Critique [of African and Pac!fic Literature: A Comparative Study]
Kristine Martin’s study makes available the relatively little-known literature of the southern Pacific basin islands. Her article has merit because it is compared with the more widely-read African literature, and she makes a significant contribution with the comparisions [comparison]. Both the Pacific selections, a recent phenomena, and the older African works are transitional literatures — striving to connect the colonized past with a post-colonial synthesis which is relevant to the author of the intended audience. Angiographic Findings in Diaphragmatic Rupture
The angiographic findings are presented in four patients with ruptured diaphragms. With rupture of the left hemidiaphragm the findings vary with the structures herniating. Gastric, mesenteric or splenic artery branches extending into the lower chest indicate herniation of the stomach, bowel or spleen. A herniating spleen is frequently ruptured. On the right, the liver is generally the only organ to herniate. Ethanol-induced Modulation of Hepatocellular Extracellular Signal-regulated Kinase-1/2 Activity via 4-Hydroxynonenal
Modulation of the extracellular signal-regulated kinases (ERK-1/2), a signaling pathway directly associated with cell proliferation, survival, and homeostasis, has been implicated in several pathologies, including alcoholic liver disease. However, the underlying mechanism of ethanol-induced ERK-1/2 modulation remains unknown. This investigation explored the effects of ethanol-associated oxidative stress on constitutive hepatic ERK-1/2 activity and assessed the contribution of the lipid peroxidation product 4-hydroxynonenal (4-HNE) to the observations made in vivo. Constitutive ERK-1/2 phosphorylation was suppressed in hepatocytes isolated from rats chronically consuming ethanol for 45 days. This observation was associated with an increase in 4-HNE-ERK monomer adduct concentration and a hepatic cellular and lobular redistribution of ERK-1/2 that correlated with 4-HNE-protein adduct accumulation. Chronic ethanol consumption was also associated with a decrease in hepatocyte nuclear ELK-1 phosphorylation, independent of changes in total nuclear ELK-1 protein. Primary hepatocytes treated with concentrations of 4-HNE consistent with those occurring during oxidative stress displayed a concentration-dependent decrease in constitutive ERK-1/2 phosphorylation, activity, and nuclear localization that negatively correlated with 4-HNE-ERK-1/2 monomer adduct accumulation. These data paralleled the decreased phosphorylation of the downstream kinase ELK-1. Molar ratios of purified ERK-2 to 4-HNE consistent with pathologic ratios found in vivo resulted in protein monomer-adduct formation across a range of concentrations. Collectively, these data demonstrate a novel association between ethanol-induced lipid peroxidation and the inhibition of constitutive ERK-1/2, and suggest an inhibitory mechanism mediated by the lipid peroxidation product 4-hydroxynonenal
Novel mutations in TARDBP (TDP-43) in patients with familial amyotrophic lateral sclerosis.
The TAR DNA-binding protein 43 (TDP-43) has been identified as the major disease protein in amyotrophic lateral sclerosis (ALS) and frontotemporal lobar degeneration with ubiquitin inclusions (FTLD-U), defining a novel class of neurodegenerative conditions: the TDP-43 proteinopathies. The first pathogenic mutations in the gene encoding TDP-43 (TARDBP) were recently reported in familial and sporadic ALS patients, supporting a direct role for TDP-43 in neurodegeneration. In this study, we report the identification and functional analyses of two novel and one known mutation in TARDBP that we identified as a result of extensive mutation analyses in a cohort of 296 patients with variable neurodegenerative diseases associated with TDP-43 histopathology. Three different heterozygous missense mutations in exon 6 of TARDBP (p.M337V, p.N345K, and p.I383V) were identified in the analysis of 92 familial ALS patients (3.3%), while no mutations were detected in 24 patients with sporadic ALS or 180 patients with other TDP-43-positive neurodegenerative diseases. The presence of p.M337V, p.N345K, and p.I383V was excluded in 825 controls and 652 additional sporadic ALS patients. All three mutations affect highly conserved amino acid residues in the C-terminal part of TDP-43 known to be involved in protein-protein interactions. Biochemical analysis of TDP-43 in ALS patient cell lines revealed a substantial increase in caspase cleaved fragments, including the approximately 25 kDa fragment, compared to control cell lines. Our findings support TARDBP mutations as a cause of ALS. Based on the specific C-terminal location of the mutations and the accumulation of a smaller C-terminal fragment, we speculate that TARDBP mutations may cause a toxic gain of function through novel protein interactions or intracellular accumulation of TDP-43 fragments leading to apoptosis
Lightning Imaging Sensor on the International Space Station: Assessments and Results from First Year Operations
