    This thesis discusses government policy on tourism Annual Cap Go Meh Festival as tourism effort under Article 14 Paragraph (1) Jo. Article 23 of Law No. 10 Year 2009 on Tourism in Singkawang. From the results of this thesis research we concluded that Local government policy on tourism annual festival Cap Go Meh in building and developing the tourism associated with the regional administration is conducting an analysis of the potential type of objects and tourist attraction that can support the organization of the festival Cap Go Meh in Singkawang. Natural tourism potential is a potential dominant object owned Singkawang, followed by the potential of religious tourism, culture and artificial. However, until now the government Singkawang has not yet prepared Tourism Development Master Plan, but the government Singkawang remains to develop and encourage the addition of a variety of attractions that can support the organizing festival Cap Go Meh thus able to act as the development of foreign national tourism and to support tourism businesses area in an effort to improve the local economy for the PAD, to develop their culture and preserve the environment and natural resources, as well as the nature of produce for progress. Factors faced by the government of Singkawang on tourism Annual Cap Go Meh Festival in the development of tourism in Singkawang namely the realization of development of tourism potential in Singkawang there are some things that factor into the supporting and inhibiting factors. The driving factor in the development of tourism potential that strong commitment, the structure of the government institutions, potential and diversity of natural resources and cultural diversity. There is no or only POor linear correlation between MgO and incompatible elements. Whereas a bit remarked correlations are observed between MgO and most compatible elements.The geochemical characteristics suggest that the strong and multiple mixing processes may have Occurred universally both in the lower crust and crust-mantle transitional zone. The weighed average composition of xenoliths shows a more mafic composition of the lower crust than that estimated from granulite data, which may imply the difference in the genesis.published_or_final_versio

    Sr, Nd Pb isotopic characteristics of granulite and pyroxenite xenoliths in Hannuoba basalts, Hebei Province, and their implications for geologic processes

    Rb-Sr, Sm-Nd and Pb isotopic systematics are reported for granulite and pyroxenite xenoliths in Hannuoba basalts, Hebei Province. The isotopic characteristics of xenoliths suggest t hat they might have formed recently , although there is no strict constraint on the precise age. The decoupling of low Rb/Sr and high [87]Sr/[86]Sr ratios implies the multi-stage evolution history and strong mixing processes. There are close ties between granulite and pyroxenites with different mineral assemblages on the Sr-Nd, Pb-Sr and Pb-Nd correlation figures. The granulite is the common end-member o f the three kinds o f pyroxenite.     Development of stable marker-free nuclear transformation strategy in the green microalga Chlorella vulgaris

    Although microalgae have valuable resources with great necessity in many biotechnological applications, no tools have been developed yet for a stable genetic transformation without antibiotic marker genes in these organisms. Chlorella is one of the most useful genus for biotechnological applications. The transfer of foreign DNA (vector or linear DNA cassette) into Chlorella by electroporation has very low stability and it is hard to screen the transformants without antibiotic marker genes. However, the marker genes have some disadvantages to host cells. To avoid the negative effects caused by the marker genes, we tried to develop a stable marker-free nuclear transformation system in Chlorella. For this, linear gene expression cassettes (LGEC) were constructed with functional domains, which are responsible for transformation, of SV40 large T antigen. The LGECs were transferred into Chlorella via electroporation and durability of the LGECs were confirmed in transgenic Chlorella. Transcription levels of the LGECs were also determined at different cell cycle sates. The LGECs integrated into the chromosomal DNA of Chlorella were stably replicated and were expressed successfully at G0-, G1-, and G2-phases.     Maximum age limitation in luminescence dating of Chinese loess using the multiple-aliquot MET-pIRIR signals from K-feldspar

    Numerical dating of loess is important for Quaternary studies. Recent progress in post-infrared infrared-stimulated luminescence (pIRIR) signals from potassium-rich feldspar has allowed successful dating of Chinese loess beyond the conventional dating limit based on quartz optically stimulated luminescence (OSL) signals. In this study we tested the multiple-aliquot regenerative-dose (MAR) pre-dose multiple-elevated-temperature post-IR IRSL (pMET-pIRIR) procedure on samples from the palaeosol S5 (∼480 ka) and S8 (∼780 ka) layers from the Luochuan and Jingbian sections, respectively. The results show that (1) compared to sensitivity-corrected signal (Lx/Tx), a higher saturation dose is observed for the sensitivity-uncorrected MET-pIRIR signals (Lx), suggesting that MAR is advantageous for dating old samples; (2) negligible fading component can be achieved using the pMET-pIRIR procedure; (3) for the sample from the top of palaeosol S5, De values (1360 + 226/-167 Gy) broadly consistent with expected De (1550 ± 72 Gy) can be obtained using the sensitivity-uncorrected 300 °C MET-pIRIR signal. Our study suggests that a De value of about 1800 Gy may be the maximum dating limit of Chinese loess using the MAR pMET-pIRIR procedure.postprin

    The technology of large-scale pharmaceutical plasmid purification by cetyltrimethylammonium bromide and Tritonx-114

    Many methods for plasmid purification have been developed, and the whole process must be designed to remove the host RNA, protein, genomic DNA and endotoxin. Currently, plasmid is mostly purified by time-consuming chromatographies. As an alternative, a new plasmid purification technology with cetyltrimethylammonium bromide (CTAB) is described. After lysis with alkali, the CTAB was directly titrated into the supernatant for plasmid precipitation, then the coprecipitated pellets were dealt with 3 M KAc and TritonX-114. Quality detection showed that the purified plasmids were free from the contamination of host RNA. In 1 mg purified plasmid, the bacterial genomic DNA, host endotoxin and protein were less than 10 g/ mg, 50 EU/ mg and 10 g/mg, respectively. The ratio of OD260/OD280 was between 1.75 - 1.85, more than 90% of the prepared plasmid presented in the supercoiled form. Further test demonstrated that the pcDNAlacZ purified with CTAB and authoritative endotoxin-free plasmid Kit had the similar transfection efficiency in vivo and in vitro. CTAB can be used for plasmid purification; the main advantages of the DNAs purified with CTAB include the avoidance of animal-derived enzymes, toxic substance like chloroform and phenol. More attractive is that the whole process has the predominance of low cost
