    The history and recent advances in research of polyprenol and its derivatives

    The reduction pathway leading to the formation of dolichol was clarified in 2010 with the identification of SRD5A3, which is the polyprenol reductase. The finding inspired us to reanalyze the length of the major chain of polyprenol and dolichol from several plant leaves, including mangrove plants, as well as from animal and fish livers by 2D-TLC. Polyprenol- and dolichol derived metabolites such as polyprenylacetone and epoxydolichol were found together with rubber-like prenol. This review focuses on analyses of polyprenol and its derivatives, including recently found epoxypolyprenol and polyprenylacetone. Attention has also been paid to the chromatographic behavior of rubber-like prenol on TLC

    The purpose of this study is to elucidate shared perspectives of problems between activities of the Ministry of Education for the revision of elementary school course of study (Music) of 1948 and preceding critical comments toward that of 1938 from Japan Teachers' Union and Association of Music Education. From its practices from 1938 to 1947, we found that the Ministry of Education put focuses on following points; which musical instruments to teach according to students' grades, effective methods of teaching how to play them and of extending abilities of singing/playing with musical scores, and treatments of Japanese indigenous melodies in composition class.     Increase in Plasma Concentrations of Geranylgeranoic Acid after Turmeric Tablet Intake by Healthy Volunteers

    Geranylgeranoic acid (GGA) is one of the most potent cancer-preventive acyclic retinoids. GGA has been shown to induce cell death in human hepatoma-derived HuH-7 cells. We have recently reported the natural occurrence of GGA and its related compounds in several medicinal herbs such as turmeric, basil, rosehip, cinnamon and others [Shidoji and Ogawa, J. Lipid Res., 45: 1092–1103, 2004]. In the present study, we performed oral administration of turmeric tablets to healthy volunteers in order to investigate bioavailability of natural GGA. By using liquid chromatography/mass spectrometry, authentic GGA was eluted at a retention time of around 18 min as a negative ion of m/z 303.4. With healthy volunteers, plasma GGA was detected prior to the tablet intake and its concentrations were increased at 2 h after its intake and maintained at higher level until 4 h, suggesting an efficient bioavailability of preformed GGA in the turmeric tablets through oral administration. These results indicated that GGA in the turmeric tablet was absorbed as an intact form from intestinal mucosa. The present study provides a clue to conduct a research for cancer preventive roles of GGA in a number of spices

    Polished rice as natural sources of cancer-preventing geranylgeranoic acid

    Geranylgeranoic acid, a 20-carbon polyprenoic acid (all-trans 3,7,11,15-tetramethyl-2,4,6,10,14-hexadecatetraenoic acid) and its derivatives were previously developed as synthetic “acyclic retinoids” for cancer chemoprevention. Recently, we demonstrated the natural occurrence of geranylgeranoic acid in various medicinal herbs (Shidoji and Ogawa, 2004). In this present study, we present several lines of evidence to demonstrate that geranylgeranyl diphosphate taken in foods could be metabolized to GGA through geranylgeraniol and geranylgeranyl aldehyde via the following steps: 1) The conversion from geranylgeranyl diphosphate to geranylgeraniol was demonstrated to occur by the action of bovine intestinal alkaline phosphatase, with a Km of 46.1 µM. 2) Geranylgeraniol oxidase-mediated conversion of geranylgeraniol to geranylgeranyl aldehyde was revealed in rat liver homogenates, which activity was mainly localized in the mitochondrial fraction. The mitochondrial enzyme showed a Km of 92.9 µM. 3) The conversion of geranylgeranyl aldehyde to geranylgeranoic acid by geranylgeranyl aldehyde dehydrogenase in rat liver homogenates was absolutely dependent on exogenously added NAD+ or NADP+. The Km of the mitochondrial geranylgeranyl aldehyde dehydrogenase was 27.5 µM for geranylgeranyl aldehyde. Taken together, our data suggest that cancer preventive geranylgeranoic acid could be a physiological metabolite from commonly consumed foods

    Association and Interaction Effect between VEGF Receptor-2 (VEGFR-2) Gene Polymorphisms and Dietary Pattern on Blood Lipids in Malaysian Adults

    Background: Vascular endothelial growth factor receptor-2 (VEGFR-2) and diet are related to blood lipid health. Gene-diet interaction studies can identify the risks. The objectives are to determine associations, and interaction effects between VEGFR-2 gene polymorphisms (rs1870377 and rs2071559) and dietary patterns on blood lipids in multi-ethnic Malaysian adults. Methods: Dietary intakes of 509 (153 Malay, 179 Chinese, and 177 Indian) Malaysians were obtained from food frequency questionnaire for the construction of dietary patterns using factor analysis. Anthropometric measurements: body mass index and blood pressure; and biomarkers: glycated hemoglobin, total cholesterol (TC), triglycerides (TG), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), and total cholesterol/HDL-C ratio were determined. Genotyping of rs1870377 and rs2071559 was performed by real-time PCR using Taqman probes. Results: Two major dietary patterns were extracted from all subjects: "Vegetables, fruits, and soy diet" (VFSD), and ‘Rice, egg, and fish diet' (REFD). There were significant genetic associations for rs2071559 with TG (p=0.048) in Malays, and in Chinese for rs1870377 with TC (p=0.001) and LDL-C (p=0.007), and rs2071559 on LDL-C (p=0.048). The interaction effects of VEGFR-2 gene polymorphisms and dietary patterns were significant in Malays between rs2071559 and REFD on TG (p=0.005); and in Chinese between VFSD and rs1870377 on LDL-C (p=0.028) after adjusting for potential confounders. There were no significant genetic or dietary associations on blood lipids in Indians (p>0.05). Conclusion: The significant associations and gene-diet interaction effects between VEGFR-2 gene polymorphisms and dietary patterns on blood lipids may pose hyperlipidemia and hypercholesterolemia risks in Malay and Chinese Malaysians
