping treatments than adjacent non-legume controls. The average improvements in soil mineral N were greater for legume BM (60 ± 16 kg N/ha; n = 5) than grain crops (35 ± 20 kg N/ha; n = 26), but soil N benefits were similar when expressed on the basis of summer fallow rainfall (0.15 ± 0.09 kg N/ha per mm), residual legume shoot dry matter (9 ± 5 kg N/ha per t/ha), or total legume residue N (28 ± 11%). Legume grain crops increased soil mineral N by 18 ± 9 kg N/ha per t/ha grain harvested. Apparent recovery of legume residue N by wheat averaged 30 ± 10% for 20 legume treatments in a subset of eight experiments. Apparent recovery of fertiliser N in the absence of legumes in two of these experiments was 64 ± 16% of the 51–75 kg fertiliser-N/ha supplied. The 25 year dataset provided new insights into the expected availability of soil mineral N after legumes and the relative value of legume N to a following wheat crop, which can guide farmer decisions regarding N fertiliser use.Mark B.     In 2007, the International Knockout Mouse Consortium (IKMC) made the ambitious promise to generate mutations in virtually every protein-coding gene of the mouse genome in a concerted worldwide action. Now, 5 years later, the IKMC members have developed highthroughput gene trapping and, in particular, gene-targeting pipelines and generated more than 17,400 mutant murine embryonic stem (ES) cell clones and more than 1,700 mutant mouse strains, most of them conditional. A common IKMC web portal (www.knockoutmouse.org) has been established, allowing easy access to this unparalleled biological resource. The IKMC materials considerably enhance functional gene annotation of the mammalian genome and will have a major impact on future biomedical research

    Irbesartan in Marfan syndrome (AIMS): a double-blind, placebo-controlled randomised trial

    BACKGROUND: Irbesartan, a long acting selective angiotensin-1 receptor inhibitor, in Marfan syndrome might reduce aortic dilatation, which is associated with dissection and rupture. We aimed to determine the effects of irbesartan on the rate of aortic dilatation in children and adults with Marfan syndrome. METHODS: We did a placebo-controlled, double-blind randomised trial at 22 centres in the UK. Individuals aged 6-40 years with clinically confirmed Marfan syndrome were eligible for inclusion. Study participants were all given 75 mg open label irbesartan once daily, then randomly assigned to 150 mg of irbesartan (increased to 300 mg as tolerated) or matching placebo. Aortic diameter was measured by echocardiography at baseline and then annually. All images were analysed by a core laboratory blinded to treatment allocation. The primary endpoint was the rate of aortic root dilatation. This trial is registered with ISRCTN, number ISRCTN90011794. FINDINGS: Between March 14, 2012, and May 1, 2015, 192 participants were recruited and randomly assigned to irbesartan (n=104) or placebo (n=88), and all were followed for up to 5 years. Median age at recruitment was 18 years (IQR 12-28), 99 (52%) were female, mean blood pressure was 110/65 mm Hg (SDs 16 and 12), and 108 (56%) were taking β blockers. Mean baseline aortic root diameter was 34·4 mm in the irbesartan group (SD 5·8) and placebo group (5·5). The mean rate of aortic root dilatation was 0·53 mm per year (95% CI 0·39 to 0·67) in the irbesartan group compared with 0·74 mm per year (0·60 to 0·89) in the placebo group, with a difference in means of -0·22 mm per year (-0·41 to -0·02, p=0·030). The rate of change in aortic Z score was also reduced by irbesartan (difference in means -0·10 per year, 95% CI -0·19 to -0·01, p=0·035). Irbesartan was well tolerated with no observed differences in rates of serious adverse events. INTERPRETATION: Irbesartan is associated with a reduction in the rate of aortic dilatation in children and young adults with Marfan syndrome and could reduce the incidence of aortic complications

Limulus appear to be determinate spawners, maturing all their eggs for the breeding season before spawning begins. On average, larger females held a larger number of eggs (63,500) than smaller females (14,500). By the end of the breeding season there was an average of 11,600 mature eggs female-1 left undeposited, regardless of female size. Larger females laid a higher percentage of the eggs they contained. Thus they not only contain more eggs, but are more effective at laying them as well. Size of spawning females ranged from about 185-300 mm prosomal width, with by far the highest concentration in the mid-size ranges. Although on an individual basis large females carry and lay the greatest number of eggs, mid-size crabs as a group contributed more to the horseshoe crab population in Pleasant Bay because they were more plentiful (net fecundity was highest for mid-size crabs). These results have implications for the management of this important species, which is harvested for bait, scientific, and biomedical uses. Incorporation of these results into models and other management tools can help predict growth rates, effects of size-selective harvest, reproductive value, and stable stage distribution of populations.This project was partially funded by MIT Sea Grant 8247-5

    Klossiella quimrensis (Apicomplexa: Klossiellibae) causes renal coccidiosis in western barred bandicoots Perameles bougainville (Marsupialia: Peramelidae) in Western Australia

    Previous studies have described a range of Klossiella species parasitic in marsupial hosts. Klossiella quimrensis is the etiologic agent of renal coccidiosis in the peramelid marsupial hosts Isoodon obesulus and Perameles gunnii in Eastern Australia, but there is no previous report of klossiellosis in Western Australian peramelids. This study describes klossiellosis diagnosed by histology of renal tissue sections collected during necropsy of 20 Perameles bougainville between 2000 and 2005. Sporonts, sporoblasts, and macrogametes were identified within parasitophorous vacuoles of epithelial cells located near the renal corticomedullary junction. The prevalence of renal coccidiosis in P. bougainville diagnosed by renal histology is estimated at 30%. Only a single unsporulated sporocyst was detected by examination of cystocentesis-collected urine, indicating that microscopic evaluation of urine samples is an insensitive diagnostic test for detection of K. quimrensis in P. bougainville. This infection in P. bougainville is indirectly associated with mild multifocal interstitial lymphohistiocytic nephritis and is likely to be only minimally pathogenic in otherwise healthy individuals. Our study also extends the host and geographic range of K. quimrensis to include P. bougainville and Western Australia

    Cutaneous papillomatosis and carcinomatosis in the highly endangered Western Barred Bandicoot

    The Western Barred Bandicoot (WBB) is a highly endangered marsupial, only existing in the wild in Western Australia. Conservation efforts using captive breeding programmes to prevent the extinction of the WBB are being hampered by a progressively debilitating, wart-like syndrome in captive and wild WBB. Multicentric proliferative lesions have been observed predominately over the face and feet of captive and wild populations of WBB. Introduction of apparently healthy, wild WBB into captive colonies of WBB affected with the skin lesions has been associated with the subsequent development of the skin lesions in the introduced bandicoots. Grossly and histologically, the smaller skin lesions resemble papillomas, whereas the larger lesions demonstrate malignant transformation into carcinomas. Chlamydial organisms have been detected in association with the lesions; however, their role in the pathogenesis of the lesions is unknown. Rarely, large intranuclear inclusion bodies have been observed in proliferative epithelium under light microscopy, suggesting a viral aetiology. Papilloma, polyoma and herpes viruses have all been reported in association with neoplastic transformation of cutaneous epithelial structures in a variety of vertebrate species. Preliminary examination of skin lesions from five WBB has utilized light microscopy, transmission electron microscopy, indirect immunohistochemistry for papillomavirus, and PCR tests for murine polyoma virus, human papilloma virus and conserved herpes virus sequences. None of these tests has definitively identified an aetiological agent. Further research is being undertaken using degenerate PCR primers targeting highly conserved regions of genes from papilloma, polyoma and herpes viruses to further investigate the possibility of a viral aetiology
