    The effects of diazinon on haematological indices and LC50(96h) of Acipenser nudiventris

    We treated 150 specimens of Acipenser nudiventris with diazinon and used 30 individuals as control to assess the haematological effects of the pesticide and LC50(96h) for the fish. The acute toxicity test lasting 96 hours was conducted on juvenile Acipenser nudiventris kept in aerated stagnant water contaminated with diazinon. The LC50(96h) values of diazinon for Acipenser nudiventris juveniles was 4.6 mg/i. The experimental groups of fish showed statistically significant difference in their erythrocyte, haemoglobine, leukocyte and differential leukocyte count (P<0.05). Acute toxicity effects of diazinon on the fish was recorded as lowered erythrocyte and lymphocyte count with a significant increase in developed neutrophile count (P<0.05). It is suggested that the change in erythrocyte and leukocyte count may have been caused as a result of disruption of haematopoiesis and a decrease in non-specific immunity level of the fish

    Some hematological and biochemical changes in blood serum of Beluga (Huso huso) after chronic exposure to diazinon

    Hematological and biochemical changes were studied in beluga (Huso huso) weighing 450±50g in the presence of b,1-3, D-glucan as a single intraperitoneal injection (0.3mg/kg body weight) once, and thereafter a chronic exposure to organophosphate, diazinon at concentration of 1.5 mg/L as a continuous bath for 63 days at 22±1ºC. Results of erythrocyte profile in fish treated with glucan, but without diazinon, generally showed significant higher values of RBC (P0.05) in the values of HCT, Hb, MCH and MCHC between the two groups. Also, fish treated with glucan, but without diazinon bath, showed significant higher values of RBC than that untreated fish up to 3 weeks post-treatment (P0.05). Furthermore, fish treated with diazinon showed higher levels of alkaline phosphatase (ALP), alanine aminotransferase (ALT), lactate dehydrogenase (LDH) than other three groups for 14-28 days post-treatment and then they decreased to the lower level for the rest of experiment. Also, fish treated with glucan-diazinon showed insignificant higher levels of these enzymes up to 21 days post-exposure compare to fish treated with glucan but without diazinon bath (P>0.05). However, such levels reduced to a lower level in this group for the rest of experiment (P>0.05). Also, fish treated with glucan-diazinon showed generally lower and higher levels of total protein and glucose concentrations in blood plasma, respectively compared to fish treated with glucan but without diazinon bath (P<0.05)

    Determination of polycyclic aromatic hydrocarbons (PAHs) in water, sediment and tissues of five sturgeon species in the southern Caspian Sea coastal regions

    Concentration levels of 16 polycyclic aromatic hydrocarbons (PAHs) consisting of naphthalene, acenaphthylene, acenaphthene, fluorine, phenantherene, anthracene, fluoranthene, pyrene, benzo(a)anthracene, chrysene, benzo(b)fluoranthene, benzo(k)fluoranthene, benzo(a)pyrene, dibenzo(a,h)anthracene, indeno(1,2,3-cd) pyrene and benzo(g,h,i)perylene were measured in water and sediment samples collected from the southern Caspian Sea coasts at four stations of Noshahr, Freydoonkenar, Sari and Amirabad during autumn, winter and spring of 2005-2006. Also, tissue samples from five sturgeon species including stellate sturgeon, Persian sturgeon, beluga, Russian sturgeon and ship sturgeon were obtained. Samples were analyzed by gas chromatography after the extraction process and the obtained data were statistically analyzed using One-Way ANOVA analysis at confidence level 95%. The mean concentrations of PAHs in water and sediment samples were 0.004-2.946 mgl-1 and 0.024- 2.336 µg g-1, respectively. No significant difference was found in the mean concentrations of PAHs among stations and seasons of the examined samples. Also, mean concentration of PAHs in the liver, muscles,gills, kidney and gonads of five sturgeon species was 0.81-1.34 µgg-1. The results of this study show that the levels of PAHs in water, sediment and sturgeon organs were below the acceptable levels of PAHs proposed by USEPA and WHO

    Effects of Zataria multiflora and Eucalyptus globolus essential oils on haematological parameters and respiratory burst activity in Cyprinus carpio

