    Regulation of Ethanol-Related Behavior and Ethanol Metabolism by the Corazonin Neurons and Corazonin Receptor in Drosophila melanogaster

    Impaired ethanol metabolism can lead to various alcohol-related health problems. Key enzymes in ethanol metabolism are alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH); however, neuroendocrine pathways that regulate the activities of these enzymes are largely unexplored. Here we identified a neuroendocrine system involving Corazonin (Crz) neuropeptide and its receptor (CrzR) as important physiological regulators of ethanol metabolism in Drosophila. Crz-cell deficient (Crz-CD) flies displayed significantly delayed recovery from ethanol-induced sedation that we refer to as hangover-like phenotype. Newly generated mutant lacking Crz Receptor (CrzR01) and CrzR-knockdown flies showed even more severe hangover-like phenotype, which is causally associated with fast accumulation of acetaldehyde in the CrzR01 mutant following ethanol exposure. Higher levels of acetaldehyde are likely due to 30% reduced ALDH activity in the mutants. Moreover, increased ADH activity was found in the CrzR01 mutant, but not in the Crz-CD flies. Quantitative RT-PCR revealed transcriptional upregulation of Adh gene in the CrzR01. Transgenic inhibition of cyclic AMP-dependent protein kinase (PKA) also results in significantly increased ADH activity and AdhmRNA levels, indicating PKA-dependent transcriptional regulation of Adh by CrzR. Furthermore, inhibition of PKA or cAMP response element binding protein (CREB) in CrzR cells leads to comparable hangover-like phenotype to the CrzR01 mutant. These findings suggest that CrzR-associated signaling pathway is critical for ethanol detoxification via Crz-dependent regulation of ALDH activity and Crz-independent transcriptional regulation of ADH.     Sleep state classification using power spectral density and residual neural network with multichannel EEG signals.

    This paper proposes a classification framework for automatic sleep stage detection in both male and female human subjects by analyzing the electroencephalogram (EEG) data of polysomnography (PSG) recorded for three regions of the human brain, i.e., the pre-frontal, central, and occipital lobes. Without considering any artifact removal approach, the residual neural network (ResNet) architecture is used to automatically learn the distinctive features of different sleep stages from the power spectral density (PSD) of the raw EEG data. The residual block of the ResNet learns the intrinsic features of different sleep stages from the EEG data while avoiding the vanishing gradient problem. The proposed approach is validated using the sleep dataset of the Dreams database, which comprises of EEG signals for 20 healthy human subjects, 16 female and 4 male. Our experimental results demonstrate the effectiveness of the ResNet based approach in identifying different sleep stages in both female and male subjects compared to state-of-the-art methods with classification accuracies of 87.8% and 83.7%, respectively

    Structural Design Of Ultra Large Ships Based On Direct Calculation Approach

    The trend in modern sea transportation is building of ever larger ships, which require application of different direct calculation methodologies and numerical tools to achieve their reliable structural design. This is particularly emphasized in case of ultra large container ships (ULCS), but also other ship types like bulk carriers or large LNG ships belong to this category. In this context some classification societies have developed guidelines for performing direct calculations and for that purpose there are several hydro-structure tools available around the world, mainly relying on the same theoretical assumptions, but having incorporated different numerical procedures. Such tools are mostly based on the application of the 3D potential flow theoretical models coupled with the 3D FEM structural models. This paper illustrates application of general hydro-structure tool HOMER (BV) in the assessment of ship structural response in waves. An outline of the numerical procedure based on the modal approach is given together with basic software description. Application case is 19000 TEU ULCS built in South Korean shipyard Hyundai Heavy Industries. Extensive hydroelastic analyses of the ship are performed, and here some representative results for fatigue response with linear springing influence are listed

    Genetic Parameter Estimation in Seedstock Swine Population for Growth Performances

    The objective of this study was to estimate genetic parameters that are to be used for across-herd genetic evaluations of seed stock pigs at GGP level. Performance data with pedigree information collected from swine breeder farms in Korea were provided by Korea Animal Improvement Association (AIAK). Performance data were composed of final body weights at test days and ultrasound measures of back fat thickness (BF), rib eye area (EMA) and retail cut percentage (RCP). Breeds of swine tested were Landrace, Yorkshire and Duroc. Days to 90 kg body weight (DAYS90) were estimated with linear function of age and ADG calculated from body weights at test days. Ultrasound measures were taken with A-mode ultrasound scanners by trained technicians. Number of performance records after censoring outliers and keeping records pigs only born from year 2000 were of 78,068 Duroc pigs, 101,821 Landrace pigs and 281,421 Yorkshire pigs. Models included contemporary groups defined by the same herd and the same seasons of births of the same year, which was regarded as fixed along with the effect of sex for all traits and body weight at test day as a linear covariate for ultrasound measures. REML estimation was processed with REMLF90 program. Heritability estimates were 0.40, 0.32, 0.21 0.39 for DAYS90, ADG, BF, EMA, RCP, respectively for Duroc population. Respective heritability estimates for Landrace population were 0.43, 0.41, 0.22, and 0.43 and for Yorkshire population were 0.36, 0.38, 0.22, and 0.42. Genetic correlation coefficients of DAYS90 with BF, EMA, or RCP were estimated to be 0.00 to 0.09, −0.15 to −0.25, 0.22 to 0.28, respectively for three breeds populations. Genetic correlation coefficients estimated between BF and EMA was −0.33 to −0.39. Genetic correlation coefficient estimated between BF and RCP was high and negative (−0.78 to −0.85) but the environmental correlation coefficients between these two traits was medium and negative (near −0.35), which describes a highly correlated genetic response to selection on one or the other of these traits. Genetic Trends of all three breeds tend to be towards bigger EMA or greater RCP and shorter DAYS90 especially from generations born after year 2000
