    Development of a Real time Evaluation System for Microteaching

    We have developed an evaluation system for microteaching. Unlike conventional evaluation methods using paper medium, our system enables evaluation comments to be inputted in real time. In addition, our system enables evaluation comments to be displayed using a video while microteaching. As a result, an evaluator is able to input a comment while watching the teaching practice and the student is able to check the comments while watching the video

    TbGT8 is a bifunctional glycosyltransferase that elaborates<em> N</em>-linked glycans on a protein phosphatase AcP115 and a GPI-anchor modifying glycan in <em>Trypanosoma brucei</em>

    AbstractThe procyclic form of Trypanosoma brucei expresses procyclin surface glycoproteins with unusual glycosylphosphatidylinositol-anchor side chain structures that contain branched N-acetyllactosamine and lacto-N-biose units. The glycosyltransferase TbGT8 is involved in the synthesis of the branched side chain through its UDP-GlcNAc: βGal β1-3N-acetylglucosaminyltransferase activity. Here, we explored the role of TbGT8 in the mammalian bloodstream form of the parasite with a tetracycline-inducible conditional null T. brucei mutant for TbGT8. Under non-permissive conditions, the mutant showed significantly reduced binding to tomato lectin, which recognizes poly-N-acetyllactosamine-containing glycans. Lectin pull-down assays revealed differences between the wild type and TbGT8 null-mutant T. brucei, notably the absence of a broad protein band with an approximate molecular weight of 110kDa in the mutant lysate. Proteomic analysis revealed that the band contained several glycoproteins, including the acidic ecto-protein phosphatase AcP115, a stage-specific glycoprotein in the bloodstream form of T. brucei. Western blotting with an anti-AcP115 antibody revealed that AcP115 was approximately 10kDa smaller in the mutant. Enzymatic de-N-glycosylation demonstrated that the underlying protein cores were the same, suggesting that the 10-kDa difference was due to differences in N-linked glycans. Immunofluorescence microscopy revealed the colocalization of hemagglutinin epitope-tagged TbGT8 and the Golgi-associated protein GRASP. These data suggest that TbGT8 is involved in the construction of complex poly-N-acetyllactosamine-containing type N-linked and GPI-linked glycans in the Golgi of the bloodstream and procyclic parasite forms, respectively

    A patient with octopus pot-shaped cardial cancer that metastasized to multiple organs

    A 71-year-old male was admitted for abdominal fullness. The condition rapidly deteriorated in a short period (3weeks), and the patient died. Autopsy revealed a protruding lesion measuring about 3 cm with erosion measuring 5mmin diameter immediately below the esophago-gastric conjugation site, suggesting primary cardial undifferentiated adenocarcinoma. In the primary focus, changes on the mucosal surface were almost normal. However, below the mucosa, infiltration of cancer cells was observed in an approximately 3 cm area along the gastric wall. Simultaneously, the site of infiltration was markedly increased in deep areas. Extra serous infiltration was observed. The morphology was special, and resembled an octopus pot, a trap used to catch octopuses in Japan, with a narrow top and a broad base.     Postazacitidine clone size predicts long-term outcome of patients with myelodysplastic syndromes and related myeloid neoplasms

    Azacitidine is a mainstay of therapy for MDS-related diseases. The purpose of our study is to elucidate the effect of gene mutations on hematological response and overall survival (OS), particularly focusing on their post-treatment clone size. We enrolled a total of 449 patients with MDS or related myeloid neoplasms. They were analyzed for gene mutations in pre- (n=449) and post- (n=289) treatment bone marrow samples using targeted-capture sequencing to assess the impact of gene mutations and their post-treatment clone size on treatment outcomes. In Cox proportional hazard modeling, multi-hit TP53 mutation (HR, 2.03; 95% CI, 1.42-2.91; P<.001), EZH2 mutation (HR, 1.71; 95% CI, 1.14-2.54; P=.009), and DDX41 mutations (HR, 0.33; 95% CI, 0.17-0.62; P<.001), together with age, high-risk karyotypes, low platelet, and high blast counts, independently predicted OS. Post-treatment clone size accounting for all drivers significantly correlated with International Working Group (IWG)-response (P<.001, trend test), except for that of DDX41-mutated clones, which did not predict IWG-response. Combined, IWG-response and post-treatment clone size further improved the prediction of the original model and even that of a recently proposed molecular prediction model, IPSS-M (c-index, 0.653 vs 0.688; P<.001, likelihood ratio test). In conclusion, evaluation of post-treatment clone size, together with pre-treatment mutational profile as well as IWG-response have a role in better prognostication of azacitidine-treated myelodysplasia patients

    Geology of Western Nepal and a Comparison with Kumaun, India

    Geologic structure of western Nepal has two tectonic systems, one being the Himalayan gneiss zone and its klippes of allochthonous nature and the other being the Midland meta-sediment zone of autochthonous or parautochthonous nature; the former is thrust over the latter along a salient tectonic zone called the Main Central Thrust zone. The Himalayan geneisses composed of high grade metamorphic rocks of the Barrovian type have a monotonous structure dipping gently northward. Meanwhile, the Chakhure-Mabu crystallines, which make up a klippe on the Midland meta-sediments, have been derived from the root of the Himalayan gneisses in the north. It is considered that the Jajarkot crystallines also form another klippe derived from the Main Central thrust zone in the north. The Midland meta-sediments show an anticlinorium structure composed of a thick sequence of platform-type sediments and are divided into five formations. The lower part of them is composed of the calcareous Ila Formation with stromatolite and the quartzite-rich Dali Formation, both of which are respectively correlative with the upper calcareous Subgroup and the middle siliceous Subgroup of the Midland meta-sediment Group of central Nepal. The Midland meta-sediments of western Nepal is considered to range in age from Riphean to Eocambrian despite of a possibility that the upper part would be of Palaeozoic in age. The Main Central Thrust zone is a profound tectonic zone characterized by intensely mylonitized rocks accompanied with characteristically blastomylonitic "augen" gneiss and garnet-mica-chlorite phyllitic schist.
