    Durch den Prozess der Urbanisierung verändert die Menschheit die Erdoberfläche in großem Ausmaß und auf unwiederbringliche Weise. Die optische Fernerkundung ist eine Art der Erdbeobachtung, die das Verständnis dieses dynamischen Prozesses und seiner Auswirkungen erweitern kann. Die vorliegende Arbeit untersucht, inwiefern hyperspektrale Daten Informationen über Versiegelung liefern können, die der integrierten Analyse urbaner Mensch-Umwelt-Beziehungen dienen. Hierzu wird die Verarbeitungskette von Vorverarbeitung der Rohdaten bis zur Erstellung referenzierter Karten zu Landbedeckung und Versiegelung am Beispiel von Hyperspectral Mapper Daten von Berlin ganzheitlich untersucht. Die traditionelle Verarbeitungskette wird mehrmals erweitert bzw. abgewandelt. So wird die radiometrische Vorverarbeitung um die Normalisierung von Helligkeitsgradienten erweitert, welche durch die direktionellen Reflexionseigenschaften urbaner Oberflächen entstehen.     A test of the CPL parameterization for rapid dark energy equation of state transitions

    We test the robustness and flexibility of the Chevallier-Polarski-Linder (CPL) parameterization of the Dark Energy equation of state w(z)=w0+waz1+zw(z)=w_0+w_a \frac{z}{1+z} in recovering a four-parameter step-like fiducial model. We constrain the parameter space region of the underlying fiducial model where the CPL parameterization offers a reliable reconstruction. It turns out that non negligible biases leak into the results for recent (z<2.5z<2.5) rapid transitions, but that CPL yields a good reconstruction in all other cases. The presented analysis is performed with supernova Ia data as forecasted for a space mission like SNAP/JDEM, combined with future expectations for the CMB shift parameter RR and the BAO parameter AA.Comment: 8 pages, 6 ps figure

    Measuring quality of grammars for procedural level generation

    Grammar-based procedural level generation raises the productivity of level designers for games such as dungeon crawl and platform games. However, the improved productivity comes at cost of level quality assurance. Authoring, improving and maintaining grammars is difficult because it is hard to predict how each grammar rule impacts the overall level quality, and tool support is lacking. We propose a novel metric called Metric of Added Detail (MAD) that indicates if a rule adds or removes detail with respect to its phase in the transformation pipeline, and Specification Analysis Reporting (SAnR) for expressing level properties and analyzing how qualities evolve in level generation histories. We demonstrate MAD and SAnR using a prototype of a level generator called Ludoscope Lite. Our preliminary results show that problematic rules tend to break SAnR properties and that MAD intuitively raises flags. MAD and SAnR augment existing approaches, and can ultimately help designers make better levels and level generators

    Clostridium perfringens epsilon toxin induces blood brain barrier permeability via caveolae-dependent transcytosis and requires expression of MAL.

    Clostridium perfringens epsilon toxin (ETX) is responsible for causing the economically devastating disease, enterotoxaemia, in livestock. It is well accepted that ETX causes blood brain barrier (BBB) permeability, however the mechanisms involved in this process are not well understood. Using in vivo and in vitro methods, we determined that ETX causes BBB permeability in mice by increasing caveolae-dependent transcytosis in brain endothelial cells. When mice are intravenously injected with ETX, robust ETX binding is observed in the microvasculature of the central nervous system (CNS) with limited to no binding observed in the vasculature of peripheral organs, indicating that ETX specifically targets CNS endothelial cells. ETX binding to CNS microvasculature is dependent on MAL expression, as ETX binding to CNS microvasculature of MAL-deficient mice was not detected. ETX treatment also induces extravasation of molecular tracers including 376Da fluorescein salt, 60kDA serum albumin, 70kDa dextran, and 155kDA IgG. Importantly, ETX-induced BBB permeability requires expression of both MAL and caveolin-1, as mice deficient in MAL or caveolin-1 did not exhibit ETX-induced BBB permeability. Examination of primary murine brain endothelial cells revealed an increase in caveolae in ETX-treated cells, resulting in dynamin and lipid raft-dependent vacuolation without cell death. ETX-treatment also results in a rapid loss of EEA1 positive early endosomes and accumulation of large, RAB7-positive late endosomes and multivesicular bodies. Based on these results, we hypothesize that ETX binds to MAL on the apical surface of brain endothelial cells, causing recruitment of caveolin-1, triggering caveolae formation and internalization. Internalized caveolae fuse with early endosomes which traffic to late endosomes and multivesicular bodies. We believe that these multivesicular bodies fuse basally, releasing their contents into the brain parenchyma

