    Osservazioni istochimiche e immunoistochimiche sull'ipofisi anteriore di un cane affetto da malattia di Cushing

    Gli adenomi del lobo anteriore dell'ipofisi sono neoplasie benigne piuttosto frequenti. Essi mostrano aspetto circoscritto e accrescimento non invasivo nei confronti dei tessuti circostanti. Il tumore a cellule secernenti ACTH, che colpisce le cellule cromofobe della pars anteriore dell'ipofisi, è uno di questi e viene associato alla sindrome di Cushing, causando evidente ipercortisolemia per stimolazione della corteccia surrenale. Un cane meticcio, femmina, di 7 anni, in terapia per sospetta Sindrome di Cushing e pervenuto a morte, è stato sottoposto ad accurato esame necroscopico, durante il quale si procedeva tra l'altro a prelevare ghiandole surrenali, polmone, fegato, cute, muscoli e intera ghiandola pituitaria. Su detti organi veniva eseguito l'esame istopatologico per la ricerca delle lesioni tipiche determinate dalla sindrome. Sulla ghiandola pituitaria, aumentata notevolmente di volume, venivano eseguite indagini istopatologiche specifiche, mediante colorazione OFG, per differenziare gli istotipi costituenti la pars anteriore. Tale colorazione ha consentito di riscontrare e identificare tra le cellule pituitarie gli elementi acidofili, basofili e cromofobi; queste ultime cellule, aumentate di numero, erano d'aspetto omogeneo e formavano un nucleo compatto e con scarso stroma al centro della ghiandola. La caratterizzazione cromatica e la differenziazione dai restanti istotipi ha reso possibile identificare la neoplasia come adenoma ipofisario di tipo "diffuso", caratterizzato per l'appunto da cellule disposte a strati, con scarso stroma e debole vascolarizzazione. Per verificare l'ormonoattività  delle cellule neoplastiche si è proceduto all'esame immunoistochimico per la ricerca dell'ACTH. Il risultato di tale indagine ha dato esito positivo, confermando l'intensa attività secernente delle cellule oggetto di studio

    Histopathological, histochemical and immunoistochemical study on a rare case of granulocytic sarcoma in a dog

    Granulocytic sarcoma is a malignant extramedullary solid tumor, composed of granulocytic precursor cells at various levels of differentiation. Three differentiation levels are considered: blastic, immature, and differentiated, and cases with unusual morphology. Nowadays the aid of more and more highly developed techniques allows to differentiate the various histotypes. We report a case of a 5 year-old female Schnautzer dog, died after a serious dyspnoea. During the autopsy the veterinary found several neoplasias in the lungs. It has been found a very large white-greyish mediastinal neoplasia, stuck at the trachea and several more in the parenchyma. Several samples were fixed in formalin, embedded in paraffin and exposed to histochemistry staining (H.E., Giemsa), cytochemistry (Naphthol-AS-D-Chloroacetate), and immunohistochemistry (anti-CD3, -CD79a, -CD45, -MPO, -CD45Ro, -CD34, -CD20, -CD68, -CD15, -CD30, -CD117, -CD235a, -Factor VIII, -elastase and anti-Pan-cytocheratine). Neoplasia, poorly circumscribed, was composed by a large number of neutrophil granulocytes with different degrees of differentiation, including elements of myeloid lineage. Cells were positive to MPO and focal to Naphtol-As-D-Chloroacetate and were negative to all the others antisera, allowing us to exclude lymphomas, small cells carcinomas, and tumors of monocytic and erythroid origin. By these characteristics we could diagnose a rare case of neutrophilic granulocytic sarcoma, of immature type progressing to mature form

    A preliminary analysis of COI and COII gene variability was carried out in Sardinian populations of Tortrix viridana. The aim of the study was to investigate the genetic structure and mitochondrial haplotype variation in populations associated with deciduous (Quercus pubescens) and evergreen (Q. suber and Q ilex) oaks. Forty-four haplotypes out of 87 individuals were found: three haplotypes showed a high frequency (23%, 16% and 10%) and were largely shared among populations. Hierarchical AMOVA showed no significant differentiation grouping populations for geographic areas or oak species, in spite of significant divergences structuring populations for different duration in egg development (early- vs late-hatching). However a high haplotype diversity and a low nucleotide diversity have been observed

