Relationship between IL-10 gene -819C/T polymorphism and the risk of inflammatory bowel disease: A meta-analysis
Background: The -819C/T polymorphism in interleukin 10 (IL-10) gene has been reported to be associated with inflammatory bowel disease (IBD) ,but the previous results are conflicting.Materials and methods: The present study aimed at investigating the association between this polymorphism and risk of IBD using a meta-analysis.PubMed,Web of Science,EMBASE,google scholar and China National Knowledge Infrastructure (CNKI) databases were systematically searched to identify relevant publications from their inception to April 2016.Pooled odds ratio (OR) with 95% confidence interval (CI) was calculated using fixed- or random-effects models.Results: A total of 7 case-control studies containing 1890 patients and 2929 controls were enrolled into this meta-analysis, and our results showed no association between IL-10 gene -819C/T polymorphism and IBD risk(TT vs. CC:OR=0.81,95%CI 0.64- 1.04;CT vs. CC:OR=0.92,95%CI 0.81-1.05; Dominant model: OR=0.90,95%CI 0.80-1.02; Recessive model: OR=0.84,95%CI 0.66-1.06). In a subgroup analysis by nationality, the -819C/T polymorphism was not associated with IBD in both Asians and Caucasians. The proposed framework uses the e-commerce information platform to collect and analyze recycling data in real time, therefore optimizing the design of the supply chain

Pancancer analysis uncovers an immunological role and prognostic value of the m6A reader IGF2BP2 in pancreatic cancer
Introduction: Pancreatic ductal adenocarcinoma (PDAC) is one of the most malignant gastrointestinal tumors worldwide with a dismal prognosis and high relapse rate. PDAC is considered a “cold cancer” for which immunotherapy is not effective. Therefore, to improve the prognosis for PDAC patients, it is urgent to explore the mechanism driving its insensitivity to immunotherapy. Materials and methods: We conducted pancancer analyses to test IGF2BP family expression and survival in patients with different cancers via TCGA and GETx databases. Then, we determined the immunological role and prognostic value of IGF2BP2 in vitro, in vivo and in clinical specimens. Results: In the present study, we found that the m6A reader IGF2BP2 was the most clinically relevant member of the IGF2BP family for pancreatic cancer. High expression of IGF2BP2 was most associated with poor prognosis and an immunosuppressive microenvironment in PDAC. By IGF2BP2 knockdown, we found that tumor cell proliferation and invasive ability were significantly diminished. Importantly, we found that IGF2BP2 expression was closely associated with high expression of immunosuppressive molecules such as PD-L1. IGF2BP2 modulated downstream PD-L1 expression by regulating its mRNA stability via m6A methylation control, and we obtained the same verification in animal experiments and human tissue specimens. Conclusion: Our study contributes to existing knowledge regarding the IGF2BP2-regulated PD-L1 signaling pathway as a potential prognostic and immune biomarker in pancreatic cancer
Effects of Salt Stress on Grain Quality and Starch Properties of High-Quality Rice Cultivars
In recent days, there has been a noticeable surge in demand for high-quality rice. However, the influences of salinity on the quality and starch properties of high-quality rice remain unclear. Three high-quality rice cultivars (Nanjing 9108, Nanjing 5055, and Nanjing 46) were studied to investigate the responses of grain quality to salt stress. There were three treatments, including a control zero salt level (0 g·kg−1, CK), and two salt levels of 0.1 g·kg−1 (0.1% salt stress, T1) and 0.2 g·kg−1 (0.2% salt stress, T2). The study involved the assessment of the appearance, milling, cooking, and eating qualities of rice. We also conducted an analysis of pasting properties, an evaluation of starch thermal properties, and an examination of the fine structure of amylopectin. The findings suggest that as the level of salt stress increases, the yield of rice gradually declines, which is primarily due to a significant reduction in the total spikelet number and the ratio of filled grains. Compared with CK treatment, the appearance and milling quality of rice were significantly improved within the T1 treatment. In addition, the protein concentration and amylose concentration were significantly decreased, the gel consistency was significantly increased, and the cooking and eating qualities were improved. In terms of starch properties, the peak viscosity, breakdown value, infrared ratio (1022/995), and short-chain-length amylopectin ratio increased significantly, while the setback value, pasting temperature, gelatinization enthalpy, relative crystallinity, and infrared ratio (1045/1022) decreased significantly. When comparing T2 with CK, the appearance quality and cooking and eating quality had deteriorated, and the milling quality was improved. The changes in the structural and physicochemical properties of starch were opposite to those in the comparison between the T1 treatment and the CK group. Accordingly, we propose that moderate salt stress has the potential to enhance rice quality, even though there may be a slight decrease in yield. This indicates that it is feasible to cultivate high-quality rice in saline–alkali beach areas
Real-World Response and Outcomes in Patients With NSCLC With EGFR Exon 20 Insertion Mutations
Introduction: This study describes treatment patterns and outcomes in patients with NSCLC with EGFR exon 20 insertions (EGFRex20ins) in the United States. Methods: The Flatiron Health electronic health record database was used to select three cohorts among patients diagnosed with NSCLC with EGFRex20ins (January 1, 2011–February 29, 2020): (1) first-line (1L) or patients receiving 1L therapy after documented EGFRex20ins; (2) second or later-line (≥2L) or patients receiving ≥2L therapy after documented EGFRex20ins; and (3) ≥2L postplatinum trial-aligned, or ≥2L patients previously treated with platinum chemotherapy whose baseline characteristics aligned with key eligibility criteria (initiating new treatment after documented EGFRex20ins and ≥1 previous treatment excluding mobocertinib or amivantamab) of the mobocertinib trial NCT02716116. Real-world end points were confirmed overall response rate, overall survival, and progression-free survival. Results: Of 237 patients with EGFRex20ins-mutated NSCLC, 129 and 114 patients were included in the 1L and ≥2L cohorts, respectively. In 1L patients, platinum chemotherapy plus nonplatinum chemotherapy (31.0%) and EGFR tyrosine kinase inhibitors (28.7%) were the most common regimens. In ≥2L patients, immuno-oncology monotherapy (28.1%) and EGFR tyrosine kinase inhibitors (17.5%) were the most common index treatments. For any 1L, ≥2L, and ≥2L postplatinum trial-aligned patients, the confirmed overall response rate was 18.6%, 9.6%, and 14.0%, respectively; the median overall survival was 17.0, 13.6, and 11.5 months; the median progression-free survival was 5.2, 3.7, and 3.3 months, respectively. Conclusions: The outcomes for patients with NSCLC with EGFRex20ins were poor. This real-world study provides a benchmark on treatment outcomes in this patient population and highlights the unmet need for improved therapeutic options
