    Circ_0000284 facilitates the growth, metastasis and glycolysis of intrahepatic cholangiocarcinoma through miR-152-3p-mediated PDK1 expression

    Background. Circular RNAs (circRNAs) are key molecules in the regulation of intrahepatic cholangiocarcinoma (ICC) progression. The purpose of this study was to analyze the function and underlying molecular mechanism of circ_0000284 in ICC. Methods. Quantitative real-time PCR was used to analyze the circ_0000284, microRNA (miR)-152-3p and pyruvate dehydrogenase kinase 1 (PDK1) expression. Cell proliferation, apoptosis, invasion and migration were executed by cell counting kit 8 assay, EdU assay, flow cytometry, transwell assay and wound healing assay, respectively. All protein expression levels were examined using western blot analysis. Cell glycolysis was analyzed by detecting glucose consumption, lactate production and ATP/ADP ratios. Target relationship was estimated by dual-luciferase reporter assay. The effect of circ_0000284 on ICC tumor growth in vivo was evaluated by constructing xenograft mice model. Results. We detected high expression of circ_0000284 in ICC tumor tissues and cells. Downregulated circ_0000284 inhibited ICC cell proliferation, invasion, migration, glycolysis, and accelerated apoptosis. MiR-152-3p was sponged by circ_0000284, and its inhibitor revoked the effect of circ_0000284 knockdown on ICC cell progression. PDK1 was a target of miR-152-3p, and its expression was suppressed by circ_0000284 knockdown. PDK1 overexpression reversed the inhibition effect of miR-152-3p on ICC cell growth, metastasis and glycolysis. In animal experiments, circ_0000284 downregulation also inhibited ICC tumor growth. Conclusion. Circ_0000284 promoted the growth, metastasis and glycolysis of ICC by miR-152-3p/PDK1 pathway, showing that circ_0000284 was a potential therapeutic target for ICC

    The distribution and demography of Euphausia superba in Prydz Bay during the austral summer 2002

    This study documents the horizontal distribution and demography of Antarctic krill (Euphausia superba Dana), collected using trawls, from the Prydz Bay region during January 2002. Euphausia superba (E. superba) was distributed primarily south of 64°S. The average density and biomass were estimated as 68.85 ind·(1 000 m-3) and 24.16 g wet weight·(1 000 m-3), respectively. Highest values were located in the open sea (depth > 3 000 m). Body length of E. superba ranged from 30 to 55 mm (N=1 758), with a mean length of 38.45±3.68 mm (SD). The overall sex ratio was approximately 1∶1, 47.6% females (41.9% sub-adults and 5.7% adults), 46.6% males (42.7% sub-adults and 3.9% adults), while 5.8% were juveniles. The population structure of E. superba exhibited geographical variation. At stations in the western part of the survey area populations had a high proportion of juveniles, a lower sexual maturity stage for males and small body size, while the opposite was found at stations in the eastern part. A latitudinal difference was found at stations along 70.5°E and 73°E transects: body length was small and the sex ratio was high in regions of high latitude, while the opposite occurred in areas of low latitude

    Single-Molecule Real-Time Transcript Sequencing Identified Flowering Regulatory Genes in Crocus Sativus

    Background: Saffron crocus (Crocus sativus) is a valuable spice with medicinal uses in gynaecopathia and nervous system diseases. Identify flowering regulatory genes plays a vital role in increasing flower numbers, thereby resulting in high saffron yield. Results: Two full length transcriptome gene sets of flowering and non-flowering saffron crocus were established separately using the single-molecule real-time (SMRT) sequencing method. A total of sixteen SMRT cells generated 22.85 GB data and 75,351 full-length saffron crocus unigenes on the PacBio RS II panel and further obtained 79,028 SSRs, 72,603 lncRNAs and 25,400 alternative splicing (AS) events. Using an Illumina RNA-seq platform, an additional fifteen corms with different flower numbers were sequenced. Many differential expression unigenes (DEGs) were screened separately between flowering and matched non-flowering top buds with cold treatment (1677), flowering top buds of 20 g corms and non-flowering top buds of 6 g corms (1086), and flowering and matched nonflowering lateral buds (267). A total of 62 putative flower-related genes that played important roles in vernalization (VRNs), gibberellins (G3OX, G2OX), photoperiod (PHYB, TEM1, PIF4), autonomous (FCA) and age (SPLs) pathways were identified and a schematic representation of the flowering gene regulatory network in saffron crocus was reported for the first time. After validation by real-time qPCR in 30 samples, two novel genes, PB.20221.2 (p = 0.004, r = 0.52) and PB.38952.1 (p = 0.023, r = 0.41), showed significantly higher expression levels in flowering plants. Tissue distribution showed specifically high expression in flower organs and time course expression analysis suggested that the transcripts increasingly accumulated during the flower development period. Conclusions: Full-length transcriptomes of flowering and non-flowering saffron crocus were obtained using a combined NGS short-read and SMRT long-read sequencing approach. This report is the first to describe the flowering gene regulatory network of saffron crocus and establishes a reference full-length transcriptome for future studies on saffron crocus and other Iridaceae plants

    Purine adenosine pathway exists widely in the body metabolism, and is involved in regulating various physiological processes. It is one of the important pathways of environmental regulation in human body. CD73 is essentially a protease that catalyzes further dephosphorylation of extracellular adenine nucleotides, hydrolyzing extracellular AMP to adenosine and phosphate. CD73 is an important part of the adenosine signaling pathway. Studies have shown that CD73-mediated adenosine pathway can convert the inflammatory ATP into the immunosuppressant adenosine. This paper aims to summarize the relevant effects of CD73 in the occurrence, development and prognosis of liver diseases such as viral hepatitis, highlight the important role of CD73 in liver diseases, especially in viral hepatitis such as HBV and HCV, and explore new clinical ideas for future treatment targets of liver diseases

