    LHPP promotes the intracellular reactive oxygen species accumulation and sensitivity of gastric cancer to cisplatin via JNK and p38 MAPK pathways

    Background. Cisplatin is the first-line chemotherapy drug for the treatment of gastric cancer (GC) patients. However, GC patients who are resistant to cisplatin often do not benefit from it. Therefore, finding a key molecule that affects cisplatin sensitivity is expected to enhance the efficacy of cisplatin in GC treatment. Methods. The human GC cell lines SGC-7901 and BGC-823 were used. The protein chip array was used to screen the cisplatin-resistance genes from the complete response and non-complete response GC patients’ tissues, then, the differential gene expression analysis, GO function annotation analysis, and KEGG pathway enrichment analysis were performed. The GC tissue chip in the GEO database was analyzed to screen the target gene. Flow cytometry, Hoechst 33342 staining assay, Western Blot, MTT, tumor sphere formation, cell cycle, and apoptosis assays were performed to explore the effect of Phospholysine Phosphohistidine Inorganic Pyrophosphate Phosphatase (LHPP) on the apoptosis, stemness, and reactive oxygen species (ROS) accumulation of cisplatin-resistant GC cells treated with cisplatin. In vivo, the cisplatin-resistant GC cell lines transfected with pcDNA-LHPP or si-LHPP were injected subcutaneously into mice to construct GC subcutaneous xenograft GC models. Results. Based on protein chip array and bioinformatics analysis, it was found that LHPP is the core molecule in the cisplatin resistance regulatory network in GC, and its expression is down-regulated in GC cisplatin-resistant tissues and cells. In vitro and in vivo experimental results show that the up-regulated expression of LHPP is closely related to the increase in sensitivity of GC to cisplatin. Mechanically, we found that overexpression of LHPP may inhibit the activation of the JNK and p38 MAPK pathways, promote cisplatininduced ROS accumulation, suppress stemness, and enhance the sensitivity of GC to cisplatin. Conclusions. Up-regulation of LHPP may inhibit the activation of the JNK and p38 MAPK pathways, attenuate stemness, and enhance the accumulation of intracellular ROS, thereby promoting cisplatin-mediated GC cell apoptosis and enhancing cisplatin sensitivity

    Tracking Nanometer-Scale Fluorescent Particles in Two Dimensions With a Confocal Microscope

    Abstract-A system for tracking multiple nanometer-scale fluorescent particles in a confocal microscope and an experimental validation is described. Position estimates of an individual fluorescent particle are generated from fluorescence intensity measurements taken at a small number of discrete locations. Tracking is achieved by combining the estimation procedure with a linear quadratic Gaussian (LQG) regulator. Multiple particles are tracked by combining the models for individual particles into a single system, applying the same LQG framework, and then cycling the control through each subsystem in turn. Experimental results are presented for single and multiple particles. For validation purposes, during each experiment images from a charge-coupled device camera were captured and analyzed offline using a standard Gaussian fit method. The estimated trajectories were in good agreement with those produced by the LQG algorithm, thereby verifying the tracking scheme

    Design of remote wireless monitoring system for coal mine main ventilator

    The main ventilator room of coal mine is remote and difficultly covered by wired network, so as to cause main ventilator monitoring system falling into information island. For above problem, a design scheme of remote wireless monitoring system for coal mine main ventilator based on mobile platform was put forward. The system uses a GPRS DTU to communicate with PLC for collecting field monitoring data, and the data is uploaded to a cloud data sever through GPRS network. On the other hand, mobile client based on Android system accesses the cloud data sever by use of socket communication mode through 3G, 4G or WiFi wireless network, so that users can remotely monitor operation status of coal mine main ventilator anytime and anywhere. The test results verify feasibility of the system

    Temperature gradient zoning of steel beams without paving layers in China

    The solar temperature field and its temperature effects on the straddle monorail tourist transportation system (SMTTS) significantly affect its mechanical performance. To investigate the distribution law of the daylight temperature field of the unpaved steel beams, the daylight temperature fields of box-type and I-sectional steel beams in different regions were measured and simulated. With the simulation results, the vertical temperature gradient curves of the unpaved steel girders were determined. Afterward, with the historical meteorological data as variables, the parameters of the extreme value model of generalized extreme value (GEV) distribution were obtained, and the representative values of temperature differences in China, based on the recurrence period, were analyzed. By analyzing the temperature difference values in 34 cities in China, an empirical prediction formula for the representative temperature difference value with three parameters was proposed, and the temperature gradient partition was established. The results indicated that the vertical temperature gradients of closed-section steel box girders are about 2–3 °C greater than that of open-section I-beams. Moreover, there are apparent geographical differences in the temperature difference values of steel girders

    Cohort profile: the Liyang cohort study on chronic diseases and risk factors monitoring in China (Liyang Study)

    Purpose The Liyang cohort study on chronic diseases and risk factors monitoring in China (Liyang Study) is a prospective population-based study which aims to investigate and identify the determinants of the most prevalent chronic non-communicable diseases (NCDs) and to evaluate the impact of demographic characteristics, lifestyle, dietary habits, cognition, disability and NCDs on the health-related quality of life.Participants Between March 2019 and June 2020, 10 056 individuals aged ≥18 years were administered a baseline survey through a multistage cluster random sampling in Liyang City, southern Jiangsu Province, China.Findings to date The Liyang Study included detailed sociodemographic, anthropometric and health-related behaviour, common NCDs and blood sample information. Moreover, the study gathered a series of data on specific scales including the activities of daily living, instrumental activities of daily living, abbreviated mental test, Food Frequency Questionnaire and EuroQol 5-Dimensions 5-Levels Scale. Of the 10 056 participants, 52.92% (n=5322) were female and 92.26% (n=9278) came from rural areas. The mean age was 49.9±16.2 years. Men were more likely to have a higher level of education, annual income and a paid job than women (p<0.05). The top three overall most prevalent NCDs in the study were hypertension (18.06%, n=1815), digestive diseases (7.88%, n=791), and arthritis or rheumatism (5.28%, n=530). Women had a significantly higher prevalence of diabetes (5.46%, n=290 vs 4.42%, n=209, p=0.016) and arthritis (6.04%, n=321 vs 4.42%, n=209, p<0.001) than men, while the opposite was true for chronic lung diseases such as chronic obstructive pulmonary disease (1.37%, n=65 vs 0.92%, n=49, p=0.032) and chronic hepatic diseases (0.80%, n=38 vs 0.47%, n=25, p=0.035).Future plans The current study will give valuable insights into the association between sociodemographic factors, health-related behaviour, diet, cognition, disability and genetic factors and the most prevalent NCDs among local community residents. Starting from 2022, a follow-up survey will be conducted every 3 years to further explore the causal relationship between the above factors and NCDs