Over two decades, the NASA Marshall Space Flight Center, the University of Alabama in Huntsville, and their partners have demonstrated the effectiveness and value of space-based lightning observations as a remote sensing tool for Earth science research and applications, and, in the process, established a robust global lightning climatology. The Lightning Imaging Sensor (LIS) on the Tropical Rainfall Measuring Mission (TRMM) provided global observations of tropical lightning for an impressive 17 years before that mission came to a close in April 2015. Now a space-qualified LIS, built as the flight spare for TRMM, has been installed on the International Space Station (ISS) for a minimum two year mission following its SpaceX launch on February 19, 2017. The LIS, flown as a hosted payload on the Department of Defense Space Test Program-Houston 5 (STP-H5) mission, was delivered to the ISS in the Dragon trunk and robotically installed in an Earth-viewing position on the outside of the ISS. Following successful activation and checkout, LIS has continuously observed the amount, rate, and radiant energy lightning within its field-of-view as it orbits the Earth. Placing LIS on the Space Station provides a great opportunity to not only extend the 17-year TRMM LIS record of tropical lightning measurements but also to expand that coverage to higher latitudes missed by the previous mission. Furthermore, this mission continues the important science focus to better understand the processes which cause lightning, as well as the connections between lightning and subsequent severe weather events. This understanding is a key to improving weather predictions and saving lives and property here in the United States and around the world. The LIS measurements, along with observations from the new Geostationary Lightning Mapper (GLM) operating on NOAA's newest weather satellites, the Geosynchronous Operational Environmental Satellite-16/17 (GOES-16/17), are being used to cross-validate both systems. An especially unique contribution from the ISS platform is the production of real-time lightning data, especially valuable for operational forecasting and warning applications over data sparse regions such as the oceans. Finally, LIS provides simultaneous and complementary observations with other ISS payloads such as the European Space Agency's Atmosphere-Space Interaction Monitor (ASIM) that is exploring the connection between thunderstorms and lightning with terrestrial gamma-ray flashes (TGFs). A systematic review of the effectiveness and cost-effectiveness of peer education and peer support in prisons.
BACKGROUND: Prisoners experience significantly worse health than the general population. This review examines the effectiveness and cost-effectiveness of peer interventions in prison settings. METHODS: A mixed methods systematic review of effectiveness and cost-effectiveness studies, including qualitative and quantitative synthesis was conducted. In addition to grey literature identified and searches of websites, nineteen electronic databases were searched from 1985 to 2012. Study selection criteria were: Population: Prisoners resident in adult prisons and children resident in Young Offender Institutions (YOIs). INTERVENTION: Peer-based interventions Comparators: Review questions 3 and 4 compared peer and professionally led approaches. OUTCOMES: Prisoner health or determinants of health; organisational/ process outcomes; views of prison populations. STUDY DESIGNS: Quantitative, qualitative and mixed method evaluations. RESULTS: Fifty-seven studies were included in the effectiveness review and one study in the cost-effectiveness review; most were of poor methodological quality. Evidence suggested that peer education interventions are effective at reducing risky behaviours, and that peer support services are acceptable within the prison environment and have a positive effect on recipients, practically or emotionally. Consistent evidence from many, predominantly qualitative, studies, suggested that being a peer deliverer was associated with positive effects. There was little evidence on cost-effectiveness of peer-based interventions. CONCLUSIONS: There is consistent evidence from a large number of studies that being a peer worker is associated with positive health; peer support services are also an acceptable source of help within the prison environment and can have a positive effect on recipients. Gene Expression During the First 28 Days of Axolotl Limb Regeneration I: Experimental Design and Global Analysis of Gene Expression
While it is appreciated that global gene expression analyses can provide novel insights about complex biological processes, experiments are generally insufficiently powered to achieve this goal. Here we report the results of a robust microarray experiment of axolotl forelimb regeneration. At each of 20 post-amputation time points, we estimated gene expression for 10 replicate RNA samples that were isolated from 1 mm of heterogeneous tissue collected from the distal limb tip. We show that the limb transcription program diverges progressively with time from the non-injured state, and divergence among time adjacent samples is mostly gradual. However, punctuated episodes of transcription were identified for five intervals of time, with four of these coinciding with well-described stages of limb regeneration—amputation, early bud, late bud, and pallet. The results suggest that regeneration is highly temporally structured and regulated by mechanisms that function within narrow windows of time to coordinate transcription within and across cell types of the regenerating limb.
