Apatite formation on electrochemically modified surface of hafnium metal in simulated body environment
Biomedical application of Hf for hard tissue repair may be expanded if it can be imparted with bone-bonding ability. Apatite formation in the body is required for artificial materials to bond to bone. We have shown that apatite can form on NaOH- and heat-treated Hf in simulated body fluid, although the degree of apatite formation is not high. In this study, we investigated apatite formation on surface-modified Hf metal in simulated body fluid. Hf substrates were soaked in NH4F solution or electrochemically polarized in H2SO4 or NH4F solution, followed by heating at 600°C. The sample soaked or cathodically polarized in NH4F solution had porous structure, and the anodically polarized sample had nanotube aggregates on its surface. F was incorporated into the sample polarized in NH4F solution, but not the sample merely soaked in the solution. NH4F was more effective than H2SO4 in facilitating apatite formation. Remarkably, the sample soaked in NH4F solution had the highest apatite-forming ability. Its porous structure was considered to play a dominant role in apatite formation. Furthermore, there was little correlation between surface energy and apatite formation. Notably, NH4F treatment is more effective than the previously reported NaOH and heat treatments in promoting apatite formation
Contextualized Automatic Speech Recognition with Attention-Based Bias Phrase Boosted Beam Search
End-to-end (E2E) automatic speech recognition (ASR) methods exhibit
remarkable performance. However, since the performance of such methods is
intrinsically linked to the context present in the training data, E2E-ASR
methods do not perform as desired for unseen user contexts (e.g., technical
terms, personal names, and playlists). Thus, E2E-ASR methods must be easily
contextualized by the user or developer. This paper proposes an attention-based
contextual biasing method that can be customized using an editable phrase list
(referred to as a bias list). The proposed method can be trained effectively by
combining a bias phrase index loss and special tokens to detect the bias
phrases in the input speech data. In addition, to improve the contextualization
performance during inference further, we propose a bias phrase boosted (BPB)
beam search algorithm based on the bias phrase index probability. Experimental
results demonstrate that the proposed method consistently improves the word
error rate and the character error rate of the target phrases in the bias list
on both the Librispeech-960 (English) and our in-house (Japanese) dataset,

Photochemical Characterization of a New Heliorhodopsin from the Gram-Negative Eubacterium Bellilinea caldifistulae (BcHeR) and Comparison with Heliorhodopsin-48C12
Many microorganisms express rhodopsins, pigmented membrane proteins capable of absorbing sunlight and harnessing that energy for important biological functions such as ATP synthesis and phototaxis. Microbial rhodopsins that have been discovered to date are categorized as type-1 rhodopsins. Interestingly, researchers have very recently unveiled a new microbial rhodopsin family named the heliorhodopsins, which are phylogenetically distant from type-1 rhodopsins. Among them, only heliorhodopsin-48C12 (HeR-48C12) from a Gram-positive eubacterium has been photochemically characterized [Pushkarev, A., et al. (2018) Nature 558, 595-599]. In this study, we photochemically characterize a purple-colored heliorhodopsin from Gram-negative eubacterium Bellilinea caldifistulae (BcHeR) as a second example and identify which properties are or are not conserved between BcHeR and HeR-48C12. A series of photochemical measurements revealed several conserved properties between them, including a visible absorption spectrum with a maximum at around 550 nm, the lack of ion-transport activity, and the existence of a second-order O-like intermediate during the photocycle that may activate an unidentified biological function. In contrast, as a property that is not conserved, although HeR-48C12 shows the light adaptation state of retinal, BcHeR showed the same retinal configuration under both dark- and light-adapted conditions. These comparisons of photochemical properties between BcHeR and HeR-48C12 are an important first step toward understanding the nature and functional role of heliorhodopsins
A blue-shifted anion channelrhodopsin from the Colpodellida alga Vitrella brassicaformis
Microbial rhodopsins, a family of photoreceptive membrane proteins containing the chromophore retinal, show a variety of light-dependent molecular functions. Channelrhodopsins work as light-gated ion channels and are widely utilized for optogenetics, which is a method for controlling neural activities by light. Since two cation channelrhodopsins were identified from the chlorophyte alga Chlamydomonas reinhardtii, recent advances in genomic research have revealed a wide variety of channelrhodopsins including anion channelrhodopsins (ACRs), describing their highly diversified molecular properties (e.g., spectral sensitivity, kinetics and ion selectivity). Here, we report two channelrhodopsin-like rhodopsins from the Colpodellida alga Vitrella brassicaformis, which are phylogenetically distinct from the known channelrhodopsins. Spectroscopic and electrophysiological analyses indicated that these rhodopsins are green- and blue-sensitive pigments (lambda(max) = similar to 550 and similar to 440 nm) that exhibit light-dependent ion channeling activities. Detailed electrophysiological analysis revealed that one of them works as a monovalent anion (Cl-, Br- and NO3-) channel and we named it V. brassicaformis anion channelrhodopsin-2, VbACR2. Importantly, the absorption maximum of VbACR2 (similar to 440 nm) is blue-shifted among the known ACRs. Thus, we identified the new blue-shifted ACR, which leads to the expansion of the molecular diversity of ACRs
