    Effect of microwave treatment on the physicochemical properties of potato starch granules

    BACKGROUND: The degree of polymerization of amylose starch in potato was so large that the gel was hardness after gelatinization. Therefore, it is one of the most important ways that the microwave treatment was used to change the physicochemical properties of starch gel to make it suitable for the preparation of instant food. RESULTS: The effect of microwave treatment on the physicochemical properties including morphology, crystalline structure, molecular weight distribution and rheological properties of potato starch granules was evaluated by treating time of varying duration (0, 5, 10, 15, 20 s) at 2450 MHz and 750 W. Scanning electron micrographs (SEM) of potato starch granules showed flaws or fractures on the surface after 5 to 10s of microwaving and collapse after 15 to 20 s. Polarized light microscopy (PLM) indicated that microwave treating damaged the crystalline structure of potato starch, such that the birefringence of starch granules gradually decreased after 5 to 10s and even disappeared after microwaving from 15 to 20 s. The molecular weight (Mw) values of potato starch and the proportion of large M(W) fraction were considerably reduced with increasing the microwave treating time from 0 to 20s. The molecular weight slowly decreased over 5 ~ 15 s microwave treating but decreased abruptly at the time of 20s microwave treating. The apparent viscosity decreased as shear rate increased and presented shear-thinning behavior. The magnitudes of the storage modulus (G’) and loss modulus (G”) obtained at each shear rate increased with duration of microwave treating from 0 to 15 s but decreased from 15 to 20 s. CONCLUSIONS: These results demonstrated that the morphology and crystalline structure was damaged by microwave treatment. The high molecular weight of potato starch above 2 × 10(8) Da was so sensitive to the vibrational motion of the polar molecules due to the application microwave energy and broke easily for longer dextran chains.     From Sky to the Ground: A Large-scale Benchmark and Simple Baseline Towards Real Rain Removal

    Learning-based image deraining methods have made great progress. However, the lack of large-scale high-quality paired training samples is the main bottleneck to hamper the real image deraining (RID). To address this dilemma and advance RID, we construct a Large-scale High-quality Paired real rain benchmark (LHP-Rain), including 3000 video sequences with 1 million high-resolution (1920*1080) frame pairs. The advantages of the proposed dataset over the existing ones are three-fold: rain with higher-diversity and larger-scale, image with higher-resolution and higher-quality ground-truth. Specifically, the real rains in LHP-Rain not only contain the classical rain streak/veiling/occlusion in the sky, but also the \textbf{splashing on the ground} overlooked by deraining community. Moreover, we propose a novel robust low-rank tensor recovery model to generate the GT with better separating the static background from the dynamic rain. In addition, we design a simple transformer-based single image deraining baseline, which simultaneously utilize the self-attention and cross-layer attention within the image and rain layer with discriminative feature representation.     Hairy root transgene expression analysis of a secretory peroxidase (PvPOX1) from common bean infected by Fusarium wilt

    Plant peroxidases (POXs) are one of the most important redox enzymes in the defense responses. However, the large number of different plant POX genes makes it necessary to carefully confirm the function of each paralogous POX gene in specific tissues and disease interactions. Fusarium wilt is a devastating disease of common bean caused by Fusarium oxysporum f. sp. phaseoli. In this study, we evaluated a peroxidase gene, PvPOX1, from a resistant common bean genotype, CAAS260205 and provided direct evidence for PvPOX1’s role in resistance by transforming the resistant allele into a susceptible common bean genotype, BRB130, via hairy root transformation using Agrobacterium rhizogenes. Analysis of PvPOX1 gene over-expressing hairy roots showed it increased resistance to Fusarium wilt both in the roots and the rest of transgenic plants. Meanwhile, the PvPOX1 expressive level, the peroxidase activity and hydrogen peroxide (H2O2) accumulation were also enhanced in the interaction. The result showed that the PvPOX1 gene played an essential role in Fusarium wilt resistance through the occurrence of reactive oxygen species (ROS) induced hypersensitive response. Therefore, PvPOX1 expression was proven to be a valuable gene for further analysis which can strengthen host defense response against Fusarium wilt through a ROS activated resistance mechanism

    Construction and Applications of Billion-Scale Pre-trained Multimodal Business Knowledge Graph

    Business Knowledge Graphs (KGs) are important to many enterprises today, providing factual knowledge and structured data that steer many products and make them more intelligent. Despite their promising benefits, building business KG necessitates solving prohibitive issues of deficient structure and multiple modalities. In this paper, we advance the understanding of the practical challenges related to building KG in non-trivial real-world systems. We introduce the process of building an open business knowledge graph (OpenBG) derived from a well-known enterprise, Alibaba Group. Specifically, we define a core ontology to cover various abstract products and consumption demands, with fine-grained taxonomy and multimodal facts in deployed applications. OpenBG is an open business KG of unprecedented scale: 2.6 billion triples with more than 88 million entities covering over 1 million core classes/concepts and 2,681 types of relations. We release all the open resources (OpenBG benchmarks) derived from it for the community and report experimental results of KG-centric tasks. We also run up an online competition based on OpenBG benchmarks, and has attracted thousands of teams. We further pre-train OpenBG and apply it to many KG- enhanced downstream tasks in business scenarios, demonstrating the effectiveness of billion-scale multimodal knowledge for e-commerce.     Meta-analysis of the association between toll-like receptor gene polymorphisms and hepatitis C virus infection

