    The Past Matters: Previous Management Strategies Modulate Current Growth and Drought Responses of Norway Spruce (Picea abies H. Karst.)

    Due to high productivity and past management approaches, the forests of Central Europe are heavily dominated by conifers, even on sites where they do not occur naturally at all. One prominent example is Norway spruce (Picea abies H. Karst.), a species considered particularly vulnerable to severe droughts, especially outside of its ecological niche where it has been widely planted over the past centuries. In the face of global change, it is a major task for foresters to increase these forests’ ability to cope with the impacts of increasing climatic extremes. Therefore, gaining more knowledge about how different management strategies affect the drought responses of trees is crucial. However, we still know little about the influence of the individual treatment history of a tree on its growth. We used a dendroecological approach to address this issue and to assess how initial spacing, structural diversity, tree size, and density regulation approaches modulate annual growth, especially in drought years. We hypothesized that stand establishment and past silvicultural treatment codetermine tree growth and drought resilience. Our study took place at the combined spacing-thinning trial Fürstenfeldbruck 612 (FFB 612) in Southern Germany, since it delivered precise long-term data covering a broad range of treatments. Based on linear mixed effect models, we showed that the individual treatment history of a tree affects its annual growth and drought responses considerably. In more detail, we found that (i) high structural diversity in the vicinity of each tree favored growth and improved a tree’s performance under drought; (ii) larger trees were more climate-sensitive; (iii) previous high variations in thinning intensity, and consequently strong fluctuations in growth, affected current growth negatively and reduced recovery from droughts. Furthermore, we sought to understand the underlying mechanisms and to draw potential implications for refining silvicultural guidelines

    Small-scale variation in available water capacity of the soil influences height growth of single trees in Southern Germany

    Aim of study: Detecting possible small-scale soil effects on height growth of single trees in monospecific stands of three important tree species (Abies alba, Fagus sylvatica, and Picea abies). Area of study: 37 mature stands along an ecological gradient in Southern Germany from the cold and wet “optimal niche zone” to warmer and drier niche zones, including gravelly soils with poor water supply. Material and methods: Measurement of achieved height and age of 15 to 20 sample trees per stand. Estimation of the available water capacity of the soil (AWC) in close proximity to sample trees based on soil texture following the German soil survey guidelines. Examining height growth depending on niche zone and AWC.   Main results: On sites (stand level) with the lowest water regime, height growth increased significantly with AWC of microsites. The estimated effect on height growth over the whole range of AWC values was almost 8 m at those sites. In contrast, the effect was negative on optimal sites. For intermediate and marginal sites, the effect was positive, albeit not significant for marginal sites. Research highlights: To our knowledge this is the first study about small-scale effects of AWC on height growth of single trees in temperate European forests. Small-scale soil variability should be considered in future scientific studies and practical evaluation, involving single tree performance at stands with low water regime. This seems particularly important in genetic environmental associations studies and in the process of selecting trees for breeding purposes in such stands

    Pooled RT-qPCR testing for SARS-CoV-2 surveillance in schools-a cluster randomised trial

    Background: The extent to which children and adolescents contribute to SARS-CoV-2 transmission remains not fully understood. Novel high-capacity testing methods may provide real-time epidemiological data in educational settings helping to establish a rational approach to prevent and minimize SARS-CoV-2 transmission. We investigated whether pooling of samples for SARS-CoV-2 detection by RT-qPCR is a sensitive and feasible high-capacity diagnostic strategy for surveillance of SARS-CoV-2 infections in schools. Methods: In this study, students and school staff of 14 educational facilities in Germany were tested sequentially between November 9 and December 23, 2020, two or three times per week for at least three consecutive weeks. Participants were randomized for evaluation of two different age adjusted swab sampling methods (oropharyngeal swabs or buccal swabs compared to saliva swabs using a 'lolli method'). Swabs were collected and pooled for SARS-CoV-2 RT-qPCR. Individuals of positive pooled tests were retested by RTqPCR the same or the following day. Positive individuals were quarantined while the SARS-CoV-2 negative individuals remained in class with continued pooled RT-qPCR surveillance. The study is registered with the German Clinical Trials register (registration number: DRKS00023911). Findings: 5,537 individuals were eligible and 3970 participants were enroled and included in the analysis. In students, a total of 21,978 swabs were taken and combined in 2218 pooled RT-qPCR tests. We detected 41 positive pooled tests (1 cent 8%) leading to 36 SARS-CoV-2 cases among students which could be identified by individual re-testing. The cumulative 3-week incidence for primary schools was 564/100,000 (6/1064, additionally 1 infection detected in week 4) and 1249/100,000 (29/2322) for secondary schools. In secondary schools, there was no difference in the number of SARS-CoV-2 positive students identified from pooled oropharyngeal swabs compared to those identified from pooled saliva samples (lolli method) (14 vs. 15 cases; 1.3% vs. 1.3%; OR 1.1; 95%-CI 0.5-2.5). A single secondary school accounted for 17 of 36 cases (47%) indicating a high burden of asymptomatic prevalent SARS-CoV-2 cases in the respective school and community. Interpretation: In educational settings, SARS-CoV-2 screening by RT-qPCR-based pooled testing with easily obtainable saliva samples is a feasible method to detect incident cases and observe transmission dynamics.
