    UXT at the crossroads of cell death, immunity and neurodegenerative diseases

    The ubiquitous expressed transcript (UXT), a member of the prefoldin-like protein family, modulates regulated cell death (RCD) such as apoptosis and autophagy-mediated cell death through nuclear factor-κB (NF-κB), tumor necrosis factor-α (TNF-α), P53, P62, and methylation, and is involved in the regulation of cell metabolism, thereby affecting tumor progression. UXT also maintains immune homeostasis and reduces proteotoxicity in neuro-degenerative diseases through selective autophagy and molecular chaperones. Herein, we review and further elucidate the mechanisms by which UXT affects the regulation of cell death, maintenance of immune homeostasis, and neurodegenerative diseases and discuss the possible UXT involvement in the regulation of ferroptosis and immunogenic cell death, and targeting it to improve cancer treatment outcomes by regulating cell death and immune surveillance

    Analysis of the Electrical and Thermal Properties for Magnetic Fe3O4-Coated SiC-Filled Epoxy Composites.

    Orderly arranged Silicon carbide (SiC)/epoxy (EP) composites were fabricated. SiC was made magnetically responsive by decorating the surface with iron oxide (Fe3O4) nanoparticles. Three treatment methods, including without magnetization, pre-magnetization and curing magnetization, were used to prepare SiC/EP composites with different filler distributions. Compared with unmodified SiC, magnetic SiC with core-shell structure was conducive to improve the breakdown strength of SiC/EP composites and the maximum enhancement rate was 20.86%. Among the three treatment methods, SiC/EP composites prepared in the curing-magnetization case had better comprehensive properties. Under the action of magnetic field, magnetic SiC were orderly oriented along the direction of an external field, thereby forming SiC chains. The magnetic alignment of SiC restricted the movement of EP macromolecules or polar groups to some extent, resulting in the decrease in the dielectric constant and dielectric loss. The SiC chains are equivalent to heat flow channels, which can improve the heat transfer efficiency, and the maximum improvement rate was 23.6%. The results prove that the orderly arrangement of SiC had a favorable effect on dielectric properties and thermal conductivity of SiC/EP composites. For future applications, the orderly arranged SiC/EP composites have potential for fabricating insulation materials in the power electronic device packaging field

    Loop-mediated isothermal amplification for detection of porcine circovirus type 2

    <p>Abstract</p> <p>Background</p> <p>Porcine circovirus type 2 (PCV2) is the primary causative agent of the emerging swine disease known as postweaning multisystemic wasting syndrome (PMWS). Nowadays, polymerase chain reaction (PCR) is still the most widespread technique in pathogen detection. Loop-mediated isothermal amplification (LAMP), a novel nucleic acid amplification method developed in 2000, will possibly replace PCR in the field of detection. To establish a LAMP method for rapid detection of PCV2, two pairs of primers were designed specially from the open reading frame 2 (ORF2) sequences of PCV2. A LAMP method for rapid detection of PCV2 was established. To compare with PCR, sensitivity and specificity of LAMP were evaluated using the optimized reaction system. The LAMP products could be determined by agarose gel electrophoresis or adding SYBR Green I dye.</p> <p>Results</p> <p>The amplification of LAMP could be obtained at 63°C for 60 min. The detection limit was nearly 1 copy of DNA plasmid, more sensitive than PCR. There was no cross-reaction with porcine circovirus type 1 (PCV1), porcine pseudorabies virus (PRV) and porcine parvovirus (PPV) under the same conditions.</p> <p>Conclusions</p> <p>LAMP is an useful rapid detection method with high sensitivity and specificity for PCV2.</p

    A global North-South division line for portraying urban development

    Rapid urbanization has tremendously changed the global landscape with profound impacts on our society. Nighttime light (NTL) data can provide valuable information about human activities and socioeconomic conditions thus has become an effective proxy to measure urban development. By using NTL-derived urban measures from 1992 to 2018, we analyzed the spatiotemporal patterns of global urban development from country to region to city scales, which presented a distinct North-South divergence characterized by the rising and declining patterns. A global North-South division line was identified to partition the globe into the Line-North and the Line-South geographically, which accorded with the socioeconomic difference from the aspects of urban population and economy. This line may keep a certain degree of stability deriving from the trends of population and economic information but also bears uncertainties in the long term

