A Nonlinear Lagrange Algorithm for Stochastic Minimax Problems Based on Sample Average Approximation Method
An implementable nonlinear Lagrange algorithm for stochastic minimax problems is presented based on sample average approximation method in this paper, in which the second step minimizes a nonlinear Lagrange function with sample average approximation functions of original functions and the sample average approximation of the Lagrange multiplier is adopted. Under a set of mild assumptions, it is proven that the sequences of solution and multiplier obtained by the proposed algorithm converge to the Kuhn-Tucker pair of the original problem with probability one as the sample size increases. At last, the numerical experiments for five test examples are performed and the numerical results indicate that the algorithm is promising
An Implementable SAA Nonlinear Lagrange Algorithm for Constrained Minimax Stochastic Optimization Problems
This paper proposes an implementable SAA (sample average approximation) nonlinear Lagrange algorithm for the constrained minimax stochastic optimization problem based on the sample average approximation method. A computable nonlinear Lagrange function with sample average approximation functions of original functions is minimized and the Lagrange multiplier is updated based on the sample average approximation functions of original functions in the algorithm. And it is shown that the solution sequences obtained by the novel algorithm for solving subproblem converge to their true counterparts with probability one as the sample size approximates infinity under some moderate assumptions. Finally, numerical experiments are carried out for solving some typical test problems and the obtained numerical results preliminarily demonstrate that the proposed algorithm is promising
Evaluation and Error Correction of the ECMWF Subseasonal Precipitation Forecast over Eastern China during Summer
Subseasonal-to-seasonal (S2S) prediction is a highly regarded skill around the world. To improve the S2S forecast skill, an S2S prediction project and an extensive database have been established. In this study, the European Center for Medium-Range Weather Forecasts (ECMWF) model hindcast, which participates in the S2S prediction project, is systematically assessed by focusing on the hindcast quality for the summer accumulated ten-day precipitation at lead times of 0–30 days during 1995–2014 in eastern China. Additionally, the hindcast error is corrected by utilizing the preceding sea surface temperature (SST). The metrics employed to measure the ECMWF hindcast performance indicate that the ECMWF model performance drops as the lead time increases and exhibits strong interannual differences among the five subregions of eastern China. In addition, the precipitation forecast skill of the ECMWF hindcast is best at approximately 15 days in some areas of Southeast China; after correcting the forecast error, the forecast skill is increased to 30 days. At lead times of 0–30 days, regardless of whether the forecast error is corrected, the root mean square errors are lowest in Northeast China. After correcting the forecast error, the performance of the ECMWF hindcast shows better improvement in depicting the quantity and temporal and spatial variation of precipitation at lead times of 0–30 days in eastern China. The false alarm ratio (FAR), probability of detection (POD), and equitable threat score (ETS) reveal that the ECMWF model has a preferable performance at forecasting accumulated ten-day precipitation rates of approximately 20∼50 mm and indicates an improved hindcast quality after the forecast error correction. In short, adopting the preceding SST to correct the summer subseasonal precipitation of the ECMWF hindcast is preferable
Bichirs employ similar genetic pathways for limb regeneration as are used in lungfish and salamanders
Bichirs are a sister group to sarcopterygian and tetrapods that can fully regenerate their endochondral-skeleton-fins. Histological and transcriptomic comparison approaches have been used to investigate the morphology and genetic basis of bichir lobe-fin regeneration, with strong down-regulation of muscle-related genes and up-regulation of ECM-related genes and developmental genes being observed. Bichir limb regeneration involves similar cellular processes to those employed by lungfish and salamander, with MARCKS-like protein (MLP) that is known to be a putative regeneration-initiating molecule in salamander, also up-regulated in the early stages of bichir lobe-fin regeneration. These gene expression results suggest that limb regeneration pathways in these amphibians have a common ancestral inheritance, consistent with evolution from endochondral-skeleton-fin structures to endochondral-skeleton-limb structures of vertebrates
Comparative iTRAQ proteomics revealed proteins associated with lobed fin regeneration in Bichirs