    Direct laser-written optomechanical membranes in fiber Fabry-Perot cavities

    Integrated micro and nanophotonic optomechanical experiments enable the manipulation of mechanical resonators on the single phonon level. Interfacing these structures requires elaborate techniques limited in tunability, flexibility, and scaling towards multi-mode systems. Here, we demonstrate a cavity optomechanical experiment using 3D-laser-written polymer membranes inside fiber Fabry-Perot cavities. Vacuum coupling strengths of ~ 30 kHz to the fundamental megahertz mechanical mode are reached. We observe optomechanical spring tuning of the mechanical resonator by tens of kHz exceeding its linewidth at cryogenic temperatures. The extreme flexibility of the laser writing process allows for a direct integration of the membrane into the microscopic cavity. The direct fiber coupling, its scaling capabilities to coupled resonator systems, and the potential implementation of dissipation dilution structures and integration of electrodes make it a promising platform for fiber-tip integrated accelerometers, optomechanically tunable multi-mode mechanical systems, or directly fiber-coupled systems for microwave to optics conversion.Comment: 10 pages, 5 figure

    Relaxing restrictions at the pace of vaccination increases freedom and guards against further COVID-19 waves

    Mass vaccination offers a promising exit strategy for the COVID-19 pandemic. However, as vaccination progresses, demands to lift restrictions increase, despite most of the population remaining susceptible. Using our age-stratified SEIRD-ICU compartmental model and curated epidemiological and vaccination data, we quantified the rate (relative to vaccination progress) at which countries can lift non-pharmaceutical interventions without overwhelming their healthcare systems. We analyzed scenarios ranging from immediately lifting restrictions (accepting high mortality and morbidity) to reducing case numbers to a level where test-trace-and-isolate (TTI) programs efficiently compensate for local spreading events. In general, the age-dependent vaccination roll-out implies a transient decrease of more than ten years in the average age of ICU patients and deceased. The pace of vaccination determines the speed of lifting restrictions; Taking the European Union (EU) as an example case, all considered scenarios allow for steadily increasing contacts starting in May 2021 and relaxing most restrictions by autumn 2021. Throughout summer 2021, only mild contact restrictions will remain necessary. However, only high vaccine uptake can prevent further severe waves. Across EU countries, seroprevalence impacts the long-term success of vaccination campaigns more strongly than age demographics. In addition, we highlight the need for preventive measures to reduce contagion in school settings throughout the year 2021, where children might be drivers of contagion because of them remaining susceptible..

    Myelination of the right parahippocampal cingulum is associated with physical activity in young healthy adults