    The present study was undertaken to investigate the effects of Zataria multiflora and Eucalyptus globolus essential oils on some haematological parameters and respiratory burst activity in common carp (Cyprinus carpio). 260 fish (30±5g) were randomly distributed in 13 treatment groups; each one in three replicates and different doses of essential oils in 16-17ºC were administrated. The fish were sampled on day 1, 2, 8, 15 and 22 after the 8-day trial. Haematological parameters (red blood cell count, haematocrit) and respiratory burst activity were then evaluated in all treatment groups. The results suggest that essential oils especially Zataria multiflora in dietary intake significantly enhanced respiratory burst activity of blood neutrophlis (P< 0.05). Meanwhile, essential oils had moderate effects on RBC and haematocrit. Significant increases in RBC and haematocrit levels were just noted in T11 treatment group (P< 0.05). This study indicates that dietary administration of Zataria multiflora and Eucalyptus globolus essential oils could be used to promote the health status of common carp during temperature stress

    Effect of Zataria multiflora Boiss essential oil on the growth of Staphylococcus aureus in the light salted fillets of silver carp (Hypophthalmichthys molitrix)

    The effects of different concentrations of Zataria multiflora essential oils at concentrations of 0, 0.005, 0.015, 0.045, 0.135, 0.405 and 0.810% on the growth of Staphylococcus aureus were studied in a food model system, light salted fillets of silver carp (Hypophthalmichthys molitrix) at storage temperature of 10ºC for 21 days. The results showed that there was no significant difference in bacterial growth between samples treated with different concentrations of Z. multiflora essential oil and control group immediately post-inoculation. However, there was significant difference in growth of S. aureus between samples treated with concentration of 0.135 of Z. multiflora essential oil and control samples (P0.05) except of 1-day post-storage. The most inhibitorial effects were observed in the samples treated with 0.405% and 0.8 10% of Z. multiflora essential oil up to 9 and 12 days post- storage, respectively

    Selected morpho-chemical features of hemocytes in farmed shrimp, Fenneropenaeus indicus in Iran

    The aim of the present study was to determine various types of hemocytes, total and differential hemocyte count and to show some indices of phagocytosis, including percent phagocytosis and phagocytic index in Indian white shrimp, Fenneropenaeus indicus in Iranian shrimp farms. The hemolymph was extracted from the shrimps and mixed with anticoagulant. It then stained and Yeast was added as foreign particles to samples. Based on the cell size and presence of the granules and nucleocytoplasmic ratio, three major groups of hemocytes, including hyaline cells, small granular cells (SGC) and large granular cells (LGC) were identified. Hyaline cell (HC) was the smallest hemocyte. HC (hyaline cell) about was 10-15%, lower quantity in comparison to some species and those of LGC and SGC were 20-25% and 60-65%, respectively, suggesting that relative percentage of differential hemocyte count in Fenneropenaeus indicus is different from other decapods. Also, in this study, it was shown that SGH and LGH could engulf the yeast particles. In Contrast to some previous studies, no engulfment was observed by hyaline cells in vitro. In this study it was showed that there are some variations in total and differential hemocyte count compare to other species in the family of Penaeidae

    Shrimp farmers have been suffering from loss of production due to White Spot Disease (WSD) in the last decades. A few studies focused on the environmental factors predisposed WSD. Our study aimed to investigate the effects of the different salinities on occurrence of WSD. The treatments were 30 ppt, 40 ppt and 50 ppt in triplicate and corresponding untreated control groups. White Spot Virus (WSV) was intramuscularly injected to the shrimp in whole treatments. PCR and histopathological findings within the measurement of some hemolymph parameters such as Differentiated Hemocyte Count (DHC), Total Hemocyte Count (THC) and Total Plasma Protein (TPP) were carried out. A positive result was observed for the occurrence of the antigen of the WSV obtained from the samples of each treatment of salinity due to the challenging treatments with the WSV. Mortality at the salinity of 30 ppt began at 72 h post inoculation as well as the salinity of 40 ppt, but at 50 ppt it started after 50 h. The Minimum and maximum count of mortality at the salinity of 30, 40 and 50 ppt were 3.5, 8.5 0.5, 4.5 and 1.5, 7.5, respectively. No mortality was observed in the untreated control groups of 30, 40 and 50 ppt during the experiment. It is concluded that the higher and the lower salinity, lesser or greater than the normal condition in exposed to WSV could lead to severe mortality of WSD

    Survey on achieving recombinant DNA vaccine (live delivery) against streptococcosis for Rainbow trout (Oncorhynchus mykiss) immunization