AbstractBackgroundPolypterus senegalus can fully regenerate its pectoral lobed fins, including a complex endoskeleton, with remarkable precision. However, despite the enormous potential of this species for use in medical research, its regeneration mechanisms remain largely unknown.MethodsTo identify the differentially expressed proteins (DEPs) during the early stages of lobed fin regeneration in P. senegalus, we performed a differential proteomic analysis using isobaric tag for relative and absolute quantitation (iTRAQ) approach based quantitative proteome from the pectoral lobed fins at 3 time points. Furthermore, we validated the changes in protein expression with multiple-reaction monitoring (MRM) analysis.ResultsThe experiment yielded a total of 3177 proteins and 15,091 unique peptides including 1006 non-redundant (nr) DEPs. Of these, 592 were upregulated while 349 were downregulated after lobed fin amputation when compared to the original tissue. Bioinformatics analyses showed that the DEPs were mainly associated with Ribosome and RNA transport, metabolic, ECM-receptor interaction, Golgi and endoplasmic reticulum, DNA replication, and Regulation of actin cytoskeleton.ConclusionsTo our knowledge, this is the first proteomic research to investigate alterations in protein levels and affected pathways in bichirs’ lobe-fin/limb regeneration. In addition, our study demonstrated a highly dynamic regulation during lobed fin regeneration in P. senegalus. These results not only provide a comprehensive dataset on differentially expressed proteins during the early stages of lobe-fin/limb regeneration but also advance our understanding of the molecular mechanisms underlying lobe-fin/limb regeneration.</p
A Comparative Evaluation of eDNA Metabarcoding Primers in Fish Community Monitoring in the East Lake
East Lake in Wuhan, China, harbors a high number of freshwater fish species of great conservation value, concurrently serving as vital resources for local livelihoods. However, the ecosystem is threatened by an array of anthropogenic activities, thus requiring consistent monitoring of the local fish community to enable more efficacious conservation management. In place of conventional surveying methods, we undertook the first analysis of the fish distribution within East Lake via metabarcoding of environmental DNA (eDNA). The accuracy and efficacy of eDNA metabarcoding rely heavily upon selecting an appropriate primer set for PCR amplification. Given the varying environmental conditions and taxonomic diversity across distinct study systems, it remains a challenge to propose an optimal genetic marker for universal use. Thus, it becomes necessary to select PCR primers suitable for the composition of fish in the East Lake. Here, we evaluated the performance of two primer sets, Mifish-U and Metafish, designed to amplify 12S rRNA barcoding genes in fishes. Our results detected a total of 116 taxonomic units and 51 fish species, with beta diversity analysis indicating significant differences in community structure diversity between the six sampling locations encompassing East Lake. While it was difficult to accurately compare the species-level discriminatory power and amplification bias of the two primers, Mifish outperformed Metafish in terms of taxonomic specificity for fish taxa and reproducibility. These findings will assist with primer selection for eDNA-based fish monitoring and biodiversity conservation in the East Lake and other freshwater ecosystems
Development of DNA aptamers targeting low-molecular-weight amyloid-β peptide aggregates in vitro
We have developed a novel functional nucleic acid aptamer to amyloid- peptide 1-40 (A(1-40)) and investigated its potential to detect A peptide fragments in neuropathologically confirmed Alzheimer brain hippocampus tissues samples. Our results demonstrate that the aptamer candidate RNV95 could detect tetrameric/pentameric low-molecular-weight A aggregates in autopsy hippocampal tissue from two neuropathologically confirmed Alzheimer disease cases. Although these are preliminary observations, detailed investigations are under way. This is the first demonstration of aptamer-A binding in Alzheimer brain tissues
Tdrd12 Is Essential for Germ Cell Development and Maintenance in Zebrafish
The regularity of Piwi-interacting RNA (piRNA) biogenesis is crucial to germline development. Functioning as Piwi-interacting proteins, Tudor domain-related proteins (Tdrds) have been demonstrated to be involved in spermatogenesis and the piRNA pathway. In this study, zebrafish tdrd12 was identified, and the maternal and germ cell-specific expression patterns of zebrafish tdrd12 were observed. Utilizing TALEN (transcription activator-like effector nuclease) techniques, two independent tdrd12 mutant zebrafish lines were generated. Although no defects were found during the generation of the primordial germ cells (PGCs) in the tdrd12-null fish progenies obtained from the heterozygous tdrd12 mutant parents, all Tdrd12-deficient fish developed into infertile males. The reduced numbers and eventually loss of the germ cells by 35 days post fertilization (dpf) led to masculinization and infertility of the Tdrd12-deficient fish. Meiosis defects of the germ cells in the tdrd12 mutants during the gonad-transitioning period were observed, revealing the indispensable functions of Tdrd12 in gametogenesis.
