    Nogo-A regulates myogenesis via interacting with Filamin-C

    Among the three isoforms encoded by Rtn4, Nogo-A has been intensely investigated as a central nervous system inhibitor. Although Nogo-A expression is increased in muscles of patients with amyotrophic lateral sclerosis, its role in muscle homeostasis and regeneration is not well elucidated. In this study, we discovered a significant increase in Nogo-A expression in various muscle-related pathological conditions. Nogo−/− mice displayed dystrophic muscle structure, dysregulated muscle regeneration following injury, and altered gene expression involving lipid storage and muscle cell differentiation. We hypothesized that increased Nogo-A levels might regulate muscle regeneration. Differentiating myoblasts exhibited Nogo-A upregulation and silencing Nogo-A abrogated myoblast differentiation. Nogo-A interacted with filamin-C, suggesting a role for Nogo-A in cytoskeletal arrangement during myogenesis. In conclusion, Nogo-A maintains muscle homeostasis and integrity, and pathologically altered Nogo-A expression mediates muscle regeneration, suggesting Nogo-A as a novel target for the treatment of myopathies in clinical settings. © 2021, The Author(s).1

    The Effects of Mitigation Measures on Flood Damage Prevention in Korea

    This study analyzed the characteristics of flood damages and the effects of structural and non-structural flood damage mitigation measures in Korea. First, a theoretical discussion of the structural and non-structural measures to mitigate flood damages was used to select the variables and devise the hypotheses. An analysis was conducted using the Auto-Regressive Integrated Moving-Average (ARIMA) time series methodology, Korean socioeconomic data, and damage characteristics of major flood events. The effects of flood damage mitigation measures on the extent of flood damages were assessed using an intervention time series model. The major findings were that the intervention effects of structural and non-structural measures were statistically significant from 1958 to 2013 (a period of 55 years) and that while the former were ineffective at mitigating flood damages, the latter were successful in doing so. Based on the above findings, policy suggestions for future flood damage mitigation measures in Korea were offered. For structural measures, the government should manage its existing facilities, recover ecosystems of damaged rivers, and devise mitigation measures for urban areas. For non-structural measures, the government should enhance its flood forecasting capacity, revise laws related to flood control and prevention, and update and rationalize land-use plans

    Perianal extragastrointestinal stromal tumor

    An extragastrointestinal stromal tumor (EGIST) is a gastrointestinal stromal tumor that arises outside of the gastrointestinal tract. Most EGISTs are located in the omentum, mesentery, and retroperitoneum. The occurrence of an EGIST at the perianal region is very rare. Herein, we report our experience with EGISTs in the perianal area and review the literature. A 70-year-old man presented to our hospital with a 2-year history of anal discomfort. A pelvic magnetic resonance imaging scan showed a homogenous, well-defined, soft tissue density mass. The patient underwent mass excision, and the pathological examination confirmed that the mass was an EGIST. The size of the tumor was 4.3×3.2 cm, and the mitotic count was 1 per 50 high-power fields. The tumor cells were immunohistochemically positive for KIT and CD34 but were negative for S-100 and alpha-smooth muscle actin. There were no other abnormal findings in the gastrointestinal tract; upon pathological review, this case was confirmed as perianal EGIST. Therefore, EGIST should be considered as a differential diagnosis of perianal masses

    How Can Insulin Resistance Cause Alzheimer’s Disease?

    Alzheimer’s disease (AD) is a neurodegenerative disorder associated with cognitive decline. Despite worldwide efforts to find a cure, no proper treatment has been developed yet, and the only effective countermeasure is to prevent the disease progression by early diagnosis. The reason why new drug candidates fail to show therapeutic effects in clinical studies may be due to misunderstanding the cause of AD. Regarding the cause of AD, the most widely known is the amyloid cascade hypothesis, in which the deposition of amyloid beta and hyperphosphorylated tau is the cause. However, many new hypotheses were suggested. Among them, based on preclinical and clinical evidence supporting a connection between AD and diabetes, insulin resistance has been pointed out as an important factor in the development of AD. Therefore, by reviewing the pathophysiological background of brain metabolic insufficiency and insulin insufficiency leading to AD pathology, we will discuss how can insulin resistance cause AD

    Improving the Electrical Connection of n-Type Conjugated Polymers through Fluorine-Induced Robust Aggregation

    Naphthalene diimide (NDI)-based conjugated polymers with bithiophene or dithienylethene (TVT) units can form large crystal domains through NDI-driven self-assembly and are widely used in organic electronic devices as n-type materials. However, improving electron transport in these semiconducting polymers has been a significant challenge mainly due to poor electrical connections between the crystal domains. Formation of an interconnected network of small domains with short-range ordering and mixed orientations could be an effective way of increasing the electron mobility (??e) of polymeric materials. The present study demonstrates the feasibility of this approach using an NDI-based polymer composed of fluorinated TVT (FTVT) units. The FTVT unit enhances intermolecular interactions between the polymer chains, leading to robust aggregation. This aggregation was found to suppress NDI-driven self-assembly, resulting in interconnected small crystal domains with short-range ordering and good thermal stability at elevated temperatures. This microstructure provided transistor devices with improved ??e by lowering energetic disorder as well as consistent electrical connectivity at different annealing temperatures