    ObjectiveThe objective of this study is to investigate the association between toll-like receptor (TLR) 3/7 gene polymorphisms and the infection by hepatitis C virus (HCV).MethodsPubMed, Embase, Web of Science, Scopus, CNKI, Wanfang Data, and SinoMed were searched to identify studies focusing on the association between the TLR3 rs3775290 or the TLR7 rs179008 single nucleotide polymorphisms (SNPs) and the HCV infection. All the related articles were collected from the inception of each database to 15 January 2023. Our meta-analysis was conducted using the allelic model, the dominant model, and the recessive model. Outcomes were presented by odds ratio (ORs) and 95% confidence interval (95%CI). The heterogeneity across studies was assessed by the I2 test. A subgroup analysis was performed to explore the source of heterogeneity. Funnel plots were drawn to assess the risk of publication bias. Review Manager 5.4 was used for statistical analysis.ResultsTen articles were finally included, among which six studies were analyzed for rs3775290 and five studies were analyzed for rs179008. Studies relating to rs3775290 included 801 patients and 1,045 controls, whereas studies relating to rs179008 included 924 patients and 784 controls. The results of the meta-analysis showed that there is no significant association between rs3775290 gene polymorphism and HCV infection (T vs. C: OR = 1.12, 95%CI 0.97–1.30; TT+CT vs. CC: OR = 1.20, 95%CI 0.73–1.96; TT vs. CT+CC: OR = 1.13, 95%CI 0.68–1.89). The recessive model showed that rs179008-T allele homozygotes had an 89% increased risk of infection by HCV compared with rs179008-A allele carriers (TT vs. AT+AA: OR = 1.89, 95%CI 1.13–3.16). The results of the subgroup analysis demonstrated that the characteristics of the control population may serve as an important source of heterogeneity. In the African populations, individuals with homozygous rs179008-T alleles had a higher risk of infection by HCV than rs179008-A allele carriers (OR = 2.14, 95%CI 1.18–3.87). We did not find that this difference existed in the European populations (OR = 1.24, 95%CI 0.43–3.56).ConclusionThere is no significant association between rs3775290 single nucleotide polymorphism and the infection by HCV. Individuals with homozygous rs179008-T alleles have a higher risk of an infection by HCV than rs179008-A allele carriers, which is statistically significant in the African populations

    Transcriptome analysis provides insights into the mechanism of carapace stripe formation in two closely related Marsupenaeus species

    Marsupenaeus japonicus has two types of phenotypic differences that are mainly reflected in the stripe pattern of the carapace. However, the underlying mechanism regulating the stripe patterns is not clear. In the present study, we first observed the composition of pigment cells and detected the contents of different carotenoids in the carapace of M. japonicus. We clearly observed the setae structure on the carapace. There were red pigment cells in the stripe pattern and yellow pigment cells in the other parts. Both red pigment cells and yellow pigment cells showed dendritic morphology. In the carapace, the content of astaxanthin was the highest, significantly (P < 0.05) higher than that of other carotenoids. Some differentially expressed genes between two pattern types of M. japonicus, may be associated with the body color formation, such as crustacyanin (CRCN), apolipoprotein D (ApoD), tubulin alpha-1 chain, cuticle protein, and ABC transporter, which were verified by quantitative PCR experiments. The amino acid composition and secondary structure of CRCN A2, CRCN C1, and ApoD were significantly different. The results of this study will help to elucidate the molecular mechanism of the differential pattern formation of M. japonicus and provide a reference for further exploration of the formation mechanism of crustacean color

    Study of an unusually high level of N-glycolylneuraminic acid (NGNA) sialylation on a monoclonal antibody expressed in Chinese hamster ovary cells