    Global health effects of future atmospheric mercury emissions

    Mercury is a potent neurotoxin that poses health risks to the global population. Anthropogenic mercury emissions to the atmosphere are projected to decrease in the future due to enhanced policy efforts such as the Minamata Convention, a legally-binding international treaty entered into force in 2017. Here, we report the development of a comprehensive climate-atmosphere-land-ocean-ecosystem and exposure-risk model framework for mercury and its application to project the health effects of future atmospheric emissions. Our results show that the accumulated health effects associated with mercury exposure during 2010–2050 are $19 (95% confidence interval: 4.7–54) trillion (2020 USD) realized to 2050 (3% discount rate) for the current policy scenario. Our results suggest a substantial increase in global human health cost if emission reduction actions are delayed. This comprehensive modeling approach provides a much-needed tool to help parties to evaluate the effectiveness of Hg emission controls as required by the Minamata Convention

    Gastric Mammalian Target of Rapamycin Signaling Contributes to Inhibition of Ghrelin Expression Induced by Roux-En-Y Gastric Bypass

    Background/Aims: Roux-en-Y Gastric Bypass, RYGB, is the most effective strategy to control body weight in morbid obesity. RYGB leads to rapid improvement of glycemic status and weight loss, which are largely attributed to the alteration of gastrointestinal hormones including ghrelin. The current study examined potential mechanisms of altered ghrelin synthesis after RYGB. Methods: Gastric mammalian target of rapamycin (mTOR) signaling, ghrelin synthesis and secretion were determined in lean or obese male mice with or without RYGB operation, as well as in obese patients pre- and post-RYGB surgery. Ghrelin expression and mTOR signaling were investigated by western blotting and immunohistochemistry. Ghrelin mRNA levels were detected by real-time PCR. Plasma ghrelin was measured by enzyme immunoassay. Results: mTOR activity in the gastric fundus was significantly lower than in the forestomachs. Both of them were decreased after 24h fasting. A significant negative correlation was found between gastric levels of phospho-S6 (phospho-S6 ribosomal protein) and proghrelin during changes of energy status. mTOR activity was activated, whereas ghrelin expression was inhibited by Roux-en-Y Gastric Bypass in both rodents and human beings. Increment of ghrelin synthesis and decline of mTOR signaling induced by rapamycin were significantly reversed by RYGB in both lean and obese mice. Administration of Ad-S6K1 (adenovirus-mediated p70 ribosomal protein subunit 6 kinase 1) from tail vein suppressed the expression of ghrelin in RYGB-operated mice relative to control animals. Conclusion: mTOR is therefore a gastric fuel sensor whose activity is linked to the regulation of ghrelin after Roux-en-Y Gastric Bypass

    Trade impact analysis on the profit and loss of agroecosystem service value in Guangdong Province

    Due to the imbalance in the supply and demand of agricultural land, there is an increasing trend of land resources being remoted utilized across regions. Within the context of regional trade, changes in the agroecosystem service value (ESV) can be used to assess consumer responsibility. However, the embodied flow of ESV and its driving factors have been largely unexplored. This paper, utilizing the latest Chinese multi-regional input–output tables and the equivalence factor method, explores the spatiotemporal characteristics of embodied agricultural land and ESV flow between Guangdong and 30 other provinces, and further seeks to construct three types of embodied ESV models to reveal the driving factors. The findings reveal that during the process of domestic trade in goods and services, Guangdong Province has an inflow of 2.10 × 107 hm2 of land from other provinces, with arable land, forestland, and grassland accounting for 25, 61, and 14%, respectively. Guangdong’s utilization of ESV is mainly dependent on external inflows, with minimal local consumption and outflows. The embodied ESV between Guangdong and other provinces is 1626.10 billion yuan, with an outflow of 325.32 billion yuan. The “Y”-shaped region consisting of the northwest, northeast, southwest, and Hainan bears significant potential ESV losses for Guangdong Province. Population growth and the intensity of ESV loss will promote the flow of cropland and forestland ESV, while economic development has a certain inhibitory effect on ESV transfer. This paper provides a new analytical perspective on issues such as the spatial distribution mismatch of land resources and ecologically unequal exchange. These insights are pivotal for promoting sustainable utilization of land resources and regional equity
