DualTable: A Hybrid Storage Model for Update Optimization in Hive
Hive is the most mature and prevalent data warehouse tool providing SQL-like
interface in the Hadoop ecosystem. It is successfully used in many Internet
companies and shows its value for big data processing in traditional
industries. However, enterprise big data processing systems as in Smart Grid
applications usually require complicated business logics and involve many data
manipulation operations like updates and deletes. Hive cannot offer sufficient
support for these while preserving high query performance. Hive using the
Hadoop Distributed File System (HDFS) for storage cannot implement data
manipulation efficiently and Hive on HBase suffers from poor query performance
even though it can support faster data manipulation. There is a project based on
Hive issue Hive-5317 to support update operations, but it has not been finished
in Hive's latest version. Since this ACID compliant extension adopts same data
storage format on HDFS, the update performance problem is not solved.
In this paper, we propose a hybrid storage model called DualTable, which
combines the efficient streaming reads of HDFS and the random write capability
of HBase. Hive on DualTable provides better data manipulation support and
preserves query performance at the same time. Experiments on a TPC-H data set
and on a real smart grid data set show that Hive on DualTable is up to 10 times
faster than Hive when executing update and delete operations.Comment: accepted by industry session of ICDE201
FDTD Modelling of Silver Nanoparticles Embedded in Phase Separation Interface of H-PDLC
We report localized surface plasmon resonance (LSPR) of silver nanoparticles (NPs) embedded in interface of phase separation of holographic polymer-dispersed liquid crystal (H-PDLC) gratings using Finite-Difference Time Domain method. We show that silver NPs exhibit double resonance peak at the interface, and these peaks are influenced by the angle of incident light. We observe a blue shift of the wavelength of resonance peak as the incident angle increases. However, the location of silver NPs at the interface has nearly no effect on the wavelength of resonance peak. Also we show near-field and far-field properties surrounding silver NPs and find that field distribution can be controlled through rotation of incident angle. Therefore, LSPR properties of silver NPs within H-PDLC gratings can be excited by appropriate wavelength and angle of the incident light
Inhibition of autophagy by 3-MA enhances IL-24-induced apoptosis in human oral squamous cell carcinoma cells
Abstract
Background
Interleukin-24(IL-24), also referred to as melanoma differentiation-associated gene-7(mda-7), is a unique member of the IL-10 gene family, which displays nearly ubiquitous cancer-specific toxicity. The most notable feature of IL-24 is selectively induced growth suppression and apoptosis in various cancer cells, with no harmful effects toward normal cells. Autophagy is a self-protective mechanism in many kinds of tumor cells that respond to anticancer treatment. It is reported that autophagy inhibition could enhance the effects of many kinds of anticancer treatments, including gene therapy. However, whether IL-24 is effective to treat oral squamous cell carcinomas (OSCC) and if autophagy inhibition could improve the anticancer effect of IL-24 towards OSCC is has not been detected.
Methods
MTT assays were carried out to determine the cell proliferation; Transfection was used to gene transfer; Western Blot was performed to detect the protein level of LC3II, P62, Beclin 1, Cleaved caspase-3, β-Tubulin and β-actin; Apoptosis rates and cell cycle alteration were analyzed using flow cytometry; Autophagy induction was confirmed by MDC staining, GFP-LC3 staining and transmission electron microscopy. Amount of IL-24 in the culture medium was quantified by ELISA. Apoptosis in vivo was analyzed by TUNEL assay. HE staining was used to observe the morphology of the samples.
Results
In the present study, we proved that IL-24 have a novel anticancer effect towards KB cells and that autophagy inhibition could improve the anticancer effect of IL-24. IL-24 treated cells showed autophagy characteristics and autophagy inhibition by 3-methyladenine (3-MA) significantly enhanced IL-24-induced apoptosis. Similar results were obtained in the KB cells xenograft tumor model.
Conclusions
These results suggest that the combination of autophagy inhibitors and IL-24 based on the AdLTR2EF1α-mediated gene transfer could be a promising way to cure OSCC.http://deepblue.lib.umich.edu/bitstream/2027.42/113230/1/13046_2015_Article_211.pd
Allogeneic Mesenchymal Stem Cell Therapy for Bisphosphonate-Related Jaw Osteonecrosis in Swine
Bisphosphonates (BPs), which are used to treat a variety of clinical disorders, have the side effect of jawbone necrosis. Currently, there is no reliable treatment for BP-related osteonecrosis of the jaw (BRONJ) due to a lack of understanding of its pathogenesis. To investigate the pathogenesis of BRONJ and observe the treatment effect of bone marrow mesenchymal stem cell (BMMSC) transplantation, we established a preclinical animal model of BRONJ in miniature pigs (minipigs). After treatment with zoledronic acid, the clinical and radiographic manifestations of BRONJ could be observed in minipigs after first premolar extraction. The biological and immunological properties of BMMSCs were impaired in the BP-treated minipigs. Moreover, the ratio of Foxp3-positive regulatory T-cells (Tregs) in peripheral blood decreased, and interleukin (IL)-17 increased in the serum of BP-treated minipigs. After allogeneic BMMSC transplantation via intravenous infusion, mucosal healing and bone reconstruction were observed; IL-17 levels were reduced; and Tregs were elevated. In summary, we established a clinically relevant BRONJ model in minipigs and tested a promising allogeneic BMMSC-based therapy, which may have potential clinical applications for treating BRONJ. © Copyright 2013, Mary Ann Liebert, Inc. 2013
Oxidation kinetics and non-Marcusian charge transfer in dimensionally confined semiconductors
Electrochemical reactions represent essential processes in fundamental
chemistry that foster a wide range of applications. Although most
electrochemical reactions in bulk substances can be well described by the
classical Marcus-Gerischer charge transfer theory, the realistic reaction
character and mechanism in dimensionally confined systems remain unknown. Here,
we report the multiparametric survey on the kinetics of lateral photooxidation
in structurally identical WS2 and MoS2 monolayers, where electrochemical
oxidation occurs at the atomically thin monolayer edges. The oxidation rate is
correlated quantitatively with various crystallographic and environmental
parameters, including the density of reactive sites, humidity, temperature, and
illumination fluence. In particular, we observe distinctive reaction barriers
of 1.4 and 0.9 eV for the two structurally identical semiconductors and uncover
an unusual non-Marcusian charge transfer mechanism in these dimensionally
confined monolayers due to the limit in reactant supplies. A scenario of band
bending is proposed to explain the discrepancy in reaction barriers. These
results add important knowledge into the fundamental electrochemical reaction
theory in low-dimensional systems.Comment: 20 pages, 4 figure