    Rapid determination of illegal additives chrysoidin and malachite green by surface-enhanced Raman scattering with silanized support based substrate

    © 2018 The Author This paper reports accurate use of silanized support based substrate for surface-enhanced Raman spectroscopy (SERS) determination of illegal additives chrysoidin and malachite green. Detection of these illegal additives is very meaningful and the developed method meets this objective in a simple, rapid and cost effective way. Detection limits for chrysoidin and malachite green in aqueous solutions reached 0.001 mg/L and 0.0001 mg/L respectively. Linear relationship between the Raman intensities and chrysoidin concentrations in range of 0.001-0.05 mg/L was found with regression coefficient (R) of 0.994. To investigate the practical application performance of the method, the substrate was applied to detect chrysoidin in Sprite at 0.01 mg/L and malachite green in fish pond water at 0.0001 mg/L. The SERS method could be an alternative approach for monitoring illegal additives and toxic harmful substances

    Endometrial Cytology as a Method to Improve the Accuracy of Diagnosis of Endometrial Cancer: Case Report and Meta-Analysis

    More and more researchers have reported that dilatation and curettage (D&C) or Pipelle had low accuracy, high misdiagnosis, and insufficient rate. Endometrial cytology is often compared with histology and seems to be an efficient method for the diagnosis of endometrial disorders, especially endometrial cancer. We report a case of misdiagnosed endometrial cancer by D&C, but with a positive cytopathological finding. Following that, a meta-analysis including 4,179 patients of endometrial diseases with cyto-histopathological results was performed to assess the value of the endometrial cytological method in endometrial cancer diagnosis. The pooled sensitivity and specificity of the cytological method in detecting endometrial atypical hyperplasia or cancer was 0.91[95% confidence interval (CI) 0.74–0.97] and 0.96 (95% CI 0.90–0.99), respectively. The pooled positive likelihood ratio and negative likelihood ratio was 25.4 (95% CI 8.1–80.1) and 0.10 (95% CI 0.00–0.30), respectively. The diagnostic odds ratio which was usually used to evaluate the diagnostic test performance reached 260 (95% CI 36–1905). So we recommend that D&C and Pipelle are still practical procedures to evaluate the endometrium, cytological examinations should be utilized as an additional endometrial assessment method

    An Efficacious Endometrial Sampler for Screening Endometrial Cancer

    Recently, the research on early detection of precancerous change and endometrial carcinoma has been focusing on minimally invasive procedures for screening. On this basis, we aim to verify the feasibility of endometrial samplers for screening endometrial cancer using Li Brush. We recruited patients undergoing hysterectomy for different diseases from the Inpatient Department of the Department of Obstetrics and Gynecology. Before surgery, endometrial cells were collected by Li Brush. The cytopathologic diagnosis from Li Brush and the histopathologic diagnosis from hysterectomy in the same patient were compared to calculate sensitivity (Se), specificity (Sp), false-negative rate (FNR), false-positive rate (FPR), positive predictive value (PV+) %, and negative predictive value (PV-). The research enrolled 293 women into this self-controlled trial. According to the hypothesis test of paired four lattices, we obtained the following indicators: Se 92.73, Sp 98.15, FNR 7.27, FPR 1.85, PV+92.73, and PV−98.15%.     Study on the distribution, abundance and growth condition of Antarctic Krill (Euphausia superba) in the Prydz Bay region, Antarctica during the Austral summer of 1999/2000

    Antarctic Krill (Euphausia superba) were collected from the Prydz Bay region during the austral summer of 1999/2000. The sample - collection was made at 15 sites in 3 longitudinal transects. Although krill were encountered at 14 sites, the abundance was comparatively low in general. The main component of the krill population was of post-larval stages. The mean numerical and weight density of krill integrated for all sampling stations in the survey area were 16.17 ind. 1000 m(-3) and 12.02 g 1000m(-3), respectively. In the survey area, stations with larger krill density are mainly located in the slope zone, except that the largest sample was collected in the open sea zone. No krill occurred in the only station of the shelf zone during our investigation. Krill collected from the slope zone were under better growth condition than those from the open sea zone, but when the survey area is considered as a whole, the growth condition is normal. The results of the comprehensive analysis of the environmental factors show that the three large sample stations seem to be related with the cold water mass at 75 m depth and the confluence area between the high and low Chl a centers at the depth of 25 m

    HOG-ESRs Face Emotion Recognition Algorithm Based on HOG Feature and ESRs Method

    As we all know, there are many ways to express emotions. Among them, facial emotion recognition, which is widely used in human–computer interaction, psychoanalysis of mental patients, multimedia retrieval, and other fields, is still a challenging task. At present, although convolutional neural network has achieved great success in face emotion recognition algorithms, it has a rising space in effective feature extraction and recognition accuracy. According to a large number of literature studies, histogram of oriented gradient (HOG) can effectively extract face features, and ensemble methods can effectively improve the accuracy and robustness of the algorithm. Therefore, this paper proposes a new algorithm, HOG-ESRs, which improves the traditional ensemble methods to the ensembles with shared representations (ESRs) method, effectively reducing the residual generalization error, and then combining HOG features with ESRs. The experimental results on the FER2013 dataset show that the new algorithm can not only effectively extract features and reduce the residual generalization error, but also improve the accuracy and robustness of the algorithm, the purpose of the study being achieved. The application of HOG-ESRs in facial emotion recognition is helpful to solve the symmetry of edge detection and the deficiency of related methods in an outdoor lighting environment