    Selecting molecular or surface centers in carbon dots-silica hybrids to tune the optical emission: A photo-physics study down to the atomistic level

    In this work, we unveil the fluorescence features of citric acid and urea-based Carbon Dots (CDs) through a photo-physical characterization of nanoparticles synthesized, under solvent-free and open-air condi-tions, within silica-ordered mesoporous silica, as a potential host for solid-state emitting hybrids. Compared to CDs synthesized without silica matrices and dispersed in water, silica-CD hybrids display a broader emission in the green range whose contribution can be increased by UV and blue laser irradi-ation. The analysis of hybrids synthesized within different silica (MCM-48 and SBA-15) calls for an active role of the matrix in directing the synthesis toward the formation of CDs with a larger content of graphitic N and imidic groups at the expense of N-pyridinic molecules. As a result, CDs tuned in size and with a larger green emission are obtained in the hybrids and are retained once extracted from the silica matrix and dispersed in water. The kinetics of the photo-physics under UV and blue irradiation of hybrid samples show a photo-assisted formation process leading to a further increase of the relative contribution of the green emission, not observed in the water-dispersed reference samples, suggesting that the porous matrix is involved also in the photo-activated process. Finally, we carried out DFT and TD-DFT calcula-tions on the interaction of silica with selected models of CD emitting centers, like surface functional groups (OH and COOH), dopants (graphitic N), and citric acid-based molecules. The combined experimen-tal and theoretical results clearly indicate the presence of molecular species and surface centers both emitting in the blue and green spectral range, whose relative contribution is tuned by the interaction with the surrounding media

    Six-Month Synbio® Administration Affects Nutritional and Inflammatory Parameters of Older Adults Included in the PROBIOSENIOR Project

    The physiological changes associated with ageing contribute to the incidence of diseases, morbidity, and mortality. For modern society, it is essential to find solutions to improve elderly people’s health and quality of life. Among promising strategies, the PROBIOSENIOR project proposed a daily six-month supplementation with new probiotic functional foods and nutraceuticals. The aim of this work was to evaluate the modulating effects of the probiotic diet on inflammatory markers and nutritional status. Ninety-seven elderly volunteers were randomly assigned to either a placebo-diet group or a probiotic-diet group (SYNBIO®). Faeces, urine, and blood samples were collected before and after the supplementation to determine serum cytokines, biogenic amines, and inflammation markers. Comparing the results obtained before and after the intervention, probiotic supplementations significantly decreased the TNF- circulating levels and significantly increased those of IGF-1. Biogenic-amine levels showed high variability, with significant variation only for histamine that decreased after the probiotic supplementation. The supplementation influenced the serum concentration of some crucial cytokines (IL-6, IL-8, and MIP-1 ) that significantly decreased in the probiotic group. In addition, the Mini Nutritional Assessment questionnaire revealed that the probiotic-supplemented group had a significant improvement in nutritional status. In conclusion, the PROBIOSENIOR project demonstrated how SYNBIO® supplementation may positively influence some nutritional and inflammatory parameters in the elderly

    Application of a new method for GWAS in a related case/control sample with known pedigree structure: identification of new loci for nephrolithiasis

    In contrast to large GWA studies based on thousands of individuals and large meta-analyses combining GWAS results, we analyzed a small case/control sample for uric acid nephrolithiasis. Our cohort of closely related individuals is derived from a small, genetically isolated village in Sardinia, with well-characterized genealogical data linking the extant population up to the 16(th) century. It is expected that the number of risk alleles involved in complex disorders is smaller in isolated founder populations than in more diverse populations, and the power to detect association with complex traits may be increased when related, homogeneous affected individuals are selected, as they are more likely to be enriched with and share specific risk variants than are unrelated, affected individuals from the general population. When related individuals are included in an association study, correlations among relatives must be accurately taken into account to ensure validity of the results. A recently proposed association method uses an empirical genotypic covariance matrix estimated from genome-screen data to allow for additional population structure and cryptic relatedness that may not be captured by the genealogical data. We apply the method to our data, and we also investigate the properties of the method, as well as other association methods, in our highly inbred population, as previous applications were to outbred samples. The more promising regions identified in our initial study in the genetic isolate were then further investigated in an independent sample collected from the Italian population. Among the loci that showed association in this study, we observed evidence of a possible involvement of the region encompassing the gene LRRC16A, already associated to serum uric acid levels in a large meta-analysis of 14 GWAS, suggesting that this locus might lead a pathway for uric acid metabolism that may be involved in gout as well as in nephrolithiasis