FZD10-targeted α-radioimmunotherapy with 225Ac-labeled OTSA101 achieves complete remission in a synovial sarcoma model
Synovial sarcomas are rare tumors arising in adolescents and young adults. The prognosis for advanced disease is poor, with an overall survival of 12-18 months. Frizzled homolog 10 (FZD10) is overexpressed in most synovial sarcomas, making it a promising therapeutic target. The results of a phase 1 trial of β-radioimmunotherapy (RIT) with the 90Y-labeled anti-FZD10 antibody OTSA101 revealed a need for improved efficacy. The present study evaluated the potential of α-RIT with OTSA101 labeled with the α-emitter 225Ac. Competitive inhibition and cell binding assays showed that specific binding of 225Ac-labeled OTSA101 to SYO-1 synovial sarcoma cells was comparable to that of the imaging agent 111In-labeled OTSA101. Biodistribution studies showed high uptake in SYO-1 tumors and low uptake in normal organs, except for blood. Dosimetric studies showed that the biologically effective dose (BED) of 225Ac-labeled OTSA101 for tumors was 7.8 Bd higher than that of 90Y-labeled OTSA101. 90Y- and 225Ac-labeled OTSA101 decreased tumor volume and prolonged survival. 225Ac-labeled OTSA101 achieved a complete response in 60% of mice, and no recurrence was observed. 225Ac-labeled OTSA101 induced a larger amount of necrosis and apoptosis than 90Y-labeled OTSA101, although the cell proliferation decrease was comparable. The BED for normal organs and tissues was tolerable; no treatment-related mortality or obvious toxicity, except for temporary body weight loss, was observed. 225Ac-labeled OTSA101 provided a high BED for tumors and achieved a 60% complete response in the synovial sarcoma mouse model SYO-1. RIT with 225Ac-labeled OTSA101 is a promising therapeutic option for synovial sarcoma
Effect of fentanyl for preterm infants on mechanical ventilation: a systematic review and meta-analysis
Introduction: Because excessive physical stress is harmful, reducing pain and discomfort in premature neonates during mechanical ventilation is a major challenge for physicians. There are no consensus and systematic review on the use of fentanyl, the most commonly used pain reliever in preterm neonates during mechanical ventilation. We aim to compare the benefits and harms of fentanyl versus placebo or no drug for preterm neonates receiving mechanical ventilation. Methods: A systematic review of randomized controlled trials (RCTs) was conducted according to the Cochrane Handbook for Systematic Reviews of Interventions. The systematic review was reported using the Preferred Reporting Items for Systematic Reviews and Meta-Analyses statement. Scientific databases such as MEDLINE, Embase, CENTRAL, and CINAHL were searched. All preterm infants on mechanical ventilation and enrolled in an RCT of fentanyl versus control were included. Results: Of 256 reports initially retrieved, 4 reports met the eligibility criteria. Fentanyl was not associated with mortality risk compared to the control (risk ratio: 0.72, 95% confidence intervals [CIs]: 0.36–1.44). No increase in ventilation duration (mean difference [MD]: 0.04, 95% CIs: −0.63–0.71) and no effect on hospital stay length (MD: 4.00, 95% CIs: −7.12–15.12) were found. Fentanyl intervention does not affect any other morbidities, including bronchopulmonary dysplasia, periventricular leukomalacia, patent ductus arteriosus, intraventricular hemorrhage (IVH), severe IVH, sepsis, and necrotizing enterocolitis. Conclusion: The present systematic review and meta-analysis failed to demonstrate the benefit of administering fentanyl to preterm infants on mechanical ventilation in mortality and morbidities. Follow-up studies are required to investigate the long-term neurodevelopment of the children
National Examination Measures in the Field of Cognitive Disorder : Analysis of Issue of National Examination for the Past Ten Years of Physical Therapist, Occupational Therapist and Speech-Language-Hearing Therapist
[目的]理学療法士(以下PT)・作業療法士(以下OT)・言語聴覚士(以下ST)の国家試験問題過去10年分から,高次脳機能障害分野に関する問題を分析し,国家試験対策の方法を検討することが目的である.[方法] 2002年から2011年のPT ・ OT ・STの過去10年分の国家試験問題から,高次脳機能障害分野の問題を抽出し,総出題数と最頻値,五肢選択形式,問題の深さ(Taxonomy),高次脳機能障害の分類について分析した.[結果]総出題数はPT41問・ OT70問・ST190問であり,最頻値はPT3問・ OT8問・ST20問であった.問題の深さに関しては,想起レベルがPT27問(66%)・ OT41問(59%)・ ST154問(81%)と最も多く、出題形式は五肢択一形式(以下Aタイプ)がPT31問(76%)・ OT55問(79%)・ST150問(79%)と出題される頻度が高かった.高次脳機能障害の出題分野については, STでは失語症に関する問題が120問(63%)と多かった.[結語]問題の傾向としてはAタイプ,想起レベルが多く,語句の意味や定義について正しい知識を身につけることが,国家試験対策の基本となることが再確認された
A human PSMB11 variant affects thymoproteasome processing and CD8+ T cell production
The Psmb11-encoded β5t subunit of the thymoproteasome, which is specifically expressed in cortical thymic epithelial cells (cTECs), is essential for the optimal positive selection of functionally competent CD8+ T cells in mice. Here, we report that a human genomic PSMB11 variation, which is detectable at an appreciable allele frequency in human populations, alters the β5t amino acid sequence that affects the processing of catalytically active β5t proteins. The introduction of this variation in the mouse genome revealed that the heterozygotes showed reduced β5t expression in cTECs and the homozygotes further exhibited reduction in the cellularity of CD8+ T cells. No severe health problems were noticed in many heterozygous and 5 homozygous human individuals. Long-term analysis of health status, particularly in the homozygotes, is expected to improve our understanding of the role of the thymoproteasome-dependent positive selection of CD8+ T cells in humans
