The Site Specific Demethylation in the 5′-Regulatory Area of NMDA Receptor 2B Subunit Gene Associated with CIE-Induced Up-Regulation of Transcription
The NMDA receptor represents a particularly important site of ethanol action in the CNS. We recently reported that NMDA receptor 2B (NR2B) gene expression was persistently up-regulated following chronic intermittent ethanol (CIE) treatment. Increasing evidence that epigenetic mechanisms are involved in dynamic and long-lasting regulation of gene expression in multiple neuroadaptive processes prompted us to investigate the role of DNA methylation in mediating CIE-induced up-regulation of NR2B gene transcription. To dissect the changes of DNA methylation in the NR2B gene, we have screened a large number of CpG sites within its 5′-regulatory area following CIE treatment. DNA methylation assays were performed to determine the direct impact of DNA methylation on the interaction between DNA and transcription factor and promoter activity. of methylated DNA decreased transcription factor binding activity and promoter activity. An additional ChIP assay indicated that the CIE-induced DNA demethylation is accompanied by increased occupation by transcription factors.These results suggest an important role of DNA demethylation in mediating CIE-induced NR2B gene up-regulation, thus implicating a novel molecular site of alcohol action
Dorsal Visual Pathway Changes in Patients with Comitant Extropia
BACKGROUND: Strabismus is a disorder in which the eyes are misaligned. Persistent strabismus can lead to stereopsis impairment. The effect of strabismus on human brain is not unclear. The present study is to investigate whether the brain white structures of comitant exotropia patients are impaired using combined T1-weighted imaging and diffusion tensor imaging (DTI). PRINCIPAL FINDINGS: Thirteen patients with comitant strabismus and twelve controls underwent magnetic resonance imaging (MRI) with acquisition of T1-weighted and diffusion tensor images. T1-weighted images were used to analyze the change in volume of white matter using optimized voxel-based morphology (VBM) and diffusion tensor images were used to detect the change in white matter fibers using voxel-based analysis of DTI in comitant extropia patients. VBM analysis showed that in adult strabismus, white matter volumes were smaller in the right middle occipital gyrus, right occipital lobe/cuneus, right supramarginal gyrus, right cingulate gyrus, right frontal lobe/sub-gyral, right inferior temporal gyrus, left parahippocampa gyrus, left cingulate gyrus, left occipital lobe/cuneus, left middle frontal gyrus, left inferior parietal lobule, and left postcentral gyrus, while no brain region with greater white matter volume was found. Voxel-based analysis of DTI showed lower fractional anisotropy (FA) values in the right middle occipital gyrus and right supramarginal gyrus in strabismus patients, while brain region with increased FA value was found in the right inferior frontal gyrus. CONCLUSION: By combining VBM and voxel-based analysis of DTI results, the study suggests that the dorsal visual pathway was abnormal or impaired in patients with comitant exotropia
Overview to the Hard X-ray Modulation Telescope (Insight-HXMT) Satellite
As China's first X-ray astronomical satellite, the Hard X-ray Modulation
Telescope (HXMT), which was dubbed as Insight-HXMT after the launch on June 15,
2017, is a wide-band (1-250 keV) slat-collimator-based X-ray astronomy
satellite with the capability of all-sky monitoring in 0.2-3 MeV. It was
designed to perform pointing, scanning and gamma-ray burst (GRB) observations
and, based on the Direct Demodulation Method (DDM), the image of the scanned
sky region can be reconstructed. Here we give an overview of the mission and
its progresses, including payload, core sciences, ground calibration/facility,
ground segment, data archive, software, in-orbit performance, calibration,

Insight-HXMT observations of Swift J0243.6+6124 during its 2017-2018 outburst
The recently discovered neutron star transient Swift J0243.6+6124 has been
monitored by {\it the Hard X-ray Modulation Telescope} ({\it Insight-\rm HXMT).
Based on the obtained data, we investigate the broadband spectrum of the source
throughout the outburst. We estimate the broadband flux of the source and
search for possible cyclotron line in the broadband spectrum. No evidence of
line-like features is, however, found up to . In the absence of
any cyclotron line in its energy spectrum, we estimate the magnetic field of
the source based on the observed spin evolution of the neutron star by applying
two accretion torque models. In both cases, we get consistent results with
, and peak luminosity of which makes the source the first Galactic ultraluminous
X-ray source hosting a neutron star.Comment: publishe
Deep Sequencing and Analysis of Transcriptomes of Pinus koraiensis Sieb. & Zucc.
The objective of this research was to study the differences in endogenous hormone levels and the genes related to reproductive development in Chinese pinenut (Pinus koraiensis) trees of different ages. The apical buds of P. koraiensis were collected from 2-, 5-, 10-, 15-, and 30-year-old plants and also from grafted plants. There were three replicates from each group used for transcriptome sequencing. After assembly and annotation, we identified the differentially expressed genes (DEGs) and performed enrichment analysis, pathway analysis, and expression analysis of the DEGs in each sample. The results showed that unigenes related to reproductive development, such as c64070.graph_c0 and c68641.graph_c0, were expressed at relatively low levels at young ages, and that the relative expression gradually increased with increasing plant age. In addition the highest expression levels were reached around 10 and 15 years of age, after which they gradually decreased. Moreover, some unigenes, such as c61855.graph_c0, were annotated as abscisic acid hydroxylase genes, and the expression of c61855.graph_c0 gradually declined with increasing age in P. koraiensis
Comparative Transport Studies of '1212' Superconductors
HgBa2CaCu2O6+δ (Hg -1212) thin films were fabricated by exchanging the T1 cations in TlBa2CaCu2O7-δ (Tl-1212) thin films with Hg cations, causing a 30-K increase in Tc. To determine how this exchange effects such a Tc increase, the irreversibility lines, temperature dependence of critical current density, and temperature dependence of Hall angle ofHg-1212 and TI-1212 thin films were measured and then compared.