    An Insight into Z-Drug Abuse and Dependence: An Examination of Reports to the European Medicines Agency Database of Suspected Adverse Drug Reactions

Published by Oxford University Press on behalf of CINP.BACKGROUND: Although originally marketed as safe alternatives to the habit-forming benzodiazepines, growing numbers of zaleplon, zolpidem, and zopiclone ("Z-drugs") clinical concerns relating to their potential of abuse, dependence, and withdrawal have been reported over time. We aimed here at assessing these issues analyzing datasets of adverse drug reactions provided by the European Medicines Agency through the EudraVigilance system. METHODS: Analyzing the adverse drug reactions databases of each Z-drug, descriptive analyses have been performed on cases and proportional reporting ratios (PRRs) computed. RESULTS: An overall number of 33 240 (e.g., 23 420 zolpidem; 9283 zopiclone; and 537 zaleplon) misuse-, abuse-, dependence-, and withdrawal-related adverse drug reactions, corresponding to some 6246 unique patients given Z-drugs, were here identified. Cases were studied and described, including demographic characteristics and clinical data such as concomitant drugs, doses, routes of administration, and outcomes of the reactions (being fatalities recorded). Considering PRR values and in comparison with zopiclone, zolpidem was more frequently involved in both misuse/abuse and withdrawal issues. Zolpidem and zopiclone presented with the same dependence risk, but zopiclone was most involved in overdose adverse drug reactions. Compared with zaleplon, zopiclone presented higher dependence and overdose-related issues but slightly lower misuse/abuse and withdrawal PRR values. CONCLUSION: Current data may only represent a gross underestimate of the real prevalence of Z-drug misuse. Caution should be exercised when prescribing those molecules, especially for patients with psychiatric illnesses and/or history of drug abuse. We recommend the need to invest in proactive pharmacovigilance activities to better and promptly detect, understand, and prevent any possible misuse potential of prescribed medications.Peer reviewe

    Azimuthal anisotropy of charged jet production in root s(NN)=2.76 TeV Pb-Pb collisions

    We present measurements of the azimuthal dependence of charged jet production in central and semi-central root s(NN) = 2.76 TeV Pb-Pb collisions with respect to the second harmonic event plane, quantified as nu(ch)(2) (jet). Jet finding is performed employing the anti-k(T) algorithm with a resolution parameter R = 0.2 using charged tracks from the ALICE tracking system. The contribution of the azimuthal anisotropy of the underlying event is taken into account event-by-event. The remaining (statistical) region-to-region fluctuations are removed on an ensemble basis by unfolding the jet spectra for different event plane orientations independently. Significant non-zero nu(ch)(2) (jet) is observed in semi-central collisions (30-50% centrality) for 20 <p(T)(ch) (jet) <90 GeV/c. The azimuthal dependence of the charged jet production is similar to the dependence observed for jets comprising both charged and neutral fragments, and compatible with measurements of the nu(2) of single charged particles at high p(T). Good agreement between the data and predictions from JEWEL, an event generator simulating parton shower evolution in the presence of a dense QCD medium, is found in semi-central collisions.     Production of He-4 and (4) in Pb-Pb collisions at root(NN)-N-S=2.76 TeV at the LHC

    Results on the production of He-4 and (4) nuclei in Pb-Pb collisions at root(NN)-N-S = 2.76 TeV in the rapidity range vertical bar y vertical bar <1, using the ALICE detector, are presented in this paper. The rapidity densities corresponding to 0-10% central events are found to be dN/dy4(He) = (0.8 +/- 0.4 (stat) +/- 0.3 (syst)) x 10(-6) and dN/dy4 = (1.1 +/- 0.4 (stat) +/- 0.2 (syst)) x 10(-6), respectively. This is in agreement with the statistical thermal model expectation assuming the same chemical freeze-out temperature (T-chem = 156 MeV) as for light hadrons. The measured ratio of (4)/He-4 is 1.4 +/- 0.8 (stat) +/- 0.5 (syst). (C) 2018 Published by Elsevier B.V.Peer reviewe