    Recent evidence suggests that individual differences in physical activity (PA) may be associated with individual differences in white matter microstructure and with grey matter volume of the hippocampus. Therefore, this study investigated the association between PA and white matter microstructure of pathways connecting to the hippocampus. A total of 33 young, healthy adults underwent magnetic resonance imaging (MRI). High angular resolution diffusion-weighted imaging and multi-component relaxometry MRI scans (multi-component driven equilibrium pulse observation of T1 and T2) were acquired for each participant. Activity levels (AL) of participants were calculated from 72-h actigraphy recordings. Tractography using the damped Richardson Lucy algorithm was used to reconstruct the fornix and bilateral parahippocampal cinguli (PHC). The mean fractional anisotropy (FA) and the myelin water fraction (MWF), a putative marker of myelination, were determined for each pathway. A positive correlation between both AL and FA and between AL and MWF were hypothesized for the three pathways. There was a selective positive correlation between AL and MWF in the right PHC (r = 0.482, p = 0.007). Thus, our results provide initial in vivo evidence for an association between myelination of the right PHC and PA in young healthy adults. Our results suggest that MWF may not only be more specific, but also more sensitive than FA to detect white matter microstructural alterations. If PA was to induce structural plasticity of the right PHC this may contribute to reverse structural alterations of the right PHC in neuropsychiatric disorder with hippocampal pathologies

    Revisiting the Past: Replicability of a Historic Long-Term Vegetation Dynamics Assessment in the Era of Big Data Analytics

    Open and analysis-ready data, as well as methodological and technical advancements have resulted in an unprecedented capability for observing the Earth’s land surfaces. Over 10 years ago, Landsat time series analyses were inevitably limited to a few expensive images from carefully selected acquisition dates. Yet, such a static selection may have introduced uncertainties when spatial or inter-annual variability in seasonal vegetation growth were large. As seminal pre-open-data-era papers are still heavily cited, variations of their workflows are still widely used, too. Thus, here we quantitatively assessed the level of agreement between an approach using carefully selected images and a state-of-the-art analysis that uses all available images. We reproduced a representative case study from the year 2003 that for the first time used annual Landsat time series to assess long-term vegetation dynamics in a semi-arid Mediterranean ecosystem in Crete, Greece. We replicated this assessment using all available data paired with a time series method based on land surface phenology metrics. Results differed fundamentally because the volatile timing of statically selected images relative to the phenological cycle introduced systematic uncertainty. We further applied lessons learned to arrive at a more nuanced and information-enriched vegetation dynamics description by decomposing vegetation cover into woody and herbaceous components, followed by a syndrome-based classification of change and trend parameters. This allowed for a more reliable interpretation of vegetation changes and even permitted us to disentangle certain land-use change processes with opposite trajectories in the vegetation components that were not observable when solely analyzing total vegetation cover. The long-term budget of net cover change revealed that vegetation cover of both components has increased at large and that this process was mainly driven by gradual processes. We conclude that study designs based on static image selection strategies should be critically evaluated in the light of current data availability, analytical capabilities, and with regards to the ecosystem under investigation. We recommend using all available data and taking advantage of phenology-based approaches that remove the selection bias and hence reduce uncertainties in results.Peer Reviewe

    High-resolution mapping of 33 years of material stock and population growth in Germany using Earth Observation data

    Global societal material stock in buildings and infrastructure have accumulated rapidly within the last decades, along with population growth. Recently, an approach for nation-wide mapping of material stock at 10 m spatial resolution, using freely available and globally consistent Earth Observation (EO) imagery, has been introduced as an alternative to cost-intensive cadastral data or broad-scale but thematically limited nighttime light-based mapping. This study assessed the potential of EO data archives to create spatially explicit time series data of material stock dynamics and their relation to population in Germany, at a spatial resolution of 30 m. We used Landsat imagery with a change-aftereffect-trend analysis to derive yearly masks of land surface change from 1985 onward. Those served as an input to an annual reverse calculation of six material stock types and building volume-based annual gridded population, based on maps for 2018. Material stocks and population in Germany grew by 13% and 4%, respectively, showing highly variable spatial patterns. We found a minimum building stock of ca. 180 t/cap across all municipalities and growth processes characterized by sprawl. A rapid growth of stocks per capita occurred in East Germany after the reunification in 1990, with increased building activity but population decline. Possible over- or underestimations of stock growth cannot be ruled out due to methodological assumptions, requiring further research.Peer Reviewe