    The main purpose of this study was survey on achieving recombinant DNA vaccine (live delivery) against streptococcusis for Rainbow trout (Onchorhynchus mykiss) Immunization. Initially, a total of 515 samples were collected from the head kidney of diseased fish (weigh 50_ 200g) in 72 farms of 8 provinces. Approximately, 40% (206 samples) of specimens were infected to Streptococcus species. Then were isolated 172 DNA samples and consequently, five pathogenic species have been identified, including S. iniae, S. faecium, S. agalactiae, S. dysgalatiae, and S. uberis. The enzyme phosphoglucomutase (PGM) has recently been discovered to play an important role in polysaccharide capsule production and virulence in S. iniae. Therefore, was initially isolated S. iniae and cloning phosphoglucomutase gene. Then, the PGM gene was amplified successfully and cloned in pTZ57R cloning vector. The recombinant plasmid was sub cloned into pETD uet-l expression vector by restriction enzymes and confirmed by PCR. Meanwhile, for amplifying simA and cpsD genes were used universal primers pNZ8148 and special for simA and cpsD genes.The recombinant bacteria Lactococcus lactis (NZ9000) was used for transformation the plasmid into Lactococcus lactis. Vaccination was performed by oral, bath and injection (peritoneal) methods. The efficiency of g2 was better than g1in these three methods and all of groups. The determination of IgM level, or detection of anti S. iniae antibody was carried out by using ELISA. The results revealed that there was a significant (p<0.05) difference between the level of IgM in all three methods and experiment groups compare to control group. The results of challenge of vaccinated fish with S. iniae showed that fish RPS in all of groups except group 3 (42.63 percent) were more than 50 percent while, in control group was 21.43 percent. The highest fish RPS was belong to group 11(61.25 percent) and statistical analyses revealed that significant (p<0.05) difference between fish vaccinated RPS, compare to control group

    Bioassay of combined hydrogen peroxide and silver ion at four life stages of Indian white shrimp (Fennerop enaeus indicus)

    Silver ion and hydrogen peroxide act synergistically as a strong disinfectant. The purpose of this study was to assess the effect of combined hydrogen peroxide 50% and silver ion 0.05% at four life stages of Indian white shrimp (Fenneropenaeus indicus) and to evaluate the feasibility of using this substance in shrimp culture. The Trimmed Spear man karber software was applied for determining EC _(50)/96h and LC sub(50)/96h with 95% confidence limit on the 6000 shrimp based on OECD static method. The experiments were conducted in Iran Shrimp Research Center, Bushehr, during 2005 -2006. The EC _(50) values of 132.5, 67.89, 55.56, 51.95 and 48 .6ppm were obtained in PL15 stage after 12, 24, 48, 72 and 96 hours, respectively. Also these were 147.57, 70.83, 60.01, 54.89, 41.19 for PL_(45) stage, and 306.43, 174.14, 113.62, 78.21, 61.96 for sub adult stage (12 plus or minus 1 grams), res pectively. In addition, the EC _(50) values of 243.25, 130.55, 75.56, 61. 18 and 51.59ppm were obtained at adult stage (20 plus or minus 2 grams), respectively. The LC sub(50) values of 239.81, 101, 74.28, 65.72 and 61.45ppm were obtained in PL _(15) stage after 12, 24, 48, 72 and 96 hours, respectively. Also thes e were 304.56, 160.12, 113.1, 93.69, 79.38 for PL _(45) stage, and 712.13 , 518.44, 265.29, 145.53, 103.76 for sub adult stage, respectively. In addition, the LC _(50) values of 827.75, 508.91, 317.3, 139.44 and 85.88ppm were obtained at adult stage, respectively. The statistical results showed that; no observed effect concentration" (NOEC) of this substance was 20ppm, and the "lowest observed effect concentration" (LOEC) was 40ppm, thus "maximum allowable concentration" (MAC) value was determined 28.8 ppm on the Indian white shrimp. Therefore this combined chemical should be used under determined MAC value with a complete precautionary as a disinfectant for Indian white shrimp

    Metamagnetic phase transition in the Ising plus Dzyaloshinskii-Moriya model

    We study the 1D ferromagnetic Ising (spin-1/2) model with the Dzyaloshinskii-Moriya (DM) interaction. We analyze the low energy excitation spectrum and the ground state magnetic phase diagram using the Lanczos method. The DM interaction-dependency is calculated for the low-energy excitation spectrum, spiral order parameter and spin-spin correlation functions. We show that a metamagnetic quantum phase transition occurs between the ferromagnetic and spiral phases. The existence of the metamagnetic phase transition is confirmed, using the variational matrix product states approach.Comment: 5 pages, 6 figure
