    Effectively Reaching the Equine Community in Kentucky

    Since 2009, Extension Specialists and County Extension Agents from the University of Kentucky in the Southeast United States have conducted an annual educational event; the Kentucky Equine Farm and Facilities Expo field-day. These annual field-days have provided practical, hands-on demonstrations and seminars for 135-250 members of the equine community per event. Participants have been a mix of horse owners in relation to farm size, number of horses and business vs. recreation. Many participants have not been previous consumers of Extension educational programming. One of the major draws of the Farm and Facility Expo field days has been the host sites, which have included various well-known Thoroughbred, Sport-Horse and Quarter Horse farms, with an increased number of participants observed when the event was held at well-known farms. Partnerships have been established with many equine related businesses, with over 30 companies displaying their products and services at the event. The educational programs at these events have been conducted by Extension Specialists, industry experts and Extension Agents and have included timely topics such as weed identification and control, vaccination protocols, farm safety, pasture management and renovation, rotational grazing, feeding and nutrition, fencing options, barn design, on-farm solar energy and many others. Post-event evaluations have been conducted and, of the more than 2,400 persons who attended these events, 84.25% of the respondents have found the information presented useful in the management of their horse operation, 78.5% planned to make at least one change in their horse operation and 90% were very satisfied with the event

    A framework for organizing cancer-related variations from existing databases, publications and NGS data using a High-performance Integrated Virtual Environment (HIVE)

    Years of sequence feature curation by UniProtKB/Swiss-Prot, PIR-PSD, NCBI-CDD, RefSeq and other database biocurators has led to a rich repository of information on functional sites of genes and proteins. This information along with variation-related annotation can be used to scan human short sequence reads from next-generation sequencing (NGS) pipelines for presence of non-synonymous single-nucleotide variations (nsSNVs) that affect functional sites. This and similar workflows are becoming more important because thousands of NGS data sets are being made available through projects such as The Cancer Genome Atlas (TCGA), and researchers want to evaluate their biomarkers in genomic data. BioMuta, an integrated sequence feature database, provides a framework for automated and manual curation and integration of cancer-related sequence features so that they can be used in NGS analysis pipelines. Sequence feature information in BioMuta is collected from the Catalogue of Somatic Mutations in Cancer (COSMIC), ClinVar, UniProtKB and through biocuration of information available from publications. Additionally, nsSNVs identified through automated analysis of NGS data from TCGA are also included in the database. Because of the petabytes of data and information present in NGS primary repositories, a platform HIVE (High-performance Integrated Virtual Environment) for storing, analyzing, computing and curating NGS data and associated metadata has been developed. Using HIVE, 31 979 nsSNVs were identified in TCGA-derived NGS data from breast cancer patients. All variations identified through this process are stored in a Curated Short Read archive, and the nsSNVs from the tumor samples are included in BioMuta. Currently, BioMuta has 26 cancer types with 13 896 small-scale and 308 986 large-scale study-derived variations. Integration of variation data allows identifications of novel or common nsSNVs that can be prioritized in validation studies

    Spikey: self-lensing flares from eccentric SMBH binaries

    We examine the light curves of two quasars, motivated by recent suggestions that a supermassive black hole binary (SMBHB) can exhibit sharp lensing spikes. We model the variability of each light curve as due to a combination of two relativistic effects: the orbital relativistic Doppler boost and gravitational binary self-lensing. In order to model each system, we extend previous Doppler plus self-lensing models to include eccentricity. The first quasar is identified in optical data as a binary candidate with a 20-yr period (Ark 120), and shows a prominent spike. For this source, we rule out the lensing hypothesis and disfavour the Doppler-boost hypothesis due to discrepancies in the measured versus recovered values of the binary mass and optical spectral slope. The second source, which we nickname Spikey, is the rare case of an active galactic nucleus identified in Kepler’s high-quality, high-cadence photometric data. For this source, we find a model, consisting of a combination of Doppler modulation and a narrow symmetric lensing spike, consistent with an eccentric SMBHB with a mass of M_(tot) =3 × 10⁷ M_⊙⁠, rest-frame orbital period T = 418 d, eccentricity e = 0.5, and seen at an inclination of 8○ from edge-on. This interpretation can be tested by monitoring Spikey for periodic behaviour and recurring flares in the next few years.     VLA FRAMEx. I. Wideband Radio Properties of the AGN in NGC 4388

    We present the first results from Karl G. Jansky Very Large Array (VLA) observations as a part of the Fundamental Reference Active Galactic Nucleus (AGN) Monitoring Experiment (FRAMEx), a program to understand the relationship between AGN accretion physics and wavelength-dependent position as a function of time. With this VLA survey, we investigate the radio properties from a volume-complete sample of 25 hard X-ray-selected AGNs using the VLA in its wideband mode. We observed the targets in the A-array configuration at 4124-12 GHz with all polarization products. In this work, we introduce our calibration and imaging methods for this survey, and we present our results and analysis for the radio quiet AGN NGC 4388. We calibrated and imaged these data using the multi-term, multi-frequency synthesis imaging algorithm to determine its spatial, spectral and polarization structure across a continuous 4124-12 GHz band. In the AGN, we measure a broken power law spectrum with α=0.06\alpha=-0.06 below a break frequency of 7.3 GHz and α=0.34\alpha=-0.34 above. We detect polarization at sub-arcsecond resolution across both the AGN and a secondary radio knot. We compare our results to ancillary data and find that the VLA radio continuum is likely due to AGN winds interacting with the local interstellar medium that gets resolved away at sub-parsec spatial scales as probed by the Very Long Baseline Array. A well-known ionization cone to the southwest of the AGN appears likely to be projected material onto the underside of the disk of the host galaxy.Comment: 22 pages, 9 figures, Accepted in Ap

    Enhancing Cardiac Rehabilitation With Stress Management Training

    BACKGROUND: Cardiac rehabilitation (CR) is the standard of care for patients with coronary heart disease. Despite considerable epidemiological evidence that high stress is associated with worse health outcomes, stress management training (SMT) is not included routinely as a component of CR. METHODS AND RESULTS: One hundred fifty-one outpatients with coronary heart disease who were 36 to 84 years of age were randomized to 12 weeks of comprehensive CR or comprehensive CR combined with SMT (CR+SMT), with assessments of stress and coronary heart disease biomarkers obtained before and after treatment. A matched sample of CR-eligible patients who did not receive CR made up the no-CR comparison group. All participants were followed up for up to 5.3 years (median, 3.2 years) for clinical events. Patients randomized to CR+SMT exhibited greater reductions in composite stress levels compared with those randomized to CR alone (P=0.022), an effect that was driven primarily by improvements in anxiety, distress, and perceived stress. Both CR groups achieved significant, and comparable, improvements in coronary heart disease biomarkers. Participants in the CR+SMT group exhibited lower rates of clinical events compared with those in the CR-alone group (18% versus 33%; hazard ratio=0.49; 95% confidence interval, 0.25-0.95; P=0.035), and both CR groups had lower event rates compared with the no-CR group (47%; hazard ratio=0.44; 95% confidence interval, 0.27-0.71; P<0.001). CONCLUSIONS: CR enhanced by SMT produced significant reductions in stress and greater improvements in medical outcomes compared with standard CR. Our findings indicate that SMT may provide incremental benefit when combined with comprehensive CR and suggest that SMT should be incorporated routinely into CR.     A Synthetic Agonist to Vasoactive Intestinal Peptide Receptor-2 Induces Regulatory T Cell Neuroprotective Activities in Models of Parkinson\u27s Disease

    A paradigm shift has emerged in Parkinson\u27s disease (PD) highlighting the prominent role of CD4+ Tregs in pathogenesis and treatment. Bench to bedside research, conducted by others and our own laboratories, advanced a neuroprotective role for Tregs making pharmacologic transformation of immediate need. Herein, a vasoactive intestinal peptide receptor-2 (VIPR2) peptide agonist, LBT-3627, was developed as a neuroprotectant for PD-associated dopaminergic neurodegeneration. Employing both 6-hydroxydopamine (6-OHDA) and α-synuclein (α-Syn) overexpression models in rats, the sequential administration of LBT-3627 increased Treg activity without altering cell numbers both in naïve animals and during progressive nigrostriatal degeneration. LBT-3627 administration was linked to reductions of inflammatory microglia, increased survival of dopaminergic neurons, and improved striatal densities. While α-Syn overexpression resulted in reduced Treg activity, LBT-3627 rescued these functional deficits. This occurred in a dose-dependent manner closely mimicking neuroprotection. Taken together, these data provide the basis for the use of VIPR2 agonists as potent therapeutic immune modulating agents to restore Treg activity, attenuate neuroinflammation, and interdict dopaminergic neurodegeneration in PD.