    Sialic or neuraminic acids of recombinant therapeutic glycoproteins produced in mammalian cells, including monoclonal antibodies, have significant impact on the half-life, stability, and biological activity of these proteins (Hossler et al., 2009; Ghaderi et al., 2012). The predominant sialic acid N-acetylneuraminic acid (NANA or Neu5Ac) is added from precursor CMP-NANA to galactose residues of N-linked glycoproteins by sialytransferases. In most mammals CMP-NANA can also be modified to its hydroxylated derivative CMP-NGNA by CMP-N-acetylneuraminic acid hydroxylase (CMAH). NGNA can then be added from CMP-NGNA to galactose residues of the N-linked glycoproteins, also by sialytransferases. However, humans cannot make functional CMAH due to an inactivating exon deletion mutation in CMAH gene (Okerblom and Varki, 2017), and therefore cannot convert CMP-NANA to CMP-NGNA. Consequently, when injected into human patients, NGNA sialic acid containing mAbs or other recombinant glycoproteins may induce immune responses, which could negatively impact pharmacokinetics or efficacy. Therefore high levels of NGNA on therapeutic mAbs or other recombinant glycoproteins are an undesirable product quality attribute. The level of total sialic acids of recombinant glycoproteins produced in Chinese hamster ovary (CHO) cells is dictated largely by the selected cell lines, upstream process, and to a lesser degree, downstream process. NGNA sialylation is generally rare in CHO cells (Könitzer et al., 2015). Hence, therapeutic glycoproteins manufactured in these cells are considered safe for human use. However, during a first-in-human (FIH) upstream process development for a novel mAb, an initially selected desirable cell line (A) was found to produce the mAb with an unexpectedly high level of the NGNA sialic acid (\u3e30%). To the best of our knowledge such high level of NGNA sialylation on a mAb produced by CHO cells has not been reported. To mitigate potential risks associated with high NGNA in human patients, a new cell line (B) that produces the mAb with very low NGNA was selected as the manufacturing cell line for this project. In order to understand the molecular mechanism causing the high NGNA content in cell line A, we initiated comprehensive genetic gap analyses using next-generation sequencing technologies and determined the differences in genomic, transcriptomic, and miRnomic profiles of the two cell lines. The results indicate spontaneous upregulation of CMAH mRNA expression, at least 10 fold higher in cell line A compared to cell line B. In this talk we will summarize the results of our studies of this unusual sialylation behavior in CHO cells

    An evaluation based on the analytic hierarchy process and GGEbiplot on French fry potato genotypes in Yunnan, China

    A total of 33 potato (Solanum tuberosum L.) cultivars and breeding clones imported from the United States and two local cultivars (Yunshu 401 and Cooperation 88, CK) were planted and evaluated. To determine their suitability for processing into French fries at five locations (e1-e5) in Yunnan Province, China, we developed a comprehensive evaluation system using the analytical hierarchy process (AHP). Eleven evaluation indicators for French fry quality, yield, and agronomic characteristics with a relative importance (weight coefficients) of 0.483, 0.301 and 0.216, respectively, were used to analyze the 35 potato genotypes (designated g1-g35).The genotypes were ranked and the results revealed that (1) on the average, the 33 potato genotypes imported from the United States showed a lower performance compared to the local cultivars. Compared with the CK, they were classified as not vigorous (Mean 5.11 vs CK 7.75), matured earlier (Mean 5.79 vs CK 1.70), and had a low resistance to late blight (Mean 3735.59 vs CK 1418.55), requiring the use of fungicides to control the disease at the five trial locations. (2) The US cultivar ‘Defender’ (g3) ranked in the top six at all five test locations because it had higher yield (29.56 t h m-2), better fry quality (4.64), higher dry matter content (20.41%), better tuber length/width ratio (1.99), longer tubers (13.57cm), stronger plant vigor (7.17) and higher resistance to late blight (AUDPC: 3134.2). (3) By using GGEbiplot analysis, superior genotypes with high and stable yields were g3 and ‘Echo Russet’ (g33). ‘Yunshu 401’ (g34) and ‘Yukon Gem’ (g4) had high but not stable yields. The ideal test environments and hence experimental locations were Luquan (LQ, e2) and Lijiang (LJ, e4) which resulted in the best discrimination between genotypes among the five experimental locations in Yunnan. Overall, the developed evaluation system based on AHP and GGEbiplot analysis including 11 evaluation indicators for French fry quality, yield and agricultural traits can be a model for evaluation and promotion of new French fry cultivars, and evaluating and selecting the test location

    Impaired long-term potentiation and long-term memory deficits in xCT-deficient sut mice

    xCT is the functional subunit of the cystine/glutamate antiporter system xc−, which exchanges intracellular glutamate with extracellular cystine. xCT has been reported to play roles in the maintenance of intracellular redox and ambient extracellular glutamate, which may affect neuronal function. To assess a potential role of xCT in the mouse hippocampus, we performed fear conditioning and passive avoidance for long-term memories and examined hippocampal synaptic plasticity in wild-type mice and xCT-null mutants, sut mice. Long-term memory was impaired in sut mice. Normal basal synaptic transmission and short-term presynaptic plasticity at hippocampal Schaffer collateral–CA1 synapses were observed in sut mice. However, LTP (long-term potentiation) was significantly reduced in sut mice compared with their wild-type counterparts. Supplementation of extracellular glutamate did not reverse the reduction in LTP. Taken together, our results suggest that xCT plays a role in the modulation of hippocampal long-term plasticity
