    Jet Algorithms and Top Quark Mass Measurement

    Mass measurements of objects that decay into hadronic jets, such as the top quark, are shown to be improved by using a variant of the ktk_t jet algorithm in place of standard cone algorithms. The possibility and importance of better estimating the neutrino component in tagged bb jets is demonstrated.     A versatile reducible polycation-based system for efficient delivery of a broad range of nucleic acids

    Synthetic vectors based on reducible polycations consisting of histidine and polylysine residues (HIS RPCs) were evaluated for their ability to deliver nucleic acids. Initial experiments showed that RPC-based vectors with at least 70% histidine content mediated efficient levels of gene transfer without requirement for the endosomolytic agent chloroquine. Significant gene transfer was observed in a range of cell types achieving up to a 5-fold increase in the percentage of transfected cells compared to 25 kDa PEI, a gold standard synthetic vector. In contrast to 25 kDa PEI, HIS RPCs also mediated efficient transfer of other nucleic acids, including mRNA encoding green fluorescent protein in PC-3 cells and siRNA directed against the neurotrophin receptor p75(NTR) in post-mitotic cultures of rat dorsal root ganglion cell neurons. Experiments to elevate intracellular glutathione and linear profiling of cell images captured by multiphoton fluorescent microscopy highlighted that parameters such as the molecular weight and rate of cleavage of HIS RPCs were important factors in determining transfection activity. Altogether, these results demonstrate that HIS RPCs represent a novel and versatile type of vector that can be used for efficient cytoplasmic delivery of a broad range of nucleic acids. This should enable different or a combination of therapeutic strategies to be evaluated using a single type of polycation-based vector

    Persistence survey of Toxic Shock Syndrome toxin-1 producing Staphylococcus aureus and serum antibodies to this superantigen in five groups of menstruating women

    Background: Menstrual Toxic Shock Syndrome (mTSS) is thought to be associated with the vaginal colonization with specific strains of Staphylococcus aureus TSST-1 in women who lack sufficient antibody titers to this toxin. There are no published studies that examine the seroconversion in women with various colonization patterns of this organism. Thus, the aim of this study was to evaluate the persistence of Staphylococcus aureus colonization at three body sites (vagina, nares, and anus) and serum antibody to toxic shock syndrome toxin-producing Staphylococcus aureus among a small group of healthy, menstruating women evaluated previously in a larger study. Methods: One year after the completion of that study, 311 subjects were recalled into 5 groups. Four samples were obtained from each participant at several visits over an additional 6-11 month period: 1) an anterior nares swab; 2) an anal swab; 3) a vagina swab; and 4) a blood sample. Gram stain, a catalase test, and a rapid S. aureus-specific latex agglutination test were performed to phenotypically identify S. aureus from sample swabs. A competitive ELISA was used to quantify TSST-1 production. Human TSST-1 IgG antibodies were determined from the blood samples using a sandwich ELISA method. Results: We found only 41% of toxigenic S. aureus and 35.5% of non-toxigenic nasal carriage could be classified as persistent. None of the toxigenic S. aureus vaginal or anal carriage could be classified as persistent. Despite the low persistence of S. aureus colonization, subjects colonized with a toxigenic strain were found to display distributions of antibody titers skewed toward higher titers than other subjects. Seven percent (5/75) of subjects became seropositive during recall, but none experienced toxic shock syndrome-like symptoms. Conclusions: Nasal carriage of S. aureus appears to be persistent and the best predicator of subsequent colonization, whereas vaginal and anal carriage appear to be more transient. From these findings, it appears that antibody titers in women found to be colonized with toxigenic S. aureus remained skewed toward higher titers whether or not the colonies were found to be persistent or transient in nature. This suggests that colonization at some point in time is sufficient to elevate antibody titer levels and those levels appear to be persistent. Results also indicate that women can become seropositive without experiencing signs or symptoms of toxic shock syndrome

    Measurement of the Higgs boson coupling properties in the H → ZZ* → 4 decay channel at √s = 13 TeV with the ATLAS detector

    The coupling properties of the Higgs boson are studied in the four-lepton (e, μ) decay channel using 36.1 fb−1 of pp collision data from the LHC at a centre-of-mass energy of 13 TeV collected by the ATLAS detector. Cross sections are measured for the main production modes in several exclusive regions of the Higgs boson production phase space and are interpreted in terms of coupling modifiers. The inclusive cross section times branching ratio for H → ZZ∗ decay and for a Higgs boson absolute rapidity below 2.5 is measured to be 1. 73 − 0.23 + 0.24 (stat.) − 0.08 + 0.10 (exp.) ± 0.04(th.) pb compared to the Standard Model prediction of 1.34±0.09 pb. In addition, the tensor structure of the Higgs boson couplings is studied using an effective Lagrangian approach for the description of interactions beyond the Standard Model. Constraints are placed on the non-Standard-Model CP-even and CP-odd couplings to Z bosons and on the CP-odd coupling to gluons

    Identification of boosted, hadronically decaying W bosons and comparisons with ATLAS data taken at √s = 8 TeV

    This paper reports a detailed study of techniques for identifying boosted, hadronically decaying W bosons using 20.3 fb −¹ of proton–proton collision data collected by the ATLAS detector at the LHC at a centre-of-mass energy √s = 8 TeV. A range of techniques for optimising the signal jet mass resolution are combined with various jet substructure variables. The results of these studies in Monte Carlo simulations show that a simple pairwise combination of groomed jet mass and one substructure variable can provide a 50 % efficiency for identifying W bosons with transverse momenta larger than 200 GeV while maintaining multijet background efficiencies of 2–4 % for jets with the same transverse momentum. These signal and background efficiencies are confirmed in data for a selection of tagging techniques

    Finishing the euchromatic sequence of the human genome

    The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead
