    Regulation of Life Span by \u3cem\u3eDAF-16\u3c/em\u3e/Forkhead Transcription Factor in \u3cem\u3eCaenorhabditis elegans\u3c/em\u3e: A Dissertation

    The insulin/IGF-1 signaling pathway plays a pivotal role in life span regulation in diverse organisms. In Caenorhabditis elegans, a PI 3-kinase signaling cascade downstream of DAF-2, an ortholog of the mammalian insulin and insulin-like growth factor-1 (IGF-1) receptor, negatively regulates DAF-16/forkhead transcription factor. DAF-16 then regulates a wide variety of genes involved in longevity, stress response, metabolism and development. DAF-16 also receives signals from other pathways regulating life span and development. However, the precise mechanism by which DAF-16 directs multiple functions is poorly understood. First, in Chapter II, we demonstrate that JNK is a novel positive regulator of DAF-16 in both life span regulation and stress resistance. Our genetic analysis suggests that the JNK pathway acts in parallel with the insulin-like signaling pathway to regulate life span and both pathways converge onto DAF-16. We also show that JNK-1 directly interacts with and phosphorylates DAF-16. Moreover, in response to heat stress, JNK-1 promotes the translocation of DAF-16 into thc nucleus. Our findings define a novel interaction between the stress response pathway (JNK) and the master regulator of life span (DAF-16), and provide a mechanism by which JNK regulates longevity and stress resistance. Next, in Chapter III, we focus on the downstream targets of DAF-16. Here, we used a modified chromatin immunoprecipitation (ChIP) method to identify direct target promoters of DAF-16. We cloned 103 target sequences containing consensus DAF-16 binding sites and randomly selected 33 targets for further analysis. The expression of majority of these genes is regulated in a DAF-16-dependent manner. Moreover, inactivation of more than 50% of these genes significantly altered DAF-16-dependent functions such as longevity, fat storage and dauer diapause. Our results show that the ChIP-based cloning strategy leads to greater enrichment of DAF-16 target genes, compared to previous studies using DNA micro array or bioinformatics. We also demonstrate that DAF-16 is recruited to multiple promoters to coordinate regulation of its downstream target genes. In summary, we identified the JNK signaling pathway as a novel input into DAF-16 to adapt animals to the environmental stresses. We also revealed a large number of novel outputs of DAF-16. Taken together, these studies provide insight into the complex regulation by DAF-16 to control diverse biological functions and eventually broaden our understanding of aging

    Upper torso and pelvis linear velocity during the downswing of elite golfers

    BACKGROUND: During a golf swing, analysis of the movement in upper torso and pelvis is a key step to determine a motion control strategy for accurate and consistent shots. However, a majority of previous studies that have evaluated this movement limited their analysis only to the rotational movement of segments, and translational motions were not examined. Therefore, in this study, correlations between translational motions in the 3 axes, which occur between the upper torso and pelvis, were also examined. METHODS: The experiments were carried out with 14 male pro-golfers (age: 29 ± 8 years, career: 8.2 ± 4.8years) who registered in the Korea Professional Golf Association (KPGA). Six infrared cameras (VICON; Oxford Metrics, Oxford, UK) and SB-Clinc software (SWINGBANK Ltd, Korea) were used to collect optical marker trajectories. The center of mass (CoM) of each segment was calculated based on kinematic principal. In addition, peak value of CoM velocity and the time that each peak occurred in each segment during downswing was calculated. Also, using cross-correlation analysis, the degree of coupling and time lags of peak values occurred between and within segments (pelvis and upper torso) were investigated. RESULTS: As a result, a high coupling strength between upper torso and pelvis with an average correlation coefficient = 0.86 was observed, and the coupling between segments was higher than that within segments (correlation coefficient = 0.81 and 0.77, respectively). CONCLUSIONS: Such a high coupling at the upper torso and pelvis can be used to reduce the degree of motion control in the central nervous system and maintain consistent patterns in the movement. The result of this study provides important information for the development of optimal golf swing movement control strategies in the future

    Development of a heating device using CPV and heat pipe

    Paper presented to the 10th International Conference on Heat Transfer, Fluid Mechanics and Thermodynamics, Florida, 14-16 July 2014.The purpose of this research was to increase the generating efficiency of CPV within a restricted area using solar tracking and heat pipe. At the same time, we demonstrated that the proposed system had the ability to extract thermal energy from a CPV using thermal absorbers containing heat pipe, which could then be used for a heating system or hot-water supply. Tracking the sun, calculating the sun’s position, reinstating the heating device towards the east again for the next day’s tracking, and system shutdown were programmed using Simulink. A comparison of the experimental results with KASSI(Korea Astronomy & Space Science Institute) mathematical data for the sun’s position confirmed that the algorithm used for the solar-tracking device was correct. As this system can collect heat from the CPV, the efficiency was much higher than that obtained using air cooling. We performed parametric analysis of the concentration ratio with respect to the operating temperature of the CPV and outlet temperature. The simulated and experimental results for the thermal absorber containing heat pipe were in good agreement.dc201

    Case study on high-resolution monitoring network of groundwater heat pump system

    With the increasing installation of shallow geothermal energy, the importance of thermal impact prediction also increases in the system design stage. In nature, it is general that heterogeneity exists and it can affect the groundwater flow as well as the transport along to the flow. When predicting heat transport under the groundwater heat pump (GWHP) operation, however, impacts of heterogeneity have rarely been considered. In this study, to detect the hydraulic and thermal feedback to the two months of GWHP operation, a dense monitoring network was constructed with 12 monitoring wells at Eumseong-gun, Republic of Korea. The temperature was monitored in high resolution via fiber-optic distributed temperature sensing. During the GWHP operation, a very dynamic flow condition was generated with the hydraulic gradient between 0.005 and 0.07. The maximum temperature change at the nearest monitoring well was 2 ?. Observed hydraulic and thermal responses showed spatially heterogeneous results. While the heterogeneous responses of hydraulic change were stronger near the geothermal wells, those of temperature change were higher near the center of the thermal plume

    Good Glycemic Control Is Associated with Better Survival in Diabetic Patients on Peritoneal Dialysis: A Prospective Observational Study

    BACKGROUND: The effect of glycemic control after starting peritoneal dialysis (PD) on the survival of diabetic PD patients has largely been unexplored, especially in Asian population. METHODS: We conducted a prospective observational study, in which 140 incident PD patients with diabetes were recruited. Patients were divided into tertiles according to the means of quarterly HbA1C levels measured during the first year after starting PD. We examined the association between HbA1C and all-cause mortality using Cox proportional hazards models. RESULTS: The mean age was 58.7 years, 59.3% were male, and the mean follow-up duration was 3.5 years (range 0.4-9.5 years). The mean HbA1C levels were 6.3%, 7.1%, and 8.5% in the 1(st), 2(nd), and 3(rd) tertiles, respectively. Compared to the 1(st) tertile, the all-cause mortality rates were higher in the 2(nd) [hazard ratio (HR), 4.16; 95% confidence interval (CI), 0.91-18.94; p = 0.065] and significantly higher in the 3(rd) (HR, 13.16; 95% CI, 2.67-64.92; p = 0.002) tertiles (p for trend = 0.005), after adjusting for confounding factors. Cardiovascular mortality, however, did not differ significantly among the tertiles (p for trend = 0.682). In contrast, non-cardiovascular deaths, most of which were caused by infection, were more frequent in the 2(nd) (HR, 7.67; 95% CI, 0.68-86.37; p = 0.099) and the 3(rd) (HR, 51.24; 95% CI, 3.85-681.35; p = 0.003) tertiles than the 1(st) tertile (p for trend = 0.007). CONCLUSIONS: Poor glycemic control is associated with high mortality rates in diabetic PD patients, suggesting that better glycemic control may improve the outcomes of these patients
